The effect of graft-versus-host (gvh) reactions was studied at the cellular level. It was found that splenic plaque-forming cells (pfc) and hemopoietic colony-forming units (efu) of nonirradiated adult F1hybrid mice were inhibited following injection with parental strain lymph node cells (107). Suppression of pfc was evident 3–6 days after injection and there was only 1% of the control pfe level by 10 days. It was found that the donor lymph node cells did not contribute significantly to the remaining pfe response. Colony-forming units were depressed in the spleen by 12 clays or earlier if larger numbers of parental lymph node cells (4.7 × 107)were injected. Suppression of efu in the bone marrow was not noted during the 12-day interval studied. These results may be interpreted as evidence that pfe precursor cells and/or efu are primary targets of the gvh reaction, but other indirect mechanisms, which are discussed, are not necessarily excluded.

A large proportion of the sera from 574 parous women was shown to contain cytotoxic antibodies against HL-A tissue antigens. After their second pregnancies, one-quarter of the women had cytotoxins, and after the sixth, one-half of the women had these antibodies. Retrospective studies of the outcome of pregnancies showed that women with antibodies had a significantly higher incidence of infants with congenital anomalies than did those without antibodies. It is postulated that antibodies produced by the mother against incompatible HL-A antigens of the fetus may have a deleterious effect upon the fetus in subsequent pregnancies.

Focal opacities or pocks form on the chorioallantoic membrane after inoculation with competent lymphocytes. Minor Boand major B histoincompatibilities can be discriminated by the sizes of the pocks (mean diameters of 0.32 and 1.02 mm). The number of minor Bopocks is directly proportional to graft-versus-host splenomegaly. The blood of donors differ greatly as initiators of large B pocks (from 1 to 5/mm8of whole blood). This variation is associated with theBgenotype of the donor but appears to be independent of theBgenotype of the embryo. The number of large B pocks is reduced by X-irradiation or Mitomycin-C treatment of donor cells. TheDofor X-irradiation is 94 R, identical with theDofor the suppession of haematopoietic colony-forming units, and the LD50for Mitomycin-C, under standard conditions, is 6.3 µ/ml. Dilution of the inocula reduces the number of large pocks in direct proportion to the reduction in cell number. Neither genetic selection of the donor nor any of the treatments reduce the size of the pocks or delay their appearance. This indicates that each pock probably marks the site of an antigen-initiated proliferation of a single donor lymphocyte, entraining thereby the pathological changes which culminate in the formation of a visible pock.

The immunosuppressive effect of antilymphocyte serum (ALS) and antithymus serum (ATS) on canine lung allografts was investigated. ALS and ATS were produced in horses by injecting either lymph nodes or thymus cells, respectively, from puppies. A total of 38 dogs received an allograft of the left lung. In 6 dogs receiving no ALS, none of the grafts survived more than 1 week. In 6 dogs receiving ALS or ATS daily beginning on the day of grafting, no prolongation of survival of the graft was observed. Of 13 dogs receiving ALS or ATS beginning 1 week before grafting, the graft survival was prolonged to more than 4 weeks in 2 and more than 12 weeks in 1. In another group of 13 dogs, not only the recipients of the graft but also the graft donors were treated with ALS or ATS beginning 1 week before grafting. Two of the grafts survived more than 4 weeks and the remaining 2, more than 12 weeks. In some of the long-term survivors, the gas exchange capacity of the grafted lungs was maintained reasonably well as determined by differential oxygen consumption and arterial blood gas values while the recipient's own lung was breathing pure nitrogen. These results suggest that treatment of canine lung allograft recipients with ALS or ATS should be started before grafting.

The administration of L-asparaginase causes regression of certain lymphoid tumours in rodents, suggesting that the enzyme may inhibit the activity of normal lymphoid cells and thus have an immunosuppressive effect. The effect of L-asparaginase treatment in mice on the survival of skin allografts and on the antibody response to i.p. injected sheep red blood cells was studied. In both experimental situations, the degree of response to the antigenie challenge was reduced. A single dose of asparaginase was effective when given at the same time as the sheep red blood cells and up to 48 hr afterward. The enzyme was unable to diminish the response to a second injection of sheep red blood cells in previously stimulated mice.
