摘要:
In Québec, 83 first-generation seed orchards destined to produce seeds from genetically improved trees have been established since 1973. Consequently, the Tree Genetic Improvement Program uses a very large quantity of pollen in order to carry out the controlled pollination projects in those seed orchards. Moreover, controlled pollination is carried out within a very short time-frame and very often the receptivity period occurs before the time of pollen dispersion. It is for these reasons that the pollen bank was created in Québec. This manuscript summarize the process of long term preservation of a large quantity of tree pollen. It is possible to preserve the pollen of coniferous trees on a large scale for up to five years. It is more difficult to manipulate the pollen of deciduous trees.

The authors attempt to determine the optimal conditions for maintaining long term viability of date palm pollen using a special method of freeze-drying. They have undertaken a systematic study of the viability over time of a sample of the same pollen stored and conserved under different conditions (freeze-drying, temperature, gaseous atmosphere and storage time).

Good preservation and storage of birch pollen (Betula verrucosaEhrh.) is desirable for two reasons; for one it enables a deeper knowledge of the biological state of the pollen and for the other, since birch pollen is a bio-indicator of air quality, its study under optimal conditions (preservation and biological state) at the biological, structural, ionic and immunological level is essential for analysing the effect of atmospheric pollution on the pollen. For this reason this study has involved both pollen collected under natural conditions and pollen from urban sites (in situ). For a period of two and half years we have studied different methods of preservation and different storage temperatures byin vitrogermination tests on a 10% sucrose Heslop-Harrison medium using rehydrated pollen. The best preservation is obtained with freeze-drying for half an hour and subsequent storage at 80°C. Thein vitrogermination test onin situpollen shows that the germination percentage is a reliable parameter for estimating the impact of atmospheric pollution on pollen because it varies in accordance with the degree of pollution at the urban sites.

In botany tentative attempts have been made to classify species, based not on the plant as a whole, but on a particular character, for example pollen characteristics. Pollen, which is involved in the transmission of male genetic information during sexual reproduction, is of particular interest in distinguishing plants and their relationships.

Bactris gasipaesKunth is important both economically and as a food resource in several countries of tropical Africa. The profitability of cultivation can be increased by genetic amelioration of natural morphotypes. We observed that pollen characteristics influence the morphological and physiological features of the fruits. This prompted us to undertake a study based upon ten morphotypes from the Yurimaguas (Peru). We here complete the morphological and ultrastructural investigation of pollen, previously started by one of us, with an analysis of the elements with respect to the importance of the role they play in relation to pollen function. A determination of the 8 major elements, which corresponds to a precise analysis of the exine, was performed by micro-X-Ray diffraction analysis using an electron probe coupled with a scanning electron microscope. At the same time, the determination of calcium, magnesium and twelve oligo-elements by inductive coupled plasma spectrometry (ICP) provided a complete mineral analysis of the pollen. A comparison of the element patterns of the morphotypes, obtained by both was carried out through a number of numerical data reduction techniques (Factor analysis and clustering). This led us to suggest two classifications: (1) that of pollen based upon mineral elements (2) that of mineral elements based upon pollen and further to look for possible multiple correlations between elements and pollens.
