The two-allele leucocyte group system 4a4bhas been tentatively tested for its influence on histocompatibility. In one individual homozygous for leucocyte antigen 4b, a relatively low incidence of other leucocyte group incompatibilities permitted a fairly reliable assessment of the importance of 4a4bgroup compatibility in skin homograft survival. Grafts obtained from four “compatible” 4b4bdonors appeared to survive longer than grafts obtained from four “incompatible” 4a4asubjects.

The in vivo intrasplonic cloning method for bone marrow cells was applied to studios on the regulation of erythropoiesis. Experiments were made to test whether erythropoietin would reverse the specific suppression of erythroid clones, caused by polycythemia. It was found that the injection of anemic plasma (erythropoietin) elicited the re-formation of erythroid clones. The kinetics of the reversal is discussed in relation to the target cell for erythropoietin and its possible morphogenetic action.

Mice of the New Zealand Black (NZB) strain develop positive antiglobulin (Coombs) tests from the age of 5 months and haemolytic anaemia of the “warm antibody” (IgG) type. They also develop lymphocytic thymomas which are transplantable, and gross splenomegaly and lymphadenopathy caused by extensive proliferation of plasma or reticulum cells. Large amounts of macroglobulin (IgM) are produced by these mice. This is probably associated with the lymphoid-plasma cells identified in impression smears of bone marrow, spleen, and lymph nodes and with the globules of FAS-positive material seen in sections of these organs. The spleen, lymph nodes, and thymus all exhibit similar histological patterns of activity. The first phase, primarly of lymphoid hyperplasia, extends from the age of 3–11 months and is followed by a second phase of massive hyperplasia of the plasma cells and reticulum cells. Similar changes are seen in the lungs and liver while the kidney glomeruli show progressive thickening and hyalinization of the basement membranes. It is suggested that the thymus of NZB mice is unable to control the balance of lymphocytes, plasma cells, and reticulum cells throughout the body and that the animals may be immunologically hyperactive to antigenic stimuli of many kinds. This implies that macroglobulinaemia, autoimmune haemolytic anaemia, leukaemia, and uncontrolled proliferation of plasma or reticulum cells, occurring singly or in combination, may all be expressions of a basic thymic dysfunction.

A congenie pair differing at a weak histocompatibility locus in the first linkage group has boon developed from the A and CBA strains of mice by repeated back-crossing to the A strain and by using the dominant coat colour allele C as a linkage group marker. Grafts of normal skin were used as the test object for determining isogenicity and congenieity. Some of the characteristics of graft rejection in the face of incompatibility at this locus are described.

Chondrocytes isolated from the nasal septal cartilage of almost mature female rabbits by the joint action of trypsin and collagenase were injected into muscles of male rabbits. Cartilage formed after transplantation of chondrocytes contained a high percentage of cell nuclei with characteristic sex chromatin; it was morphologically similar to septal cartilage. In older transplants slight resorption of cartilage was observed.

An isoantigenic lipoprotein isolated from a mouse sarcoma is rendered soluble by the addition of the detergent Triton |MX 100 or by incubation with snake venom. Evidence for the identity of the active agent in venom as phospholipase A was obtained by fractionation of snake venom and through studies of the heat stability of the enzyme. Incubation of the lipoprotein with the purified enzyme resulted in a marked decrease in the amount of lipoprotein-bound phosphatidyl ethanolamine. Lysophosphatide produced by the action of the enzyme on lipoprotein-bound phosphatides appeared to account for the increased solubility of the lipoprotein since the lipoprotein could also be rendered soluble by the addition of lysoleeithin. Lipoprotein brought into solution by added lysoleeithin or through the action of the venom enzyme remained soluble after passage through a Sephadex column and the presence of lysophosphatide in the recovered preparations was demonstrated. Lipoprotein brought into solution by these methods was largely homogeneous by electrophoretic criteria. At pH S.4 the solubilized lipoprotein was highly poly-disperse in the ultracentrifuge, while at pH 12, it showed a single sedimenting boundary (S = 3.9).Retention by Sephadex (5–200 of a small proportion of the antigenic material after incubation with phospholipase A suggests that isoantigenic specificities are present in molecules of molecular weight less than 200,000. The ability of the lipoprotein to absorb antibodies specific for components determined by theII-2locus was increased after digestion with purified venom enzyme and, to a lesser extent, when it was solubilized by added lysoleeithin.
