SUMMARYThe rosette inhibition assay for immunosuppressive activity of antilymphocyte globulin or plasma has been studied in an effort to improve its reliability. Important changes include the elimination of calcium and magnesium ions from salt solutions used in the assay, the use of deoxyribonuclease to prevent lymphocyte clumping, and pretreatment of plasma samples (heating at 63 C for 10 min followed by acrinol precipitation) to prevent nonspecific inhibition of rosette formation. The use of a graded dose response potency assay against a house standard is discussed. A significant correlation was established between the in vitro activity of several series of antilymphocyte globulin or antithymocyte globulin preparations and their ability to prolong skin graft survivial in primates. The best correlation was achieved using a potency estimate relative to a house standard, rather than the conventional titer estimate.

SUMMARYRejection of a xenograft by BALB/c mice results in a highly potent immune peritoneal cell population. By using two in vitro assays in parallel, it has been possible to show that there are at least two active cell types. The first assay detects the detachment of target cells from plastic, which is referred to as direct cytotoxicity. The second reflects the inhibition of incorporation of a deoxyribonucleic acid precursor,125I-iododeoxyuridine, into target cells, which could occur either as a result of direct cytotoxicity or as a consequence of growth inhibition (cytostasis). With a rabbit antimouse brain serum which is cytotoxic for thymus-derived T cells, it has been shown that direct cytotoxicity requires T cells, whereas the cytostatic response can be largely independent of T cell function during the assay.

SUMMARYAntisera produced against sarcoma I (SaI) cells in C57BL/6 and CBA mice were tested for their cytotoxic activity against normal A lymphocytes and SaI tumor cells, using guinea pig or rabbit complement. Whereas high levels of cytotoxic activity against normal A lymphocytes were revealed with both antisera using either complement, only rabbit complement produced significant lysis of tumor cells under the standard conditions of the cytotoxic test (30 min, 37 C). However, upon prolonged incubation, guinea pig complement also produced high levels of lysis of SaI cells with the C57BL anti-SaI, but not with the CBA anti-SaI serum. Because these two antisera contain antibodies directed mainly against the H-2K and H-2D specificities, respectively, these results seemed to suggest that SaI cells are lysed in the presence of guinea pig complement with anti-H-2K antibodies but not with anti-H-2D antibodies. This conclusion was further supported by the use of antisera of restricted specificity prepared by selective absorption of the C57BL anti-SaI serum with CBA or DBA/2 cells. However, a C57BL anti-A serum which did not contain antibodies capable of lysing SaI target cells on prolonged incubation with guinea pig complement did not inhibit their subsequent lysis with the C57BL anti-SaI serum. These findings might suggest that under these modified conditions, mainly tumor-specific antigen-antibody systems are revealed with guinea pig complement, whereas with rabbit complement, under the standard conditions of the cytotoxic test, all antigen-antibody systems are detected.

SUMMARYThe methodology of the conventional mixed lymphocyte culture test has been simplified and miniaturized by developing a micromethod employing Terasaki trays with a final culture volume of 20 μl containing between 10,000 and 20,000 responding and stimulating cells each/culture. This represents one-tenth the cell concentration of the conventional mixed lymphocyte culture. For this new test, a special microharvester has been designed and developed. The test allows accurate and reproducible results giving a high level of discrimination between the different levels of response.

SUMMARYCross sensitization between the products of theH-2KandH-2Dloci has been demonstrated by an experimental design in which recipients were sensitized with tissue bearing determinants controlled by one locus (e.g.,H-2K), and then challenged with skin grafts expressing determinants controlled by the other locus (e.g.,H-2D). In one combination, experimental grafts were rejected faster than first-set control grafts. Cellular rather than humoral mechanisms appeared to mediate this effect. In a second combination, prolonged survival of experimental grafts were observed. This enhanced survival may have been influenced by serum antibody, either to H-2 or Ia determinants.

SUMMARYMouse skin maintained in organ culture for 7 days was transplanted across the H-Y histocompatibility barrier in two inbred strains: C57BL/6 and C3H/An. Cultured grafts were invariably rejected and at a faster rate than noncultured control grafts. Recipients of the cultured male skin grafts rejected subsequent noncultured male skin grafts in accelerated fashion.

SUMMARYA single transfusion of 200 ml of donor blood 14 days before renal transplantation in prospectively DL-A tissue typed beagle littermates appeared to have an effect on graft survival. Seventeen per cent of the recipients did respond to the transfusion with formation of lymphocytotoxic and haemagglutinating antibodies. These “responder dogs” rejected kidney grafts in an accelerated way, compared with the “nonresponders” and with the nontreated control dogs. Responsiveness appeared to occur in pairs of littermates, which suggests that responding potency is genetically determined. There was histological evidence of acute arteritis in the renal grafts of responders, whereas cell-mediated rejection was noted in nonresponders.

SUMMARYBlocking antibody directed against donor responding cells has been found to develop in HL-A-mixed lymphocyte culture-identical donor-recipient combinations after renal transplantation in which splenectomy was not performed. Cytotoxic antibody associated with rejection episodes and detected only by the discriminatory mixed lymphocyte culture developed in other HL-A-mixed lymphocyte culture-identical combinations as well. It disappeared as blocking antibody appeared and the post-transplant course became uneventful. In addition, both cytotoxic and blocking activity were shown to develop and coexist in a patient who received two successive renal transplants: cytotoxic against donor 1 (kidney rejected), and blocking against donor 2 (kidney accepted). These findings are taken to be strong evidence in favor of qualitatively demonstrable separate antibody molecules dealing with two separate functions, i.e., cytotoxicity versus enhancement or immunoregulation.

SUMMARYCanine cryopreserved autologous peripheral buffy coat cells were infused after an otherwise lethal regime of busulfan and cyclophosphamide. In seven dogs so treated, restoration of hematopoiesis occurred within 7 days, and six dogs recovered completely. Comparable colony-forming unit in agar activity was present in freshly processed and cryopreserved buffy coat cells and proved to correlate with in vivo marrow recovery. It was concluded that the peripheral blood is a convenient source for obtaining cells for autologous marrow reconstitution in canines.
