SummaryA procedure is described for the large scale purification and sterilization of ceruloplasmin and apoceruloplasmin. This procedure has the following advantages over others previously described:1Ceruloplasmin and its apoenzyme are prepared in large volumes, using equipment and facilities which are available in any laboratory where the Cohn ethanol method is employed.2The starting material (Fraction IV‐1) is abundantly available and is otherwise wasted where major emphasis is placed on the preparation of Dried Fibrinogen, Antihemophilic Globulin, Immune Serum Globulin and Normal Serum Albumin.3Other useful plasma components contained in Fraction IV‐1 may be recovered without denaturation; for example, Plasma Thromboplastin Antecedent.4The copper‐saturated ceruloplasmin is stable under procedures commonly employed for the destruction of viral contaminants; namely, (a) heat‐treatment for ten hours at 60° C in the final container, or (b) treatment with a variety of concentrations of beta‐propiolactone (at 4° C or 37° C).5The use of batch‐type adsorption and elution with DEAE‐cellulose avoids more tedious procedures such as column chromatography and high speed centrifugation.6The ethanol chloroform denaturation step occurs late in the process where volumes are small and a closed system may be employed. Other proteins denatured in this step occur in trace quantities and workers are protected from chloroform vapors.RésuméLes auteurs décrivent une méthode pour la purification et la stérilisation de grandes quantités de ceruloplasmine et d'acoperuloplasmine. Cette méthode a, sur les méthodes précédeniment décrites, les avantages suivants:1La ceruloplasmine et son apoenzyme peuvent être préparés en grande quatitité dans tout laboratoire équipé pour le fractionnement à l'éthanol selon Cohn.2Le matériel de départ (fraction IV —1) s'obtient facilement et en graiide quantité et est en outre inutilisé quand on prépare du fibrinogène desséché, de la globuline antihémophilique, des iminuns globulines sériques et de la sérum‐albumine normale.3D'autres composants utiles contenus dans la fraction IV‐1 peuvent être récupérés sans être dénaturés, par exemple le PTA (plasma thromboplastin antecedent) ou facteur XI.4La ceruloplasmine saturée en cuivre reste stable malgré les traiteinents usuels ntilisée pour détruire les virus, notamment: a) traitement par la chaleur à 60° C pendant 10 heures, ou b) traitement à diverses concentrations de β‐propriolactone à 4° C ou à 37° C.5L'utilisation de la méthode d'absorption et d'élution avec la DEAE‐cellulose évite les méthodes ennuyeuses telles que la chromotographie sur colonne et les centrifugations à haute vitesse.6Dans cette méthode, l'étape de dénaturation par le chloroforme et l'éthanol se situe tardivement, au moment où l'on opère sur de petits volumes, si bien qu'un système en circuit fermé peut être utilisé. Das Verfahren hat die folgenden Vorzüge gegenüber früher beschriebenen:1Coeruloplasmin und sein Apoenzym können in großen Mengen in jedem Labor, das für Alkoholfraktionierung eingerichtet ist, mittels den dort vorhandenen Geräten gewonnen werden.2Das Ausgangsmaterial, Fraktion IV1, ist in großen Mengen verfügbar. SummaryThe effect of increasing ethanol Concentration and decreasing pH on the separation of Fraction II +III from human plasma on the heat stability of the protein fraction in the supernatant fluid remaining has been studied. When the ethanol concentrations used to separate Fraction II + III were 30 to 35% and the pH and temperature maintained at 6.3 to 6.8 and ‐5° C, respectively, the plasma protein fractions obtained from the supernatant fluids showed electrophoretically, an albumin concentration of 88 to 92%. On heating for 10 hours at 60° C very little of a fast moving component in the ultracentrifuge and very little change in the electrophoresis patterns were observed. Nephelometric changes due to heating were also quite small. The ratio of the area of the main ultra centrifugal component of the heated fractions to that of the unheated was 0.97 to 0.90.Thus, these findings stand in contrast to the results obtained with earlier methods which approximated the corresponding steps of the Cohn group [1]. When the ethanol concentration used to separate the Fraction II + III proteins was 25%, the fractions obtained from the supernatant fluid showed electrophoretically, an albumin concentration of 80 to 83%. On heating of this fraction for 10 hours at 60° C the amount of fast moving component in the ultracentrifuge was larger and the change in the electrophoretic patterns was greater than those observed above. Nephelometric changes were also greater. The ratio of the areas of the main ultracentrifuge component of the heated fraction to that of the unheated was 0.92.RésuméL'effet de l'augmentation de la concentration de l'éthanol et de la diminution du pH sur la séparation des fractions II et III du plasma humain et sur la thermostabilité des fractions protéiniques dans le liquide surnageant