SUMMARYSkin grafts from C57L/J mice to C3H/HeJ mice arc rejected in 8 days. Rejection is accelerated by injecting C57L/J spleen or lymph node cells 3 hr before skin grafting. The acceleration is greater if the cells are obtained from C57L/J mice previously immunized with sheep red blood cells. This further increase is not due to increased antigenicity of the lymphoid cells obtained from sheep RBC-stimulated donors. Instead, we believe that it is related to the graft-versus-host effect which occurs following the injection of allogenic lymphoid cells, and which tends to weaken host responses. When many of the injected cells are already committed to react against sheep RBC, the graftversus- host effect would be lessened, and the host reaction thereby intensified.

SUMMARYThe51Cr-release assay for measurement of cytotoxic reactions, with its many advantages, has hitherto been applicable only to mouse cell types which occur free from contamination with erythrocytes. A simple method is presented which can selectively lyse contaminating erythrocytes from tissue cell preparations without damaging the tissue cells. The tissue cells can then be labelled and tested by the51Cr-release cytotoxicity assay. The usefulness of the method is shown by demonstrating its success with suspensions of mouse spleen cells and with a hemorrhagic ascites sarcoma.

SUMMARYMice, grafted with allogeneic skin, were given various combinations of 6-mercaptopurine (6-MP), cortisol and plasma from six different pools of rabbit and anti-mouse lymphocyte plasma (ALP) of widely dissimilar potencies. Allograft survival times produced by ALP were frequently prolonged by 6-MP. No combination of 6-MP and ALP gave results which were inferior to those obtained with ALP alone. Cortisol did not alter allograft survival times when given with either ALP alone or in conjunction with both ALP and 6-MP.

SUMMARYThe immunological competence of blood of White Leghorn chicken donors, as measured with the “graft-versus-host” reaction (GVHR), increased rapidly in the first 3 weeks, then at a slower rate reached a plateau level at about the 12th week of age. A comparable age-related elevation in the GVHR level of blood was observed in Game chicken donors, but the magnitude of the host embryo splenomegaly was much greater and the plateau level was attained by the 16th week of age. The age profiles of individual donors and the WL-2 donors as a group indicate that the competence of blood does not rise steadily with increasing age but fluctuates considerably. During the ages of 1-12 weeks, elevated levels of competence occurred frequently at the 3rd, 6th, and 12th weeks, and depressed levels, at the 4th and 8-9th weeks of age. The increase with age in the GVHR capacity was correlated with a corresponding change in mononuclear cell concentration of donor blood. Similar variations in immune competence were observed in the spleen and thymus. The variable nature of alloimmune responsiveness is attributable to fluctuations in the level of competent cells in blood and other tissues. A rapid maturation of the immunological capacity during the first few weeks is inferred from the elevation in the GVHR capacity despite an increase in body size at this time. A less rapid elevation and subsequent maintenance of a “plateau level” during the 4- to 22-week period indicate that the growth more nearly parallels that of body growth.

SUMMARYThe ability of intrasplenic grafts of ovarian and testicular tissue to condition adult animals to accept subsequent skin homografts was investigated utilizing congenic lines of mice differing with respect to their Ii-1, H-3 or H-l and Ii-S -alleles. The results indicated that, although genetic disparity was crucially important in determining the tolerance-inducing capacity of these tissues, they were about equally effective in the three combinations tested. This was the case despite the fact that in situations in which skin grafts were accepted, ovarian grafts survived while those of testicular tissue did not.

SUMMARYThe post-transplant serums of 14 related kidney transplant recipients were studied for the presence of cytotoxin against donor lymphocytes by a cytotoxin tests. Five of the 14 recipients' serums demonstrated cytotoxins; four of these 5 recipients suffered acute homograft rejection. In all 4 individuals the presence of cytotoxin preceded the clinical diagnosis of rejection. Seven of the 14 recipients did not demonstrate serum cytotoxin; only one of these 7 suffered clinical rejection. Three of the 14 recipients had post-transplant serums with suggestive cytotoxin activity. One of these 3 suffered acute rejection several months later which was preceded by serum cytotoxin activity. It is concluded that serum cytotoxin against donor lymphocytes occurs post-transplant in some recipients, and usually precedes acute rejection.

SUMMARYMethotrexate given i.v. followed by large doses of citrovorum factor can modify the allograft reaction and prolongation of renal function can be obtained. Reversal of the allograft reaction has been shown. The drug is, however, extremely toxic in spite of the use of the citrovorum factor and no successful drug schedule was found. White cell depression, however, did not occur.
