SummaryFor the preparation of larger amounts of human plasminogen one depends upon the availability of a suitable residue fraction from a plasma fractionation center. The method of isolation has to be adapted according to the composition of this starting material.A method has been worked out for the isolation of a highly enriched plasminogen preparation from the residual globulin fraction B as obtained in the fractionation of pooled plasma according toNitschmann, Kistler and Lergier[5, 81. The seven steps of the procedure are a succession of precipitations and extractions and no operation is involved which could not be easily carried out on a large scale. Conditions are such as to facilitate the reproducibility of the results.The effectiveness of the single steps in the purification process was determined mainly by means of the tosylarginine‐methylester (TAME) hydrolysis measured in a pH‐stat after activation of the preparations with streptokinase. The analytical method is described in detail. According to this method, the final preparations of plasminogen are about 30 times as active as the starting material (fraction B) and 100–110 times as active as pooled plasma when solutions of identical protein conrentration are compared. The purification factor appears much higher when based on measurements of the caseinolytic activity.The yield is about 20% of the activity in plasma and more than 30 % of the activity in the starting material.According to studies in the ultracentrifuge and with the immunoelectrophoresis, the final preparation is not a homogeneous protein.The plasminogen can be pasteurized in solution at pH 4 (10 hours at 60° C) with only little loss of activity. Activation can be achieved by streptokinase, by urokinase or spontaneously in 50% glycerol. The activated preparation shows a high fibrinolytic activity in vitro.RésuméLa préparation de quantiéks importantes de plasminogène humain dépend de l'existence d'une fraction riche en ce proferment provenant d'un centre de fractionnement du plasma. La méthode d'isolation doit être adaptée à la composition de cette fraction initiale.Les auteurs ont mis au point une méthode permettant d'obtenir une priparation très riche en plasminogène à partir du résidu de la fraction globulinique B; cette fraction provient du fractionuement d'un «pool» de plasmas selonNitschmann, Kistler et Lergier[5, 81. La méthode comprend sept étapes, consistant en une succession de précipitations et d'extractions; elle ne comporte aucune opération pi ne pourrait être facilement effectuée sur une large échelle; les conditions sont prévues pour faciliter la reproductibilité des résultats.L'efficacité de chaque étape du procédé de purification a été déterminée principalement par la méthode de l'hydrolyse du tosylarginineméthylester (TAME) mesurée dans un pH‐stat après activation des préparations par la streptokinase. La méthode analytique est décrite en détail. Par cette mithode les préparations finales de plasminogène sont à peu près 30 fois plus actives que la fraction initiale (fraction B) et 100 à 110 fois plus actives que le «pool» plasmatique, quand on compare des solutions de teneur protidique identique. Le facteur de purification parait beaucoup plus élevé quand il est basé sur la mesure de l'activité caséinolytique.Le rendement est environ de 20% de l'activité du plasma et de plus de 30% de l'activité de la fraction initiale.L'étude de la préparation finale en ultracentrifugation et en immuno‐électrophoèse montre qu'il ne s'agit pas d'une protéine homogéne. Le plasminogène peut être pasteurisé en solution à pH 4 (10 h à 60°C) sans qu'il résulte une perte d'activité appriéiable. La préparation activée est douée d'une forte activité fibrinolytique in vitro.ZusummenfassungZur Gewinnung größnerer Mengen von humanem Plasminogen ist man auf eine Restfraktion aus einem Plasmafraktionierungszentrum angewiesen, in der sich dieses Proferment angereichert findet. Die Isolierungsmethode wird sich nach der Zusammensetzung des Ausgangsmaterials zu richten haben.Es wurde eine Methode ausgearbeitet fůr die Gewinnung eines hochangersicherten Plasminogenpräparates aus der Restglobulinfraktion (B), die bei der Fraktionierung von Mischplasma nachNitschmann, Kistler und Lergier[5, 8] anfällt. Das ***7stufige Verfahren wird genau beschrieben. Die Bedingungen sind