The erythrocytes of two inbred strains of rats, Lewis and B.N., differ because of antigens 1 and 3, respectively, which are determined by allelic genes. The survival times of Lewis and B.N. skin homografts on rats of an F2hybrid population were evaluated with respect to the correspondence between donor and host for these antigens. Homografts did not survive longer than 12 days in any case where an incompatibility existed, although a significant number of grafts were maintained longer on hosts compatible for antigens 1 or 3. The data suggested that the antigens were products of an important histocompatibility locus. Genetic tests showed the genes determining antigens 1 and 3 to be in the same chromosome linkage group as that determining rat antigenB(4) and indicated a possible allelic relationship. Cytotoxic antibodies developed in some F2hybrids following hyperimmunization with Lewis or B.N. tissues. The presence of cytotoxic antibodies in the few F2hybrids tested was unrelated to their possession of the 1 and 3 antigens, suggesting that the Lewis and B.N. strains differ for an additional, independent system of serologically distinguishable antigens.

Using coisogenic strains of mice differing by strong or weak, or strong-plus-weak histocompatibility factors, a study was made of the induction and duration of immunity. Immunization with spleen cells in the presence of only anH-2incompatibility was rapid in onset but of short duration, while immunization across the weaker,H-3barrier was slow in onset and of longer duration. When strong and weak factors were combined, immunity was of shorter duration than when only the strong factor was involved. The long-term immunity that normally results from skin grafting was found to be abolished if the recipient was preimmunized with lymphoid cells. Removal of the regional nodes early in the development of immunity following skin grafting prevented permanent immunity.

Using coisogenie strains of mice, differing only by weak histocompatibility factors, long-lasting tolerance was induced in adult animals by the injection of spleen cells. The interrelationship between tolerance and immunity was explored and a theory proposed based upon the induction or selection of “preimmune” host lymphoid cells that have some degree of specificity for the antigen. It is suggested that these cells, representing an early step in the direction of immunity, are vulnerable to suppression or destruction by excessive doses of antigen.

Embryo cell transplantation antigens have been measured by the titration of isoantisera absorbed with allogeneic embryo cells, using a microcytotoxic test. H-2 antigens appear and increase in 9-day embryos at approximately equal rates in two of the three strains examined; results with a third strain suggest the possibility of strain-dependence of transplantation antigen development. The cells of 9-day mouse embryos are considerably less antigenic than adult lymphoid cells.

Studies were undertaken to investigate the effect of early thymectomy on the induction of immunological tolerance in adult life. C3H mice thymectomized at 6 days of age retain the capacity to reject allogeneic (C3H ± A) F1hybrid skin grafts, but are nonetheless more easily rendered immunologically tolerant to (C3H ± A) F1hybrid skin grafts in adult life than are sham-thymectomized mice. The immunological reactivity of spleen cells from mice thymectomized at 6 days of age is intermediate in magnitude between that of mice thymectomized as newborns and sham-thymectomized mice. The wasting disease associated with thymectomy at birth does not affect mice thymectomized at 6 days of age.

In a randombred population of Swiss-Webster mice a study has been made of survival rates following lethal exposure to radiation, 750 r. When antibiotics were given, infusions of marrow from a pool of 30–60 donors were as effective in inducing survival as infusions of autologous marrow. Survival was less good when the marrow infusions came from a single donor, or from a pool of 15 donors, or from a pool of 120 donors, irrespective of the numbers of cells injected. The implication of these observations and the probable usefulness and limitations of marrow from pools of donors are discussed on the basis of H-2 hetero-genicity known to exist in the population.

In an attempt to obtain more information about human transplantation antigens, substances derived from placental tissue were identified with the use of leukocyte agglutinin inhibition as a test system. The concentration and purification of the active fractions are described. On the basis of its stability to chemical procedures a lipoprotein structure for the active principle seems most likely.
