SummaryA method to prepare stable ceruluplasmin concentrates in one step from different globulin fractions is described.In an acetone‐temperature variable system at constant pH and ionic strength, the lipoproteins are separated from ceruloplasmin. Because of their high cerulo‐plasmin content residual globulin fractions from retroplacental serum are used as starting material.For further fractionation of the acetone extracted ceruloplasmin concentrate an adsorption chromatography method using DEAE‐cellulose or DEAE‐Sephadex is employed under such conditions that only ceruloplasmin is adsorbed to the ion exchanger. The rest of the proteins are filtered off and the ceruloplasmin is eluted at 0.3 M sodium chloride concentration and pH 6.8. In using DEAE‐Sephadex, an elution technique is described which gives a ceruloplasmin concentration of more than 5 % in the final eluate.The method may be applied to smaller scale laboratory experiments as well as larger scale batch processing. Both variants of the method are described.After sterile filtration the ceruloplasmin solution is stable for years at + 4°C, if protected from light.In intravenous administration to animals and humans the ceruloplasmin solutions obtained by this method have been found non‐toxic, non‐pyrogenic and free of significant side‐effects.RésuméOn décrit une méthode en un temps pour préparer des concentrés stables de céruloplasmine à partir de différentes fractions de globulines.Les lipoprotéines sont séparées de la céruloplasmine dans un système à l'acétone à température variable à PH et force ionique constante. Par suite de leur haute concentration en céruloplasmine, on utilise comme matériel de départ les fractions de globulines résiduelles provenant du sérum rétroplacentaire.Ensuite, le fractionnement du concentré de céruloplasmine extraite par l'acétone est obtenu grâce à une méthode chromatographique d'absorption utilisant la cellulose DEAE, ou le Sephadex DEAE de telle manière que, seule, la céruloplasmine soit absorbée sur l'échangeur d'ion. Le solde des protéines est filtré et la céruloplasmine est éluée par une solution de chlorure de sodium 0,3 M à PH 6,8. On décrit une méthode d'élution avec le Sephadex DEAE qui donne une concentration de céruloplasmine de plus de 5% dans l'éluat final.Cette méthode peut être appliquée aussi bien pour l'étude expérimentale au laboratoire que pour la préparation de grande quantité. Globulin‐Fraktionen von Retroplacentar‐Serum werden wegen ihres hohen Coeruloplasmingehaltes als Ausgangsmaterial bevorzugt.Die weitere Fraktionierung des Aceton‐extrahierten Coeruloplasmins geschieht durch Chromatographie auf DEAE‐Cellulose oder DEAE‐Sephadex. Dabei werden die Bedingungen so gewählt, daß nur das Coeruloplasmin an den Ionenaustauscher gebunden wird. Die übrigen Proteine werden abfiltriert. Das Coeruloplasmin wird dann mit 0,3 M Natriumchloridlösung bei pH 6,8 eluiert. Die beschriebene Méthode gestattet es, bei Gebrauch von DEAE‐Sephadex, Coeruloplasmin‐Lösungen mit einer Konzentration von mehr als 5% im letzten Eluat herzustellen.Die Méthode kann sowohl im Labormaßstab wie auch für die Präparation größerer Mengen im «batch»‐Verfahren angewandt werden. Summary1Young red blood cells are slightly more resistant to mechanical trauma and destruction induced by rapid freezing and thawing than are older cells.2A mathematical model is presented which takes account of the random destruction and destruction according to age. SummaryAntibodies to both red cell and colon antigens were detected in the serum of a patient with ulcerative colitis and Coombs'‐positive hemolytic anemia. The erythrocyte‐coating material proved to be a 7 S γ‐globulin, as found in typical acquired hemolytic anemia of the warm antibody type. No cross reactivity between the erythrocyte and colon antibodies could be demonstrated in absorption studies. A possible explanation for the simultaneous presence of both antibodies in this patient lies in the hypothesis of an underlying genetic predisposition to aberrant immunologic response in patients with ulcerative colitis.RésuméDes anticorps contre les érythrocytes et les antigènes coliques ont été mis en évidence dans le sérum d'un patient atteint de colite ulcéreuse et de maladie hémolytique avec Coombs positif. On a pu démontrer que les substances fixées aux érythrocytes étaient des γ‐globulines 7 S, comme on les trouve dans les anémies