SUMMARYA monospecific serum which recognizes antigen HL-A2 (Mac) of the HL-A system was systematically absorbed with homogenates of fresh organs of three individuals possessing this antigen. Spleen was shown to be the richest in this antigen, followed in order by lung, liver, intestine, kidney, and heart. Aorta is rather poor while fat and brain contain practically none.

SUMMARYThe effect of topical radiation on heterotopically transplanted cardiac allografts has been investigated in dogs. Seventy-three transplants were done in this series, with 10 animals serving as controls, and the remaining 63 receiving 2400-rad radiation to the graft in 6 equal doses, over a period of 3 weeks. Seventy-five per cent of the radiated grafts maintained viability for periods up to 10 weeks, while 90% of the nonradiated transplants were rejected by the 12th postoperative day. In addition, 12 intact canine hearts radiated with identical dose and fractionation provided histological controls for the radiated transplanted group. Electrocardiographic changes and alteration in lactic dehydrogenase values proved to be the most sensitive parameters of early rejection.

The order of loci of Brachy, tufted, andH-2in linkage group IX has been analyzed in three different backcross experiments. Recombination figures for segmentsT-tfandtf-H-2varied markedly according to strains and alleles involved. The recombination within H-2 antigenic components was accompanied by the recovery of the corresponding chromosome products in classical fashion. These results confirm the orderT, tufted,H-2, 11 (K), 4 (D) and indicate, moreover, that H-2 components 4 (D) and 11 (K) are determined by discrete chromosome units. The variability of recombination results reported in this and other publications points to an unusual degree of chromosomal complexity and genetic instability for chromosome IX of the mouse. At least one new combination of H-2 antigens (new allele) was discovered in mice descending from wild ancestors.

SUMMARYA method for assaying cell-mediated cytotoxicity against cultured target cells in 10 µl of medium is described. The test possesses sufficient sensitivity to detect tumor-specific as well as histocompatibility antigens. Lymphoid cells from the peritoneal cavity, spleen, and peripheral blood exhibited strong cytotoxicity, while lymph node cells were less effective. Reduction of target cell survival was also observed with cells from mice receiving either single or multiple immunizations.

SUMMARYSoluble HL-A transplantation antigens, isolated from spleens of tissue-typed donors, have been examined by diethylaminoethyl ion-exchange chromatography and the several specificities expressed by each individual were shown to be present on separable molecules by examining the peak positions of each specificity in the column fractions. This was done by inhibition of platelet complement fixation or of cytotoxic reactions using well defined HL-A typing sera. A tentative beginning has been made to map the sequence of elution of the molecules carrying different HL-A specific determinants. There is evidence against the view that the products of the first HL-A sublocus have any family relationship as seen by diethylaminoethyl adsorptive capacity. For the second sublocus, there are insufficient data to draw such a conclusion. Certain non-HL-A (leuco) platelet antigens eluted from the columns quite separately from those determined by theHL-Alocus. HL-A antigens having the same properties as those isolated from human spleens can also be obtained from lymphoid cell suspended tissue cultures established from peripheral leucocytes of tissue-typed donors.

SUMMARYImmunosuppressive therapy with high doses of methotrexate beginning 1 day after allogeneic bone marrow transplantation was evaluated for its effectiveness in preventing or delaying the lethal graftversus-host disease in dogs. A short-term regimen of methotrexate for 6 days after transplantation was compared with a long-term regimen during 102 days postgrafting. Unrelated donor-recipient pairs mismatched by canine histocompatibility testing were used. The recipients were given combined infusions of donor marrow and leukocytes to provide a test system in which the graft-versus-host syndrome is acute and rapidly fatal. Recipients were prepared for transplantation by 1200 R of total body irradiation. Five control dogs, not receiving immunosuppressive therapy, died between 9 and 14 days after transplantation with graft-versus-host disease. Eight of the 9 dogs receiving short-term methotrexate treatment died between 14 and 21 days with graft-versus-host disease, and 1 dog lived 136 days. Seven of the 10 dogs given long-term methotrexate treatment died between 18 and 68 days, and 3 dogs are alive more than 180 days after transplantation. Statistical analysis of the survival data demonstrated the superiority of the long-term methotrexate treatment. The results show that stable long-term chimerism can be achieved in mismatched recipient dogs when intensive methotrexate is begun immediately after marrow transplantation and continued for a prolonged period of time.

SUMMARYAdult F1hybrid mice suffering from the graft-versus-host reaction demonstrated increased fecal loss of protein, as shown by the51Cr-excretion test, as well as severe hypoalbuminemia. Histological lesions were evident in the small bowel of all of the animals undergoing runting. The significance of these findings in relation to the wasting observed in F1homologous disease is discussed.

SUMMARYAntimurine thymus sera were produced in New Zealand rabbits by 3 i.v. injections of 1 × 1099viable thymic cells from CBA/J, C57BL/6J, or A/J strain mice on three occasions at fortnightly intervals, followed by bleeding 7 days after the third injection. Groups of CBA/J and C57BL/6J mice, receiving A/J skin grafts, were treated with anti-CBA/J, anti-C57BL/6J, or anti-A/J thymus sera, or with normal rabbit serum, in two dosage schedules. The rabbit anti-A/J thymus serum also was given to A/J mice receiving C57BL/6J skin grafts. The cytotoxicity of each serum for lymphocytes from each of the strains of mice was tested in vitro and their lymphopenic effects were assessed in vivo. Strain-specific effects of the antithymus sera on the survival of skin allografts were shown when they were given in 3 doses following grafting and, perhaps, when CBA/J and C57BL/6J mice were injected daily with the anti-A/J thymus serum. No evidence for such effects could have been predicted from the cytotoxicity of the sera for lymphocytes in vitro nor from their lymphopenic effects.

SUMMARYRabbit sera raised by hyperimmunisation with mouse thymocyte subcellular fractions were strongly immunosuppressive when tested for their ability to prolong the life of mouse skin grafts. Antisera raised against spleen cell fractions were only mildly immunosuppressive, as were antisera prepared by less vigorous immunisation with thymocyte fractions. Different techniques for lysing the cells and fractionating the lysates were examined. A soluble fraction from mouse thymocytes gave rise to a moderately immunosuppressive antiserum. A subcellular fraction from human blood peripheral lymphocytes was used to immunise horses, and the antisera showed in vitro lymphocytotoxicity.