so präzisiert, daß die Ergebnisse gut reproduzierbar sind. Es werden nur Fällungen und Extraktionen ausgeführt, so daß sich das Verfahren ohne weiteres in praktisch beliebig großen Ansätzen durchführen Iäßt. Der Erfolg der Anreicherung wurde vor allem durch Messung der TAME‐Spaltung im pH‐Stat nach Streptokinaseaktivierung kontrolliert. Die Analysemethode wird näher beschrieben. Die Schlußpräparate zeigen nach dieser Meßmethode einen Anreicherungsfaktor von etwa 30 gegenüber dem Ausgangsmaterial und von 100–110 gegenüber Mischplasma. Die Anreicherungsfaktoren, die durch Caseinolysemessungen angezeigt werden, sind viel höher.Die Ausbeute beträgt über 300%, bezogen auf die Ausgangsfraktion und etwa 20 % bezogen auf Plasma.Ein einheitliches Protein liegt nach Untersuchungen in der Ultrazentrifuge und mit der Immunoelektrophorese nicht vor. Unser Plasminogenpräparat läßt sich bei pH 4 ohne wesentlichen Verlust pasteurisieren (10 Stunden bei 60° C).Die Aktivierung kann in bekannter Weise durch Streptokinase, Urokinase oder spontan (in 5

SummaryA new method of experimental erythrophagocytosis is proposed whereby the interaction of antigen, antibody and complement can be studied. The effect of antibody, temperature, chelating agents, incubation time, volume, reagent concentration and complement components is discussed. The enhancing effect of complement on phagocytosis is demonstrated. It appears thatC'1,C'4andC'2are the only complement components involved in erythrophagocytosis. C'3is not necessary.RésuméUne nouvelle méthode d'érythrophagocytose expérimentale est décrite où l'action entre l'antigéne:, l'anticorps et le complément peut être étudiée. SummaryThiols and other reducing agents, such as sodium borohydride, dissociate 19 S isoagglutinins into 6 S subunits with concomitant loss af agglutinating activity. Using thiols little or no activity was recovered upon reoxidation, whereas the effert of borohydride was reversible; in some instances up to 100% of the original activity was recoverd upon dialysis in the presence of oxygen.The dissociation produrts were alkylated with iodoacetamid to block the sulf‐hydryl groups and prevent reassociation.In a new serie of experiments amixtureof isoagglutinins anti‐A and anti‐B was treated with borohydride and reaggregated by dialysis. It was possible to recover a fraction of antibodies with double specificity, bearing on the same molecule anti‐A as well as anti‐B activity. These “mixed” antibodies were brought into evidence by absorption and elution methods and also by mixed agglutination between type A buccal epithelial cells and type B erythrocytes.ZusammenfassungAnti‐A und Anti‐B Isoagglutinine des 19 S‐Typs können durch Disulfid‐spaltende Reduktionsmittel wie Merkaptane oder Borhydrid in inaktive Untereinheiten zerlegt werden. Während die Aktivität nach Behandlung mit Merkaptanen nicht mehr nachgewiesen werden konnte, stieg sie bei den mit Borhydrid behandelten Proben nach oxydativer Dialyse an, in einigen Fällen bis zum Ausgangswert.Die Spaltprodukte, deran Sulfhydrylgrnppen zur Verhinderung der Reaggregierung mit Monoiodazetamid blockiert wurden, agglutinieren nicht mehr, sind aber befähigt, die Agglutination durch native Isoagglutinine zu hemmen.Durch Behandlung eines Gemisehes von Anti‐A und Anti‐B‐Isoagglutininen mit Borhydrid und anschlieβlender Dialyse in Gegenwart von Sauerstoff gelang die Her‐stellung von hntikörpern doppelter Spezifität. Letztere wiesen auf demselben Molekül sowohl Anti‐A als auch Anti‐B‐Aktivität auf. Sie konnten einerseits mit Absorptions‐ und Elutionsmethoden, andererseits durch gemischte Agglutination von Epithelzellen der Blutgruppe A und Typ B‐Erythrozyten nachgewiesen werden.RésuméL'emploi de réducteurs des ponts disulfures tels que les thiols ou le borohydrure de sodium ont permis de scinder les isoagglutinines anti‐A et anti‐B en subunités avec perte du pouvoir agglutinant. Avec les thiols l'activité n'a pas pu être récupérée de façon satisfaisante, tandis qu'avec le borohydrure le titre remonte après dialyse oxydative jusqu'à atteindre parfois sa valeur initiale.Les produits de scission que l'on acyle au moyen de monoiodoacétamide pour bloquer les groupes sufhydryles et empêcher ainsi une réassociation ne sont plus agglutinants, mais sont inhibiteurs.Dans une seconde série d'expériences, en