AbstractMononuclear cells, present in bone marrow and peripheral blood, have been isolated from red cells and granulocytes using a ficoll‐hypaque density centrifugation process. Cells isolated by this process which uses centrifuge tubes may become contaminated. In 19 studies in our laboratory we used Ficoll‐Hypaque treatment to isolate mononuclear cells from cellular residues obtained during plateletpheresis using a modified 600‐ml polyvinylchloride (PL‐146) plastic bag with the Haemonetics blood processor V‐50 or the Fenwal CS‐3000 blood processor. The 600‐ml PVC plastic bag was modified by sealing its vertical edges using radio frequency to form a narrow bag with a volume of approximately 200 ml. A 125‐volume of diluted apheresis cellular residue was collected, and the mononuclear cells were isolated as follows: the diluted cellular residue was layered onto 75 ml of Ficoll‐Hypaque with a specific gravity of 1.077 and was centrifuged at 260gfor 30 min at 22°C. The supernatant plasma was removed. The mononuclear cell layer was transferred to a sterile 600‐ml transfer bag, and the cells were washed with saline. Of the 4.24d±0.9 times 109mononuclear cells applied to the gradient, approximately 3.73±0.8times109cells were recovered. The recovered cells consisted of 77.3±8% lymphocytes, 19.0±7% monocytes, and 3.6±3% granulocytes. There was no significant difference in tissue culture growth in the CFU‐GEMM assay of mononuclear cells whether the plastic tube or the plastic bag system was used. Aerobic bacteriologic cultures were negative. The PL‐146 plastic bag system used in this study proved to be a significant aid in isolating mononuclear cells

AbstractHepatitis B virus infectivity can be removed from human plasma used to prepare a gamma‐globulin product by ion‐exchange chromatography with commercially available resins. A pooled human plasma sample free of hepatitis markers was contaminated with 10,000 chimpanzee infectious doses of hepatitis B virus per milliliter from a known infectious inoculum. The contaminated plasma was diluted to yield a weakly positive radioimmunoassay result for HBsAg and then processed over a double‐column chromatography system containing anionic ion‐exchange resins to yield highly purified gamma‐globulin. Two chimpanzees were each inoculated intravenously with this product while a control animal received non‐treated material. The 2 experimental animals each received a potential challenge of 3,000 ID and showed no serologic evidence of the disease during 9 months of evaluation. AbstractThe hypothesis that experimentally determined survival times ofTreponema pallidumin stored donorblood could be related to the number of treponemes initially present in the treponeme‐blood mixtures was investigated by inoculating rabbits with three graded doses of treponemes suspended in donor blood and stored at 4°C for various periods of time. The storage periods up to which living treponemes could be detected in the testes of the inoculated rabbits as well as those storage periods up to which seroconversion occurred are related to the number of treponemes present in the treponeme‐blood mixtures. Increasing numbers of treponemes present in the donor blood resulted in longer periods up to which positive results in both parameters were found. All available evidence suggests that the upper limit of seroconversion coincides with the upper limit of treponemal survival. AbstractA positive direct antiglobulin reaction of a baby's cells, found during antenatal testing at Cardiff, was caused by a low‐frequency antigen which has been identified as Rea(ISBT No. 700019). Investigations with this, only the second example, anti‐Rearevealed another Re(a+) propositus in testing 6,635 random donors at Oxford; no further positive was found in 4,770 random donors at Cardiff.
