SUMMARYSkin grafts were made on inbred rabbits varying in number of generations of inbreeding. For an average of 6, 12, and 16 generations of sib-mating, the percentages of graft acceptances were 12.5, 21.6, and 43.7 respectively. The number of loci with allelic differences was estimated for the parents at zero generation, for the different inbred lines. No females permanently accepted grafts from their male sibs, but the average survival of 106 days was much longer than that for grafts between inbred lines. Natural selection in favor of heterozygosis was discussed.

SUMMARYNormal murine spleen cells were cultured with syngeneic, allogeneic, or hemiallogeneic spleen cells that had been treated with mitomycin- C; subsequent incorporation of tritiated thymidine was measured. Cell donors were genetically alike except for differences at the H-2 locus. When mitomycin-treated cells were mixed with normal cells, thymidine incorporation in allogeneic cultures significantly exceeded that in hemiallogeneic cultures; incorporation in both allogeneic and hemiallogeneic cultures was significantly greater than in syngeneic cultures. No significant difference could be demonstrated between groups of parent-vs.-Fi and Fr-vs.-parent cultures, nor between reciprocal allogeneic combinations. The results support the conclusion that antigenic disparity can induce DNA synthesis in splenic cells in mixed cultures. Conventional immunological incompatibility alone is insufficient to explain all of the results. The ability of the mixed-culture test to distinguish homozygous and hemizygous differences in dosage of antigens did not extend to the recognition of differences in number or in strength of antigens defined by serological tests.

SUMMARY Ehrlich ascites carcinoma transplants in normal and irradiated C3H mice were examined for the presence of y-globulin on their surface. Results obtained by the mixed agglutination test and the antiglobulin consumption test showed y-globulin attached to the surface of tumor cells from immunologically competent hosts. The amount of y-globulin bound to cells from irradiated mice was significantly reduced. Judged by the indirect fluorescence reaction circulating antibody against the tumor cells appeared in the serum after tumor rejection, while it was absent in serum from mice carrying a tumor. Antibody against red cells or leucocytes was not detected in either of these sera. These findings strongly suggested in vivo absorption of antibody by the tumor graft. This antibody might be directed against transplantation tissue antigens as well as tumor specific components on the tumor cell surface.

SUMMARYA new method for assaying human leukoagglutinins has been developed that is a measure of intercellular dissociation and migration of centrifuged leukocytes in capillary tubes by the force of gravity. Comparative studies indicate that the capillary migration method is 10-100 times more sensitive than microscopic agglutination. It, is highly reproducible and rapidly performed. Utilizing sera from the National Institutes of Health serum bank, the method has been adapted for leukocyte typing and cross-matching. The increased sensitivity appears to account for the higher incidence of reproducibly positive responses determined by capillary migration than by microscopic agglutinations. This was most prominent with antisera primarily developed for use in cytoxicity tests. The method is simple, requires no special equipment, and may be partially automated.

SUMMARYAlice bearing Ehrlich ascites carcinoma show a modified immune response to primary and secondary skin allografts. The present experiments were undertaken to investigate the role of Immoral factors in cell-free Ehrlich ascites fluid (CFEAF) in the reaction between host and allograft. Administration of CFEAF did not alter rejection of primary skin allografts. However, accelerated rejection of second and third allografts was suppressed by continued administration of CFEAF. Rejection of the second and third allografts did occur, but the rejection pattern and survival time were typical of primary graft rejection. Selective inhibition of the immunological memory of the host, indicated by suppression of accelerated secondary graft rejection, taking place in the presence of a repeated and concurrent “primary” immunological response to a second allograft, suggests there is a qualitative difference between the primary and secondary immune response to cellular antigens. Further investigation of the mechanism by which CFEAF selectively suppresses the secondary immune response may lead to a better understanding of this important biological process and additionally, may provide information about the growth of tumor cells in immunologically competent hosts.
