AbstractInheritance of an excess of immunoglobulin allotypes in one haplotype was encountered which could not be explained by the assumption of a duplicated locus. The surplus of allotypes was related to markers on the CH3 domain of γ3 chains. Two such cases were investigated extensively. The IgG3 molecules were isolated by gel filtration and by absorption on protein A. Only the usual combination of allotypes appeared to be present on the IgG3 molecules. The supernumerary markers were found in one case on IgG2 molecules and in the other case on IgG1 molecules. This followed from investigations of eluates after separation of the subclasses by immune absorptions. A hypothesis was proposed to explain these events by mutation of a particular position of an otherwise homologous stretch of γ‐subclass

AbstractA new technique has been developed to absorb selectively autoantibodies from sera. Red cells maximally coated with IgG antibodies are subjected to discontinuous, combined density and pH gradient centrifugation. The method yields a large quantity of antibody‐poor, intact red cells that retain antigenicity. Red cells prepared in this manner absorb antibody as efficiently as do uncoated red cells and nonspecific absorption does not occur. AbstractThe equivalent of 5–7 units of platelets, isolated from a single donor with the IBM Blood Cell Processor 2997 using a dual stage separation chamber, was frozen with the cryoprotectant dimethylsulfoxide (DMSO). The DMSO‐saline solution was added directly to the platelets, and the platelets were frozen in a polyvinyl chloride plastic bag by storage in a ‐80°C mechanical freezer. Washing the thawed platelets with a phosphate‐buffered sodium chloride‐dextrose solution, pH of 5.0, removed about 95% of the DMSO. In vitro freeze‐thaw‐wash recovery was 80%, and in vivo51Cr platelet recovery was 31%. AbstractAnti‐D levels in blood taken immediately postpartum from Rh‐negative women who have been given antenatal Rh immune globulin and the anti‐D levels in the cord blood of their babies were measured semiquantitatively by a low ionic polybrene Auto Analyzer technique. When both mother and baby were Rh‐negative a comparison of their anti‐D levels was made against the interval between injection of the passive anti‐D and delivery. Mothers' levels decreased as the interval increased, the regression coefficient for 73 maternal sera was ‐0.74. The drop in levels of cord blood was much less than that of the mothers' levels. The regression coefficient was ‐0.294 for 74 cord sera (one set of twins). The marked difference between cord blood and maternal blood levels of anti‐D at delivery supports the hypothesis that the transfer of IgG across the placenta is predominantly from mother to fetus and not fro

AbstractA serum sample from a nontransfused male containing anti‐Rh27 and an additional weakly reacting antibody was investigated. One absorption of the serum with RzRlcells left only anti‐Rh27 while repeated absorption with RlRlcells or autologous cells had no effect. Treatment of the serum with reducing agents destroyed all activity suggesting the antibody was predominantly IgM. Tests with monospecific antiglobulin reagents in the indirect antiglobulin test revealed very weak reactions with anti‐IgM and anti‐IgA, no reactions with anti‐IgG, and strong reactions with anti‐C3(C3c + C3d) and anti‐C4. AbstractThis report presents the first prospectively followed case of hemolytic disease of the newborn due to anti‐Jk3 alone. A Samoan woman with known anti‐Jk3 was closely monitored during her third pregnancy. Because of elevated ΔOD values in the amniotic fluid and rising titer of serum antibody, labor was induced at 36 weeks gestation. The cord red blood cells were coated with IgG, and anti‐Jk3 was eluted from the cord cells and the placenta. AbstractA survey of the occurrence of lympho‐agglutinins was carried out on seeds of 150 species of wild and cultivated plants. Potent agglutinins were detected in 17 extracts by employing lymphocytes of man, guinea pig, goat and sheep in the bioassay. Except for one species each of Mimosoideae, Euphorbiaceae and Clusiaceae, all the agglutinins were found in fabaceous seeds. Two of the agglutinins, namely,Parkia biglandulosaandErythrina arborescens, were found to be monospecific for guinea pig and human lymphocytes, respectively, while the others agglutinated lymphocytes of 2–4 different species.