hémolytiques acquises à autoanticorps du type chaud. Aucune action croisée entre les anticorps anti‐érythrocytes et anti‐colou n'a pu être démontrée grâce aux tests d'absorption. Summary1The inhibiting effect of soluble and tissue fixed human gamma globulin (HGG) on the hemagglutination of anti‐D (Rho) sensitized red blood cells by Coombs serum, called Antiglobulin Consumption Test (AGC‐test), have been compared.2The amount of HGG required to give a certain consumption was about five times larger for tissue fixed than for soluble HGG. The minimum amount of HGG detectable averaged 1.0 μg/ml solution for soluble and 5.1 μg/ml suspension for tissue fixed HGG. Steric hindrance, differences in time and rate of reaction are mentioned as possible explanations for this difference.3Statistical evaluation demonstrates the high variance of the total of our tests on the basis of 95% probability of occurrence. Individual test groups (tests performed simultaneously on one plate) seem to have less variability. The consequences for the practical performance of the AGC‐test are discussed and approximative amounts of tissue fixed gamma globulin for various results of a routine AGC‐test are calculated. The test is qualified as a semiquantitative technique.Résumé1L'effet inhibiteur des gamma‐globulines humaines (GGH) en solution ou fixées aux tissus sur le test d'hémagglutination d'érythrocytes D (Rho) sensibilisés par l'anti‐D (Rho) incomplet et mis en présence de sérum anti‐globuline ou test de consommation de l'antiglobuline (test de CAG) ont été comparés.2La quantité nécessaire de GGH pour donner une certaine consommation est environ cinq fois plus grande pour les gamma‐globulines fixées aux tissus que pour les gamma‐globulines solubles. La quantité minimum de GGH décelable est de 1 μg/ml de solution pour les gamma‐globulines solubles et 5,1 μg/ml de suspension pour les gamma‐globulines fixées aux tissus. Un empêchement stérique, des modifications dans le temps et le degré de réaction sont mentionnés, comme des explications possibles à ce phénomène.3L'étude statistique démontre la haute variance du total de nos tests, sur la base d'un coefficient de sécurité de 95%. Les groupes de tests individuels (tests pratiqués simultanément sur une seule plaque) semblent présenter une variabilité moindre. Ce test est considéré comme une technique semi‐quantitative.Zusammenfassung1Es wurde die Hemmwirkung von gelöstem und gewebsfixiertem menschlichem Gammaglobulin (HGG) auf die Hämagglutination Anti‐D‐(Rho)‐sensibilisierter Erythrozyten durch Coombs‐Serum [sog. Antiglobulinkonsumptionstest (AGC‐Test)] vergleichend untersucht.2Falls das HGG an Gewebe fixiert war, war die für einen gegebenen Antiglobulinverbrauch benötigte HGG‐Menge ca. fünfmal größer, als wenn sich das HGG in Lösung befand. Die Nachweisgrenze lag bei gelöstem HGG bei 1,0 μg/ml, bei gewebsfixiertem HGG bei 5,1 μg/ml. Dieser Unterschied beruht offensiehtlich auf sterischen Behinderungseffekten. Als weitere Ursachen sind aber auch Unterschiede im zeitlichen Ablauf der Reaktion in Erwägung zu ziehen.3Bei der statistischen Bearbeitung der Ergebnisse zeigte sich auf der Stufe der 95 %‐Wahrscheinlichkeit eine erhebliche Streuung. Einzelversuchsansätze auf derselben Platte zeigen jedoch eine geringere Streuung. SummaryThe authors have adapted the Auto‐Analyzer to the performance of Kidd and Duffy blood typings. Coombs antiglobulin serum is not used in the procedure. Kidd typing is done with papain‐treated blood. SummaryA severe haemolytic transfusion reaction due to Wraincompatibility is described. It was characterized by jaundice and oliguria; the latter lasted for 16 days and necessitated three courses of haemodialysis. Although the patient recovered from the immediate effects of the transfusion reaction, she died several weeks later from complications.The anti‐Wraantibody in the patient's serum was of natural origin; nevertheless, its mode of action was strictly incomplete. A similarly incomplete natural anti‐Wrawas present in the serum of the patient's daughter; two further examples of incomplete natural anti‐Wrawere observed, one of which was found by serial tests on 160 normal sera.These findings are discussed in conjunction with the extreme rarity of haemolytic transfusions reactions due to anti‐Wra, of which only one example has been published previously.Among 3,753 Zürich blood donors tested for the