traitant au borohydrure un mélange d'agglutinines anti‐A et anti‐B et en laissant les suhunités se réaggréger par dialyse il a été possible d'obtenir une certaine proportion d'anticorps à double spécificité, c'est‐à‐dire portant sur la même molécule à la fois un site anti‐A et un site anti‐B. SummaryA case is described of inherited positive direct Coombs' reaction combined with a weakly developed N factor (N2) in a mother and her baby. By way of explanation of the phenomenon the hypothesis is put forward that the weak N factor is due to the fact that the N antigen (N2) in this case possesses few determinant groups and that the carrier group of proteins is therefore exposed and able to react with the Coombs' serum.RésuméOn décrit un cas de réaction directe de Coombs positive congénitale avec un facteur N (N2) faiblement développé chez une mère et son enfant. Pour expliquer ce phénomène, on avance l'hypothèse que le facteur N faible est dû au fait que, dans ce cas, l'antigène N (N2) ne possède que peu de groupes déterminants et que les protéines porteuses de groupe sont de ce fait exposées et réagissent sans autre avec le sérum antiglobuline.ZusammenfassungEs wird ein Fall von vererbtem positivem direktem Coombstest in Kombination mit einer schwachen N‐Eigenschaft (N2) bei einer Mutter und ihrem Kind beschrieben. SummarySera from normal adults of the groups A, B and AB were studied for group specific blood group substance. This was found to be present in the vast majority of cases. After division of the groups A and AB into A subgroups, non‐secretors showed significantly lower values than secretors within each A subgroup. When secretors and non‐secretors were considered separately, both categories revealed significantly lower values for group A, than for group A,. No difference in the content of B substance could be demonstrated in secretors and non‐secretors of groups B and AB.The inhibition which is produced by the serum is considered to be group specific, since O sera which were absorbed free of iso‐antibody did not inhibit anti‐A or anti‐B.A study of sera from a series of twins did not reveal any evidence in support of the assumption that the amount of blood group substance should be determined by genetic factors.RésuméLes sérurns d'adultes normaiix de groupe A, B et AB ont été étudiés quant à leur substance spécifique de groupe sanguin. Celle‐ci s'est avérée être présente dam la grande majorité des cas. Après avoir séparé les groupes A et AB en sous‐groupes de A, les non‐secréteurs présentaient des valeurs nettement plus basses que les secréteurs daus chacun des sous‐groupes de A. Si les secréteurs et les non‐secréteurs étaient examinés séparément, les deux catégories présentaient des valeurs plus basses pour A2que pour A1. On n'a pas pu mettre en évidence de différence dans la contenance en substance B chez les secréteurs et non‐secékteurs des groupes B et AB.L'inhibition qui est produite par le sérum est considérée comme étant spécifique de groupe, puisque les sérums O qui ont été absorbés et qui sont francs d'iso‐anti‐corps n'inhibent pas l'anti‐A et l'anti‐B.Une étude de sérum de jumeaux ne soutient pas d'une manière évidente l'hypothèse selon laquelle la quantité de substance de groupe sanguin peut être déterminée par un facteur génétique.ZusammmenfassungSeren von normalen Erwachsenen der Gruppen A, B und AB wurden auf ihren Gehalt an Blutgruppensubstanz untersucht. In der überwiegenden Mehrzahl der Seren wurden solche Substanzen festgestellt. Nach Unterteilung der Gruppen A und AB in die entsprechenden A‐Untergruppen zeigte sich, daß innerhalb der A‐Untergruppen Seren von Nicht‐Sekretoren einen signifikant niedrigen Gehalt an Gruppensubstanz enthalten als Seren von Sekretoren. Bei gesonderter Betrachtung der Sekretorenseren und der Nicht‐Sekretorenseren zeigte sich, daß in beiden Fällen der Gruppensubstanzgehalt bei der Gruppe A2signifikant niedriger ist als bei der Gruppe Al. Bei den Seren von Sekretoren und Nicht‐Selcretoren der Gruppen B und AB wurde kein Unterschied im B‐Substanzgehalt festgestellt.Die durch diese Seren bewirkten Agglutinationshemmungen sind als spezifisch zu betrachten, da bei O‐Seren, bei denen die Isoantikörper wegabsorbiert worden waren, keine Hemmwirkung auf Anti‐A bzw. RésuméLa recherche des antigènes Gm dans les taches de sang sec présente un