résiduel a été étudié. Quand les concentrations d'éthanol utilisées pour séparer la fraction II + III est de 30 à 35%, et, le pH et la température maintenue de 6,3 à 6,8 et à ‐5° C respectivement, les fractions plasmatiques obtennes à partir du surnageant montrent à l'analyse électrophorétique une concentration d'albumine se situant entre 88 et 92%. Après chauffage de 10 heures à 60' C, il y a très peu de composant à sédimentation rapide à l'ultracentrifugation et l'image électrophorétique ne présente que de très faibles modifications. Les modifications nephelométriques dues au chauffage sont aussi très faibles. Le rapport des surfaces des composants principaux observés à l'ultracentrifugation entre les fractions chauffées et non chauffées est de 0,97 à 0,98.Ainsi, ces découvertes contragtent avec les résultats obtenus avec les produits de fractionnement fabriqués selon les anciennes méthodes dérivdées des données du groupe de Cohn [1]. En effet, lorsque la concentration en éthanol pour la préparation des fractions II+III du plasma est de 25%, les fractions obtenues dans le liquide surnageant montrcnt à l'analyse électrophorétique une concentration d'albumine de 80 à 82%. Après chauffage de cette fraction à 60° C pendant 10 heures, la quantité du composant à sédimentation rapide à l'ultracentrifugation est plus grande, et les modifications de l'image électrophorétique sont plus marquées que celles observées avec la méthode précédente. Les modifications nephelométriques sont aussi plus accentuées. Le rapport des surfaces des composants principaux observés à l'ultracentrifugation entre les fractions chauffées et non chauffées est de 0,92.Zusammenfassung:Der Einfluß steigender Äthanolkonzentrationen und sinkender pH‐Werte bei der Abtrennung der Cohnschen Fraktion II+III aus humanem Plasma auf die Wärmestabilität der Proteinfraktion in der überstehenden Lösung wurde untersucht. Sofern die Äthanolkonzentratiou zur Abtrennung der Fraktion II + III auf 30–35% erhöht wurde, während das pH 6,3–6,8 und die Temperatur ‐5° C betrugen, wies die im Überstehenden verbleibende Proteinfraktion einen Alburningehalt von 88–92% auf. Nach dem Erwärmen einer 5%igen Lösung dieser Fraktion während 10 h auf 60° C waren in der Ultrazentrifuge nur eine sehr geringe schnellere Komponente und in der Elektrophorese nur kleine Änderungen featstellbar. Die Trübungszunahme durch die Wärmebehandlung war gering. Das Verhältnis der Hauptkomponente der erhitzten zur unerhitzten Lösung in der Ultrazentrifuge war 0,97 bis 0,98.Diese Befunde sind im Widerspruch zu den mit älteren Methoden, die sich an die Cohnschen Verfahren [1] anlehnten, gefundenen Ergebnissen. Wenn die Äthanolkonzentration zur Abtrennung der Fraktion II + III 250% betrug, zeigte die Proteinfraktion in der überstehenden Lösung einen Albumingehalt von 80–82%. Nach der Wärmebehandlung dieser Fraktion während 10 h bei 60° C war die schnelle Komponente in der UZ größer und die Äderungen in der Elektrophorese waren stärker ausgeprägt. SummaryThe modifications put on the alcohol fractionation procedure ofNitschmann., KistlerandLergier[8] since 1954 are communicated. The yields and purities realised were calculated from the results of about 150 fractionation runs, 100 1 of plasma each. The actual method allows the isolation of 48% of the total plasmaproteins as albumin (98% pure in electrophoretic analysis), 9,8% as γ‐Globulin (practically 100% pure) and 3,4% as clottable fibrinogen. A scheme summarizes the details of the entire procedure.RésuméLes modifications apportées depuis 1954 à la méthode de fractionnement deNitschmann, KistleretLergier[8] sont décrites. Le rendement et la pureté des differents fractions ont été calculées à partir des résultats de 150 fractionnements de 100 litres chacun. La méthode actuelle permet d'obtenir 48% des protéines plasmatiques totales sous forme d'albumine (degré de pureté électrophorétique de 98%), 9,8% sous forme de γ‐globuline (degré de pureté pratiquement de 100%) et 3,4% sous forme de fibrinogène coagulable. SummaryThe serum proteins of seven primates are studied by immunoelectrophoresis using heterologons immune serums anti‐Man, anti‐Cynomolgusandanti‐Lemur. The individual patterns are described and the homologies of a certain number of proteins were established with thofie of the reference species (Man,CynomolgusandLemur). The electrophoretic mobilities of different proteins are calculated. Zoological implications are discussed.RésuméLes protéines sériques de sept espèces de primates ant été étudié es en immunoélectrophorèse au moyen d'immunsérums hétérologues anti‐Homme, anti‐Cynomolguset anti‐Lemur. Les images immnno‐Clectrophoritiques sont décrites et les homologies de certaines protéines ant été établies avec celles des trois espèces de référence (Homme,CynomolgusetLémur). Les mobilités électrophorétiques de différentes protéines ant été calculées. Summary1Complete leuco‐agglutinin was found in a total of 17.5% of a group of pregnant women comprising 52 unpaired