Wraantigen, two Wr(a+) persons were found. The pedigree of one of these, which shows independent segregation ofWraof the gene responsible for Lewis red cell groups and ofFya, is presented.RésuméUne réaction hémolytique grave due à une incompatibilité transfusionnelle selon le système Wraest décrite. Cette réaction s'est manifestée par un ictère et de l'oligurie; cette dernière persista pendant 16 jours et nécessita une hémodialyse par trois fois. Un anticorps naturel anti‐Wraidentique a été trouvé dans le sérum de la fille de la malade; deux autres exemples d'anti‐Wranaturel incomplet ont été observés, dont l'un a été trouvé en pratiquant des examens en série sur 160 sérums normaux.Ces données sont discutées sur la base du fait que les réactions transfusionnelles hémolytiques dues à un anti‐Wrasont extrêmement rares; un seul exemple a été décrit jusqu'alors.Parmi 3753 donneurs de sang de Zurich, deux personnes seulement ont été trouvées Wr (a+). Obschon sich die Patientin von den unmittelbaren Folgen der Transfusionsreaktion erholte, verstarb sie mehrere Wochen später an deren Spätfolgen.Bei dem im Patientinnenserum gefundenen Anti‐Wrahandelte es sich trotz seines rein inkompletten Charakters um einen natürlichen Antikörper. Bei der Tochter der Patientin wurde ein gleichartiger inkompletter natürlicher Anti‐Wra‐Antikörper gefunden. Zwei weitere Antikörper dieser Art wurden beobachtet. Der eine wurde bei einer Reihenuntersuchung von 160 Normalseren gefunden.Diese Beobachtungen werden im Hinblick auf die extreme Seltenheit von hämolytischen Transfusionsreaktionen, bedingt durch Wra‐Inkompatibilität, diskutiert. Bis anhin ist lediglich ein einziger Fall einer solchen Reaktion beschrieben worden.Unter 3753 Zürcher Spendern wurden zwei Wra‐positive Personen gefunden. Résumé239 sérums de Japonais ont été étudiés avec 7 SNagg anti‐Gm(b) dont la spécificité avait été vérifiée sur un panel de 50 sérums de sujets de race blanche. Les résultats obtenus démontrent que dans la population Japonaise le facteur Gm(b) est constitué par au moins 3 éléments définis sérologiquement et permettant de décrire 5 phéno‐types différents.Des exemples sont rapportés, montrant que dans les autres races, et en particulier chez les noirs, le phénomène est d'une plus grande complexité.Quelques faits précis permettent de penser que cette structure en mosaïque n'est pas l'apanage du facteur Gm(b), et qu'également les facteurs Gm(a) et Gm(x) ne sont pas des unités insécables.SummaryThe sera of 239 Japanese persons were tested with 7 anti‐Gm(b) antisera of the SNagg type whose specificity had been verified on a panel of 50 Caucasian serum samples. The results obtained show that Gm(b) in the Japanese is made up of at least 3 elements: these are serologically well defined and enable one to recognize 5 distinct phenotypes.Further results are reported which indicate that the complexity of Gm(b) is still greater in other races, especially in Negroes.Certain observations suggest that this mosaic structure is not limited to Gm(b) but that it may also apply to Gm(a) and Gm(x).ZusammenfassungMit 7 Anti‐Gm(b)‐SNagg‐Seren, deren Spezifität an 50 Serumproben von Angehörigen der weißen Rasse erhärtet worden war, wurden 239 Seren von Japanern untersucht. SummaryThe extract ofCrotalaria mucronataseeds contains anti‐A + B agglutinins and detects, like that of aPhaseolus lunatusvariant, considerable differences among B samples which by both lectins are put in the same order of sensitivity.Variation is continuous, all B samples becoming positive under suitable conditions while O samples remain negative. SummaryPlant agglutinins are described which act more strongly as temperature is decreased.Crotalaria mucronataas well as variants ofPhaseolus lunatuscontain pronounced cold agglutinins against B cells while their action on A1‐cells is not enhanced with decreasing temperature. So the structures for anti‐A and anti‐B action which are assumed to be situated in one and the same lectin molecule would display different susceptibilities to temperature. However, such lectins resemble other anti‐A + B lectins in that eluates fromA1as well as from B cells behave qualitatively like the original extract except that O cells fix anti‐A1ofPhaseolus lunatusextract. The two specificities are hardly or not at all separable.RésuméDes phytoagglutinines sont décrites, dont l'action augmente lorsque la température diminue.