intérêt consiéirable en médicine légale. En effet, les résultats obtenus sont particulièrement nets et la recherche de ces antigènes demande une quantité de tache bien inférieure à celle que l'on doit employer pour la détection de ceux des autres systèmes.Ces antigènes peuvent être mis en évidence non seulement dans les taches récentes, mais aussi, quoique de façon moins facile, dans des taches anciennes où ils semblent se conserver au moins pendant une dizaine d'années.Leur recherche doit cependant se faire avec des sérums bien choisis et de titre soigneusement déterminé, inférieur à celui qui est préconisé habituellement pour la mise en évidence du facteur Gm dans les sérums. L'origine des sérums anti‐Gm utilisés paraît jouer un rôle essentiel: chaque fois que cela est possible, il faut utiliser des sérums anti‐Gm provenant de sujets normaux (Snagg) et non des anticorps provenant de malades atteints de polyarthrite chronique rhumatismale (Ragg). Cette condition est particuliàrement rigoureuse quand il s'agit de grouper des taches anciennes.SummaryThe determination of the Gm antigens in dried blood stains is of considerable importance in medico‐legal work, since the results obtained are particularly clear‐cut and a much smaller amount of stain is required than for determining other antigens.The Gm antigens can be demonstrated in fresh stains and also, although with less ease, in old stains; they appear to retain their activity for at least ten years.The antisera to be used must be carefully selected, their titre must be exactly determined and it should be lower than that of the antisera usually recommended for Gm determinations in serum. It appears that one of the conditions for obtaining good results, especially when working on very old stains, is to use anti‐Gm sera from normal persons (Snagg), but not sera from patients suffering from rheumatoid arthritis (Ragg).ZusammenfassungDer Nachweis der Gm‐Antigene in trockenenBlutflecken begegnet in der Gerichtsmedizin einem erheblichen Interesse. Tatsächlich sincl die Ergebnisse meist eindeutig; zudem benötigt man wesentlich weniger Material als bei der Bestimmung der Antigene anderer Systeme.Dime Antigene lassen sich nicht nur in frischen, sondern anch, wenn auch weniger leicht, in alten Flecken nachweisen. Ihre Konservierbarkeit beträgt mindestens 10 Jahre.Die Bestimmung hat mit wohlausgewahlten Seren mit genau kontrolliertem Titer zu erfolgen, wobei der letztere niedriger sein muß als es zur Gm‐Bestimmung in Seren gewöhnlich vorgescbrieben wird. Die Herkunft der Seren spielt eine große Rolle. Soweit immer möglich ist den Anti‐Gm‐Seren von normalen Individuen (Snagg) gegenüber denjenigen von Patienten mit primär chronischer Polyarthritis (Ragg) der Vorzug zu geben. Diese Voraussetzun

ZusammenfassungEs wird über die Haüufigkeit erblicher Serumeigenschaften und der ihnen zugrundeliegenden Gene in Island berichtet. Folgende Gen‐Frequenzen konnten beobachtet werden: Hp1= 0,3856; Gma= 0.3934; Gmx= 0,1981; Gc1= 0,759. Diese Werte liegen im Bereich der bisher für europäische Populationen mitgeteilten Gen‐Frequenzen.SummaryA study was made of the frequencies of hereditary serum factors and of the genes governing them in Iceland. The following gene frequencies were observed: Hp1= 0.3856; Gma= 0.3934; Gmx= 0.1981; Gc1= 0.759. Summary1A method is described for the isolation and purification of urokinase from human urine. The procedure includes: adsorption on and elution from the cation exchanger, cellulose phosphate; precipitation with 20% ethanol; adsorption on and elution from ion exchange resin XE‐97; dialysis to remove salts; and drying from the frozen state. Sterilization is accomplished by treating with betapropiolactone (1:6000 to 1:8000).2When necessary, pyrogenicity may be removed by incubation of urokinase in 0.15 M NaCl solution at pH 7.2–7.4 for four days at 37° C.3The resulting urokinase passes standard tests for sterility, safety, and pyrogenicity. It has a potency of 1000 activator units per mg protein and is stable under normal storage conditions.4A method is described forin vitrourokinase activation of human profibrinolysin in 50% glycerol.5A simple, reliable and economical assay system is described for urokinase activity.Résumé11° Une méthode pour isoler et purifier l'urokinase à partir de l'urine humaine est décrite.