and biparae, where leuco‐agglutinin was found in 6%, and 262 women having had three or more pregnancies, in which case leucoagglutinin was found in 20%. The frequency of leuco‐agglutinin was found to rise with the number of pregnancies. Leuco‐agglutinin was, however, also found in non‐transfused primogravidae.2In this material incomplete leucocyte antibody was found in non‐transfused pregnant women, but in the case of pregnancies it was of much more rare occurrence than complete, thus it was found by Coombs' consumption test in four out of 55 pregnant women with complete leuco‐agglutinin. Three of these four women had never received blood transfusions.3It was not possible to establish any relation between the frequency of abortion in the women who were examined and the occurrence of leuco‐agglutinin in their serum.4The frequency of premature delivery, however, was found to be significantly higher in women with leuco‐agglutinin than in women without such agglutinin.5Marked neonatal neutropenia was not found in the offspring of mothers of this series with leucocyte antibody in their serum. No difference in the leucocyte averages was found, whether or not the leuco‐agglutinin could be proved to have passed the placenta. The leucocyte averages, however, were found to be significantly lower in children whose mothers had incomplete leucocyte antibody in their serum than in children whose mothers had complete ‐ but no incomplete ‐ leucocyte antibody.Résumé1° Les leuco‐agglutinines ant été trouvées chez 17,5% d'un groupe de femmes enceintes comprenant 52 unipares et bipares ‐ où 6% prbsentaient des leuco‐agglutinines ‐ et 262 femmes tripares ou pluripares ‐ où 20% ant présenté des leuco‐agglutinines. La fréquence des leuco‐agglutinines augmente avec le nombre des grossesses; mais on rencontre aussi des leuco‐agglutinines chez les primipares n'ayant jamais requ de transfusions.2On a trouvé aussi des anticorps anti‐leucocytaires incomplets chez des femmes enceintes non transfusées. Toutefois, en présence de grossesses, la fréquence des anticorps incomplets est inférieure à la fréquence des anticorps complets. En effet, grâce à la méthode de consommation de l'anti‐glohuline humaine, ces anticorps incomplets ant été trouvés chez 4 femmes enceintes parmi les 55 qui présentaient des leuco‐agglutinines complètes.3II n'a pas été possible d'établir une quelconque relation entre la fréquence des avortements et la présence des leuco‐agglutinines chez les femmes examinées.4La fréquence des accouchements prématurés, par contre, est augmentée d'une manière significative chez les femmes présentant des leuco‐agglutinines par rapport à celles qui n'en possèdent pas.5II n'a pas été observé de neutropénie marquée du nouveau‐né chez les enfants de femmes présentant des leuco‐agglutinines sériques. II n'a pas été constaté de différence du nombre moyen des leucocytes, qu'il yait transfert diaplacentaire ou pas des leuco‐agglutinines. Mais le nombre moyen des leucocytes est nettement plus bas chez les enfants de mères présentant dans leur sérum des anticorps anti‐leucocytaires incomplets que chez les enfants de mères présentant dans leur sérum des anticorps anti‐leucocytaires complets sans anticorps incomplets.Zusammenfassung:1Eine Gruppe von Schwangeren wurde auf das Vorkommen von Leukoagglutininen untersucht. Solche fanden sich bei 17,5% der untersuchten Frauen. Bei 52 Erst‐ und Zweitgebärenden betrug der Prozentsatz 6%; bei 262 Dritt‐ und Mehrgebärenden betrug er 20%. Die Häufigkeit des Vorkommens von Leukoagglutininen steigt somit mit der Zahl der Schwangerschaften an. Es wurden aber auch bei Primigravidae, welche nie Transfusionen erhalten hatten, Leukoagglutinine gefunden.2Im vorliegenden Untersuchungsgut wurden auch bei Schwangeren, die nie Transfusionen erhalten hatten, inkomplette Leukozytenantikorper gefunden. Diese sind allerdings bei Schwangeren viel seltener als die kompletten Antikörper. So wurdeu mittels des Coombs‐Konsumptionstestes bei 55 Schwangeren, deren Seren komplette Leukoagglutinine enthielten, nur bei vier inkomplette Leukoantikörper gefunden. Drei dieser vier Frauen hatten nie Bluttransfusionen erhalten.3Im vorliegenden Untersuchungsgut fehlt eine Beziehung zwischen dem Vorkommen von Leukoagglutininen und der Anzahl der Aborte.4Hingegen waren Frühgeburten bei Frauen mit Leukoagglutininen signifikant haufiger als bei Frauen, die keine solchen Antikörper aufwiesen.5Bei den Neugeborenen von Müttern mit Leukozytenantikörpern wurde keine ausgesprochene Netropenie gefunden. Ebenso fand sich in jenen Fällen, bei denen die Plazentarpassage der Leukozytenantikörper nachgewiesen werden lconnte, kein Unterschied der Leukozytenzahl gegenüber jenen Fällen, bei denen der Nachweis der Passage nicht erbracht wurde. SummaryA haemolytic transfusion reaction is reported in which no incompatibility was demonstrable in vitro. The determination of the survival time of51Cr‐tagged red cells from several donors showed markedly different disappearance rates.
