A fraction of rabbit lymph node cells and splenic cells has been prepared by differential centrifugation of sonically disrupted cells: a high‐speed centrifugal sediment from the supernate of a lower speed centrifugation. This cell fraction gives complement fixation with rabbit anti‐rabbit‐leucocyte serum and gives evidence of containing nearly all the complement‐fixing isoantigen of such cells.The antibody in these sera which causes the rejection or suppression of homotransferred lymph node cells can be removed from such sera by absorption with the cell fraction. From the specific aggregates thus obtained the suppressive antibody can be eluted with recovery of 25% of the antibody present in the original serum. The suppressive antibody can be absorbed from such eluates by the cell‐fraction. The antibody thus eluted shows properties of a pseudoglobulin.

Two iso‐antigens of the mouse, not related to any known histocompatibility system, are described as H‐5.A and H‐6.A. H‐5.A is present on the red cells, kidney, testis, and lung of C3H/St mice and in smaller amounts on spleen, brain, and muscle. It has been detected on the red cells of 10 out of 29 sublines tested. H‐6.A has been found on red cells, testis, brain, and spleen, and in smaller amounts on other organs of C3H/He mice. The tissue distribution appears to be roughly similar in C57 and A mice. The antigen has been found on the cells of 13 strains. Both antigens appear to behave as a single unit, and neither has the same strain or tissue distribution as any previously described histocompatibility factor.

The mechanism of rejection of the transplanted dog heart was studied in experiments in vivo and in vitro. Vascular permeability was measured by determining the degree of tissue radioactivity after intravascular injection of radioiodinated albumin (RISA). One hour after transplantation, vascular permeability was increased. This was more pronounced when the rejection was accelerated. In experiments in vitro perfusion of the isolated heart with homologous plasma demonstrated slightly increased capillary permeability one hour after the onset of perfusion; perfusion with sensitized plasma caused a significant rise in vascular permeability. Cellular reaction was absent in the homografted heart one hour after transplantation. In only 3 of 25 perfusions in vitro was cellular reaction present characteristic of homograft rejection. This occurred in hearts perfused with sensitized plasma. The results illustrate that homograft rejection can commence with signs of increased vascular permeability one hour after transplantation. They further show that humoral factors may play a predominant role in homograft rejection.

Burroughs Wellcome 57‐322 and 6‐methyl mercaptopurine have prolonged survival of canine renal homografts. Urethan plus 6‐azauracil achieved moderate prolongation, but cyclophosphamide and vinblastine sulfate were ineffective at the dosages studied.

摘要:
Aspartic acid concentration is increased in liver of foreign bone marrow treated, irradiated mice. The serine and glycine concentration of liver, plasma, and muscle is decreased in these animals. The rate at which aspartic acid is labeled by carbon‐14 injected as carbon‐14 glucose is different in foreign marrow chimeras as compared to isologous marrow treated irradiated mice. This suggests that the increase in aspartic acid of liver is due to a fundamental alteration in metabolism of this amino acid in foreign marrow chimeras.

Fetal circulations in 3 placentas of the monozygotic quadruplets of the nine‐banded armadillo (Dasypus novemcinctusLinn.) are described. In none of the 3 quadruplet sets were vascular communications between the placental sectors of the respective embryos found. The relation of this finding to the tissue incompatibility existing within the armadillo quadruplet sets and its significance in the phenomenon of chimerism are discussed.

Development of homotransplantation antigens was investigated by determining the capability of hemopoietic tissues from C3H fetal, neonatal, or adult mice to induce transplantation immunity in adult LAF1mice. Transplantation immunity in LAF1mice was estimated by measuring the rejection of a challenge dose of adult C3H bone marrow. C3H antigens were detected in 90‐day‐old spleen, neonatal and embryonic (17½‐ and 15½‐day‐old) liver. No antigenicity could be found in 13½‐day‐old liver.

Studies were conducted with rats and guinea pigs to determine whether their platelets share transplantation antigens in common with skin. Relatively high dosage inocula of blood platelets (equivalent in cytoplasm to that present in 4‐60 × 107spleen cells) from B.N. donors inoculated into adult Lewis rats failed to excite more than marginal sensitivities to subsequent skin homografts having the same genetic origin as the platelets. These sensitivities are most probably provoked by contaminating leucocytes. Serum from B.N. rats inoculated with homologous platelet concentrates provoked immediate PCA‐type cutaneous reactions when administered intradermally to Lewis recipients. The i.d. injection of homologous platelet concentrates into guinea pigs specifically presensitized by homotransplantation of skin provoked the formation of delayed type hypersensitivity reactions attributable to the presence of isoantigens in platelets. In the light of the findings of other workers, it seems reasonable to conclude that platelets do share at least some transplantation antigens in common with skin in this species.

The earliest appearance of immunological competence against transplantation antigens has been shown in 3‐day‐old chick skin since when it was grafted on the chorioallantoic membrane weak splenomegaly was induced in the embryo. This has been confirmed by the ability of skin from 2‐day‐old parental strain chicks to reject itself when grafted on genetically tolerant F1hybrid adults. The greatest concentration of lymphocytes appears in chick skin at 15 days of age when grafts of such skin can function as effectively as a source of lymphocytes as adult spleen. Skin from parental strain newly hatched and day‐old chicks contained so few lymphocytes that it was “tolerant‐responsive” and such grafts remained on the genetically tolerant F, adults. These results suggest explanations for many curious phenomena in skin transplantation in chickens.

Several criteria, including survival of chimeras, Fe59uptake, microscopic appearance of stained, fixed tissues, and serological identification of erythrocytes and nucleated hemopoietic cells, have been used to show that peritoneal fluid cells of the mouse can, on transplantation to a heavily irradiated host, proliferate and differentiate into apparently normal, permanent hemopoietic and lymphopoietic tissues. The efficiency of peritoneal fluid cells is far smaller than that of bone marrow cells and appears to be like that of white cells from circulating blood of the normal mouse.
