Urine protein selectivity was determined by the radial diffusion immunoassay method in 32 renal transplant patients. In the 19 patients studied serially normal selectivity (&thetas; = 60° or more) was not seen before 3 weeks post-transplantation even in the presence of excellent renal function. Eight patients achieved normal selectivity within 12 weeks after transplantation and seven more between the 12th and 40th weeks. There was a highly significant correlation between selectivity and both serum creatinine (P< 0.001) and creatinine clearance (P< 0.001). No significant correlation was found between selectivity and the 24-hr urine protein excretion (0.1 >P> 0.05). In all 13 rejection episodes the selectivity was subnormal. During these rejections there were five cases of a sharp decline in selectivity, three with poor selectivity and further deterioration, one with a subnormal but rising selectivity, and, where no earlier specimens were available, four grossly subnormal selectivities. The subnormal selectivity occurring in seven of nine patients on the last determination before a rejection suggested that it was a sensitive indicator of glomerular damage with possibly a predictive capacity. After rejection the selectivity increased by 11–50° in the eight instances in which subsequent specimens were available. Donor source and total ischemia time did not significantly affect the development of normal selectivity.

The effect of adult thymectomy on the immunosuppressive action of antilymphocyte serum was evaluated. Antilymphocyte serum suppressed the immune rejection of a tumor xenograft growing in the hamster cheek pouch. The degree and duration of this immunosuppression was studied in thymectomized and intact animals. Tumor growth was evaluated by comparing tumor incidence and tumor volume in groups of animals bearing tumor implants. Thymectomy at 28 days of age significantly enhances the immunosuppressive effect of antilymphocyte serum and delays recovery from this suppression.

A new method for performing massive skin grafts involving 30–40% of the total body skin was developed. Massive skin grafts were made across H-2 differences in inbred mice. In four different situations of antigenic disparity, the survival time of massive skin grafts was between 2.0 and 2.6 times greater than that of small (1.3 X 1.3 cm) grafts. In contrast, minute (0.3 X 0.3 cm) grafts showed a shorter survival time. To investigate the mechanism underlying the prolonged survival of massive grafts, experiments were done with placement of massive grafts followed by small grafts, and with simultaneous placement of massive and small grafts. The mechanism proved to be complex, with three factors contributing: a nonspecific depression of immune reactivity, possibly due to the stress reaction caused by the initial operation and postoperative trauma; a depression in immunological reactivity specific for the type of skin transplanted, possibly due to transient immune tolerance; and a nonspecific reaction dependent upon the relation between the size of the graft and the cellular dynamics of the rejection process.

The necropsy of an orthotopic cardiac homograft recipient which survived 5 months without immunosuppressive therapy is described. The animal succumbed to biventricular failure caused by valvular lesions and arterial insufficiency due to chronic rejection. The absence of acute myocardial rejection and arteritis is ascribed to an unusual donor-graft histocompatibility.

The relationship between 7 antigenic groups of lymphocytes was studied in 36 families with 144 children and a population of 2208 people. Gene frequencies of the individual antigens were deduced from the phenotype of the subjects in the population and the parents in the families as well as the genotype of the latter. In families these antigenic determinants were inherited en bloc, confirming the concept of Dausset, van Rood, Ceppellini, Amos and others that these antigens belong to a single complex locus, theHL-Alocus. Evidence is presented to suggest that there are two mutational sites in theHL-Alocus. One site corresponds to antigens 1 and 2 (LA series of Payne-Bodmer). The second site is principally defined by the antithetic groups 5 and 6 (roughly 4d and 4c). Two other groups, 3 and 7 (approximately 4a and 4b of van Rood), which behave allelic but are more difficult to define sharply, appear to constitute a partially independent site next to the 5–6 site. The mutational site next to 5 appears to be group 7, whereas the site next to antigen 6 could be either 3 or 7. Thus theHL-Alocus seems to have two sites, of which one is composed of two partially independent subunits.

The microdroplet lymphocyte cytotoxicity test was examined thoroughly in an effort to increase the reproducibility of the test. The discrepancy rate in a large series of tests was reduced from 5.16% at the start of this study to the present 0.95% by introducing certain modifications in the technique. Variables connected with the isolation of lymphocytes, handling of antisera, quality of antisera, amount of complement, incubation temperature, duration of incubation, fixing of reactions, and reading of reactions were studied. The method which has resulted appears to be reproducible, simple, and readily usable on a large scale.

Single daily injections of 400 mg/kg of chloramphenicol in rabbits combined with sublethal irradiation (600R) are well tolerated for long periods, prolong allograft survival and suppress the establishment of skin reactivity to diphtheria toxoid. Chloramphenicol given in this way alone or irradiation alone has little effect on these reactions. The results of this combined treatment are probably due to anti-inflammatory as well as anti-immune effects.

Lifespan and weight of separate, paired 129 strain mice was compared with lifespan and weight of simple skin-anastomosed parabionts, skin-plus-coelomic-anastomosed parabionts, and pseudoparabionts. The results indicate that skin anastomotic parabiosis does not significantly alter the lifespan of normal mice. The addition of coelomic anastomosis to the parabiotic procedure significantly decreased lifespan compared with either separate pairs or simple skin parabionts. Pseudoparabionts had a significantly shorter lifespan compared with any of the other groups. The results also show that the effects of parabiosis are greater on weight than on lifespan. Both types of parabionts weighed less than separate pairs, as did pseudoparabionts.
