SUMMARYFour normal human subjects were immunized with alligator erythrocytes. The development of in vitro lymphocyte blastogenesis and humoral antibody to these cells was followed at serial intervals after immunization. Humoral antibody was not detected in preimmunization samples of serum. Significant in vitro lymphocyte blastogenesis to alligator erythrocytes was seen with all four subjects prior to immunization. This reaction represented either a primary immune response in vitro or an example of antigenic cross reactivity. Following immunization, there was a rapid increase in the blastogenic response. This effect was noted before antibody was detected in the serum and persisted long after antibody could no longer be measured. There was, however, a correlation between antibody formation and in vitro lymphocyte blastogenesis. It is concluded that in vitro lymphocyte blastogenesis is a more sensitive index for both the development and the persistence of an immune response to a xenogeneic cell than is the measurement of humoral antibody.

SUMMARYSequential serum samples were obtained from horses immunized two different methods of immunization using purified populations rat thymocytes or splenic lymphocytes. In vitro antibody titers were determined, and the ability of each sample to prolong skin graft survival, inhibit acute inflammation, or induce acute toxicity was studied. Although the number of horses immunized was small, the following preliminary conclusions were indicated. (1) The lymphocytotoxicity titers of the sera more closely paralleled the antiinflammatory activity than the immunosuppressive potency. (2) The acute toxic effect of unabsorbed serum correlated with the hemagglutination titer; however, after absorption with erythrocytes, the acute toxicity was independent of the level of hemagglutination, lymphocytotoxicity, and antiinflammatory or immunosuppressive potency. The histological lesions indicated a separate antibody population responsible for this activity, which was induced only by the serum obtained late in immunization from the animal immunized with spleen lymphocytes and not by the serum obtained from the three animals immunized with thymocytes with or without adjuvant. (3) Sera obtained early after both methods of immunization with either antigen were immunosuppressive. The immunosuppressive potency of serum induced with Freund's adjuvant did not decrease with booster injections; however, that induced without adjuvant eventually was lost, as were all other biological activities.

SUMMARYRats injected i.v. with 1 µc/g of32P chromium phosphate showed a significant depression of their antibody synthesis against sheep red blood cells. The phagocytic activity was also reduced. The animals exhibited a marked reduction in circulating mononuclear cells. Skin graft rejection was unaffected. Lymphopenia alone is not a factor sufficient to prevent graft rejection. The possible mode of action of32P chromium phosphate on immune reactions is discussed.

SUMMARYA microprocedure was developed for typing fresh white blood cells as well as those stored at 4 or —20 C. The test does not require viability of the cells. The principle of the procedure relies on absorption of typing sera of known specificity with white blood cells and testing of the absorbed sera for residual antibodies by means of a standard cytotoxicity test with fresh lymphocytes which contain corresponding antigens. The entire test is performed in microliter volumes. Studies performed thus far have allowed reliable typing of white blood cells for several HL-A specificities.

SUMMARYC57BL spleen or lymph node cells cultured for 4 days with mitomycin- treated CBA or A. CA spleen cells were tested in vitro for cytotoxicity against CBA and A.CA fibroblastic target cells. It was found that the cytotoxicity of C57BL spleen or lymph node cells toward target cells of the same genotype as those to which they had been preexposed in such a mixed leukocyte culture was greater than the cytotoxicity toward the other target cell used. This finding shows that immunologically competent cells acquire specific cytotoxicity toward H-2-incompatible target cells carrying the stimulating antigen under the conditions of the mixed culture.

SUMMARYHuman fetal cells grown in tissue culture monolayers were tested using the technique of mixed agglutination and were found to possess some of the antigens controlled by theHL-Alocus. Gestational specimens ranging from 6 weeks through maturity were studied. All fetal homogenates, as well as cells derived from numerous individual fetal organs, demonstrated similar patterns of reaction by mixed agglutination when tested with a panel of 22 human isoantisera. We have been unable to detect HL-A antigens on whole trophoblastic syncytia by direct mixed agglutination testing and have not demonstrated that this material is capable of absorbing out antibodies directed against HL-A specificities. Absorption studies utilizing human spermatozoa also indicate that spermatozoa, like the trophoblastic syncytia, either do not express their transplantation antigens or are lacking in these specificities.

SUMMARYThe effect of previous sensitization to rabbit antilymphocyte yglobulin (ALγG) and the effect of previous paralysis to aggregatefree normal rabbit γ-globulin on a subsequent course of ALγG were examined. Sensitization of mice to ALγG partially reversed the immunosuppressive action of a subsequent course of ALγG on skin allografts, provided that the interval between the sensitizing and challenge doses was long enough. Induction of paralysis to aggregate-free normal rabbit γ-globulin in mice potentiated the immunosuppressive effect of ALγG on skin allograft survival. Aggregated normal rabbit γ-globulin was neither “immunogenic” nor “tolerogenic” in our experiments and did not change the immunosuppressive effect of ALγG. Our study supports but does not prove the contention that the duration of the immunosuppressive effect of ALγG is related to either the production and/or the utilization of anti-ALγG antibody. The clinical implications of these findings are briefly discussed in relation to the use of ALγG in clinical organ transplantation.

SUMMARYHistocompatibility antigens have been detected on melanoblasts and hair follicle cells by means of test grafting of skin from allophenic mice containing two homozygous subpopulations of cells with different alleles at theH-2locus. Melanoblasts bore a visible color marker (e.g.,B/B ↩ b/b) and hair follicle cells were marked at the agouti locus (A/A ↩ a/a). In skin grafts to parental isogenic strains, there was selective survival of those melanoblasts and hair follicle cells whose visible phenotypes were from the sameH-2strain as was the host, while cells of the foreignH-2strain were destroyed. Genotype-specific homograft rejection by the hosts was highly localized to the homologous cells, even when the genotypic cell strains were admixed within a single hair bulb. Donor grafts had some individual hairs with both black and brown pigment granules attributable toB/Bandb/bcells, respectively, or intermediate degrees of agouti banding attributable to presence of bothA/Aanda/acells; only one pure strain contributed to such follicles after rejection of incompatible cells. The morphogenesis of mosaic hairs was strikingly modified when one of the two clonal components was eliminated. Hair follicle H-2 antigens in the in situ skin also sometimes differed from H-2 antigens of the contained melanoblasts, e.g., when C57BL/6 pigment cells entered a BALB/c hair and there produced pheomelanin. “Allogeneic inhibition” did not occur in any of these animals despite the presence of diverse H-2 antigenic cell types.
