SUMMARYControl and bursectomized chicks were i.m. injected with 6 mg of cyclophosphamide (Cy) (0.15 ml in saline) at hatching (Cy1×) or at hatching and the day after hatching (Cy2×). The visual basophilic lymphoid cells of the bursal follicles were replaced by large pale reticular cells in the 2-day-old Cy-treated chicks. No regeneration was observed in the bursas of chicks from the Cy2× group, while basophilic lymphoid cells appeared in the bursa of 2-week-old chicks from the Cy1× group. Germinal centers (GC) in the spleen and cecal tonsil were absent, reduced in number, or numerous in chickens from the Cy2×, Cy1×, and control groups, respectively. The Cy treatment reduced the number of plasmacytic cells. Bursectomy and a single injection of Cy were more effective in depressing the immune response than was either treatment alone. Bursectomy plus Cy1× and Cy2× produced agammaglobulinemia

SUMMARYThe production of tissue-typing reagents for dog leucocytes by allografting procedures is described. Skin and small bowel grafts were efficient in producing informative sera. A negative correlation was found between the duration of graft survival and the subsequent development of cytotoxic antibodies. It was shown that these reagents were useful in the selection of littermate donor-recipient pairs for transplantation experiments. Evidence presented from family testing and tissue transplantation supports the existence of a major (DL-A) histocompatibility locus in dogs

SUMMARYAn in vitro graft reaction system in which rat lymphocytes were sensitized in culture against mouse fibroblasts, then were tested for their capacity to effect immune lysis of mouse cells, was found to be based on a bicellular lymphoid interaction. Rat thymus cells, which by themselves are incapable of effecting the lytic reaction following sensitization on mouse fibroblasts, when combined with spleen cells, confer specific lytic activity on the mixed cell population. Experiments were made to define the tissue origin and properties of the spleen cells which are capable of interacting with the thymus cells. A mixed population of bone marrow and thymus cells, combined and cultured on mouse cells, did not manifest immune lysis of mouse target cells. Mixed populations consisting of spleen cells of thymectomized, X-irradiated mice inoculated with rat marrow and of rat thymus cells combined and cultured on mouse fibroblasts did show lytic activity. Thus, bone marrowderived cells within the spleen interact with thymus cells to effect lysis of target monolayers. The capacity of such bone marrow cells to recognize transplantation antigens was studied. Rat bone marrow-derived cells (following colonization of B6D2F1 mouse spleens), when admixed with rat thymus cells, could be sensitized and would effect lytic activity on C3H, but not on C57BL and DBA/2 strain, fibroblasts. Thus, the rat bone marrow-derived cells seemed to recognize the recipient's mouse antigen, since they manifested unresponsiveness toward fibroblasts of similar antigenic constitution. We conclude that: (1) the in vitro graft reaction system is based on interaction between thymusprocessed and nonthymus-processed lymphocytes; (2) the nonthymus-processed cells derive from the bone marrow but are not identical with the original bone marrow cells. They seem to represent a differentiation product of bone marrow cells; (3) both the thymus- and the nonthymus-processed cells are capable of recognizing the mouse antigens. The question of which is the killer cell remains open. The necessary participation of thymus cells in the killing process is discussed

SUMMARYA simple hypothermic immersion storage technique is described by which pig livers were successfully stored for up to 8 hr, as tested by orthotopic allotransplantation. This simple technique should be applicable to human liver transplantation and is recommended as more suitable for short-term storage than are currently used complex perfusion techniques. This period of storage should allow adequate time for recipient operation and tissue typing

SUMMARYElevation of serum enzymes often accompanies tissue damage in vivo. The aim of this study was to determine whether the output of the enzyme lactate dehydrogenase (LDH) during a 15-min perfusion of an isolated kidney was a measure of acute ischaemic renal damage. The results indicated that, in general, LDH output increased with increasing time of total renal ischaemia. Also, the duration of warm ischaemia and LDH output of the excised kidney were compared as indices of renal damage. This damage was assessed by the postoperative function of the remaining kidney which had undergone the same ischaemia as the one excised. The LDH output of the excised kidney showed the closer correlation with subsequent renal function

SUMMARYResults of this study indicate that the mixed lymphocyte reaction (MLR) can be used to monitor the influence of adult thymectomy as early as 4 weeks following thymus removal. Beginning 16 weeks postthymectomy, the MLR was capable of detecting the recovery of competence in animals grafted with syngeneic thymus tissue. It is suggested, therefore, that the MLR may serve as a sensitive, quantitative, and relatively accurate test for determining the immunological status of lymphoid compartments following adult thymectomy

SUMMARYThe microabsorption technique developed in this laboratory has been utilized to demonstrate HL-A1, 2, 3, 9, 5, 7, and 8 antigens on frozen leukocytes, kidney, and spleen cells and on established lymphoid cell lines. No discrepancy was noted between HL-A phenotypes of 10 healthy donors determined by means of the standard lymphocytotoxicity test (LCT) and by means of microabsorption with frozen leukocytes. Most, but not all, HL-A antigens corresponding to those on lymphocytes could be demonstrated on kidney and spleen cells by this technique. It was also shown that cultured cells of established lymphoid cell lines possess their original HL-A antigens after long-term propagation

SUMMARYThe relative efficacy of azathioprine, chlorambucil, methylprednisolone, and total body γ-radiation, alone and as combined therapy, in suppressing tetanus antitoxin production in mice was determined. An in vivo “live-die” assay for serum antitoxin titer was developed.
