AbstractA transient maternal folic acid deficiency resulted in alterations in the concentrations of adenosine triphosphate (ATP), diphosphate (ADP), and monophosphate (AMP) in developing rat embryos. At the end of the treatment period the ATP level was markedly reduced and the pool of adenine (as ATP, ADP, and AMP) was decreased in experimental embryos. AbstractOffspring of rats treated with vitamin A, cortisone, or both drugs were studied with the aid of35S‐sulfate to investigate whether sulfated‐mucopolysaccharides have a role in cleft palate production. Vitamin A alone or combined with cortisone produced a high frequency of cleft palate. Cortisone alone produced no cleft palates. Autoradiographic results showed that35S‐sulfate incorporation in palatal tissue of control fetuses and those from rats treated with both drugs was almost similar, whereas isotope incorporation was several times higher than this in the vitamin A‐treated group and slightly less than this in the cortisone‐treated group. AbstractFifty A × C 9935 strain inbred rat litters were examined near term; 88/315 fetuses (27.9%) showed renal malformations (renal agenesis and hydronephrosis). Unilateral renal agenesis occurred more often on the right side (11.5%) than the left (3.6%), especially in males. Renal agenesis was always associated with absence or hypoplasia of a uterine horn or vas deferens and epididymis on the corresponding side. However, the ovaries or testes were present and intact. Hydronephrosis was seen more often on the left side (11.4%) than the right (3.5%). There was no significant sex difference in the frequency of renal agenesis and hydronephrosis. In hydronephrosis the ureteropelvic junction was patent, and hydroureter always accompanied moderate hydronephrosis. Atresia near the vesicoureteral junction was considered as a cause of hydronephrosis. There was a close morphogenetic relation between renal agenesis and hydronephrosis

AbstractLigation of the uterine vasculature was performed on the 10th or 18th day of gestation in 114 Wistar rats. In each rat the blood vessels of one uterine horn were ligated at the level of the oviduct, midhorn, or the cervix; the opposite horn served as a control. Fetal weight and mortality data were analysed for the effect of the vascular interruption and the effect of intrauterine position. It was found that patency of the ovarian artery was not essential for normal fetal growth and that the loop‐artery configuration of the vessels to the uterine horn was not a determinant of the position effect. The time in gestation at which the ligation was performed had a decided effect on ultimate fetal weight: the later in gestation, the greater the fetal growth retardation. Various factors that could be responsible for the position effect are discussed. It is suggested that the association between fetal weight and intrauterine position is species dependent and is determined by unknown factors, not singly by hemodynamics, myometrial pressure, or time of implantatio

AbstractA technique was utilized for intrauterine deposition of acetazolamide, to study the locus of action resulting in malformations in fetal rats. A 30‐gauge needle, attached to a Hamilton microliter syringe, was inserted through the uterine wall, and acetazolamide was deposited in three different regions. Deposition in the yolk sac and/or decidua capsularis produced typical forelimb deficiency in 7/46 surviving fetuses, while injection into the exocoelom produced 2/9 typically deformed fetuses. In contrast, deposition of acetazolamide into the decidua basalis and/or chorioallantoic placenta produced only 1/46 typically deformed fetus, suggesting that the chorioallantoic placenta is not the locus of acetazolamide action. The small amount of teratogen needed to produce the typical deformity when injected in the yolk sac or exocoelom, plus the total lack of deformed fetuses from control and noninjected implantation sites, eliminated disturbance of maternal metabolism as a causative factor in acetazolamide teratogenesi

AbstractTo test the direct teratogenic effects of trypan blue in chick embryos 1 μl of 0.1% dye (dissolved in isotonic saline or 9‐day chick embryonic serum) was injected into the vitelline veins at 72 hours of incubation. It was carried from the injection site into the embryonic circulation within seconds after injection. The types and sites of abnormalities produced were noted 1 day later. Of the various types of controls, 65% were normal, 18% died within 1 day of treatment, and 17% had abnormalities. None among the experimental specimens was normal, 62% died within 1 day of treatment, and 38% had abnormalities. AbstractPregnant mice were injected once with 10 or 20 mg/kg cyclophosphamide. A large embryolethal effect was induced by treatment on the 3rd and on the 8th and 9th days of pregnancy. Mortality was probably due to a direct effect of cyclophosphamide on the embryos.
