SummaryExamples of anti‐A were obtained from various subjects: random donors; selected group O and B subjects, before and after the injection of A substance; the mother of an infant severely affected with ABO‐haemolytic disease; and normal newborn infants.Approximately 90% of the random group O sera would haemolyse group A red cells when the ratio of serum: cells was 40:1, and all group O sera tested were haemolytic when the ratio was 200: 1. Approximately 30% of group O sera failed to agglutinate pig group A red cells and these sera were mainly those with a low haemolysin titre when tested against human group A red cells.Selected sera were fractionated on a DEAE‐cellulose column. In many cases elution was carried out only with 2 phosphate buffers, 0.02 M to obtain a fraction containing 7Sγ globulin and 0.2 M to obtain a fraction containing 19Sγ globulin, but in some cases intermediate fractions were obtained. The concentration of 7Sγ and 19Sγ in all fractions was estimated by an immunological method. It was confirmed that anti‐A in group B subjects, before and after stimulation with A substance is predominantly 19Sγ globulin whereas in group O subjects in addition to 19Sγ anti‐A there is often some 7Sγ anti‐A before stimulation and usually considerable 75γ anti‐A after stimulation. The finding of Rockey and Kunkel [17] that considerable amounts of anti‐A may be present in “intermediate” fractions obtained from a DEAE‐cellulose column, that is fractions containing only low concentrations of 7Sγ and 19Sγ globulin, was confirmed.Various serological characteristics of these different fractions were studied: (1) Anti‐A in the 0.2 M fraction (19Sγ) was far more easily neutralized by A substance than the anti‐A in the 0.02 M fraction (7Sγ). (2) Whereas the agglutination produced by the anti‐A in the 0.02 M fraction was greatly enhanced in a medium of serum, compared with saline, agglutination produced by anti‐A in the 0.2 M fraction was not enhanced by serum. (3) Only the anti‐A in the 0.2 M fraction was found to agglutinate Apcells although the anti‐A in the 0.02 M fraction would sensitize Apcells to agglutination by an anti‐7Sγ globulin serum. (4) Anti‐A in the 0.2 M fraction bound more complement than antibody of equivalent titre in the 0.02 M fraction.In all these tests, antibody eluted in intermediate fractions and thought to correspond with the 11‐12S antibody described by Rockey and Kunkel 117] behaved in the same way as 19Sγ anti‐A recovered in the 0.2 M fraction.It is clear that there is no single criterion of “immune” anti‐A; an ability to hacmolyse group A cells readily is possessed by most 19Sγ anti‐A and all 7Sγ anti‐A, whereas the ability of a serum, after partial neutralization, to sensitize A cells to an antiglobulin serum is almost confined to those containing 7Sγ anti‐A.RésuméDes anti‐A ont été obtenus chez différents sujets: des donneurs pris au hasard; des sujets de groupe B ou O avant et après l'injection de substance A; de mères d'enfant atteints gravement de maladies hémolytiques ABO; de nouveaux‐nés normaux.A peu près 90% des sérums de groupe O testés hémolyse les érythrocytes de groupe A lorsque le rapport sérum/érythrocytes est de 40:1 et tous les sérums O testés sont hémolytiques quand le rapport est de 200:1. Diese stammten von Blutspendern, ausgewählten O‐ und B‐Individuen, vor und nach Injektion von A‐Substanz, von normalen Neugeborenen sowie von der Mutter eines Kindes mit schwerem ABO‐Morbus hämolyticus.Ungefähr 90% der O‐Seren hämolysierten A‐Zellen, sofern das Serum‐Zellver‐hältnis 40:1 betrug; bei einem Verhältnis von 200:1 erwiesen sich alle O‐Seren als hämolytisch aktiv. Ca. 30% der O‐Seren vermochten Schweineerythrozyten der Gruppe A nicht zu agglutinieren. Es handelte sich dabei in der überwiegenden Mehrzahl um jene Seren, die gegenüber menschlichen A‐Zellen einen niederen Hämolysintieter aufwiesen.Einige ausgrwählte Seren wurden auf DEAE‐Zellulosekolonnen fraktioniert. In den meisten Fällen wurde die Elution nur mit zwei Phosphatpuffern vorgenommen. Mit dem 0,02 M‐Puffer wurde die Fraktion, welche das 75‐γ‐globulin enthielt, eluiert. Zur Elution der Fraktion, in der das 19S‐γ‐Globulin enthalten war, wurde der 0,2 M‐Puffer verwendet. In einigen Fällen wurden Zwischenfraktionen gewonnen. Der Gehalt der Fraktionen an 7S‐γ‐ und 19S‐γ‐Globulin wurde immunochemisch überprüft. Es bestätigte sich, daβ die Anti‐A‐Antikörper bei B‐Individuen vor und nach der Stimulation mit A‐Substanz vornehmlich den Charakter von 19S‐γ‐Globulinen aufweisen; bei O‐Individuen findet man vor der Stimulation nebst 19S‐Antikörpern auch eine gewisse Menge von 7‐S‐Antikörpern. Nach der Stimulation mit A‐Substanz steigt bei O‐Individuen der Gehalt an 7‐S‐Antikörpern meist stark an. Der Befund von Rockey und Kunkel [17], die beträchtliche Mengen von Anti‐A in Zwischenfraktionen mit geringem 7S‐γ‐ und 19S‐γ‐Globulingehalt gefunden hatten, wurde bestätigt.Die serologischen Eigenschaften dieser Fraktionen wurden untersucht. Dabei ‐Fraktion blieb dieser Effekt aus. 3. Lediglich die in der 0,2 M‐Fraktion ent‐haltenen Anti‐A‐Antikörper vermochten Ap‐Zellen zu agglutinieren. Die Antikörper der 0,02 M‐Fraktion sensibilisierten Ap‐Zellen, so daβ diese durch Anti‐7S‐γ‐Globulinseren agglutiniert wurden. 4. Die Anti‐A‐Antikörper der 0,2 M‐Fraktion vermochten mehr Komplement zu binden, als titergleiche Antikörper der 0,02 M‐Fraktion.In all diesen Versuchen verhielten sich die in den Zwischenfraktionen enthaltenen Antikörper (11‐12S‐Antikorper van Rockey und Kunkel [17]) wie die in der 0,2 M‐Fraktion enthaltenen 19S‐Antikörper.Es ist klar, daβ es kein verläβliches Einzelkriterium für den «Immuncharakter» van Anti‐A‐Antikörpern gibt. SummaryIn accord with published reports it was found that the RNA derivatives cytidine sulfate and eytidylic acid inhibit Rh agglutinations, but only weakly. A number of other compounds structurally related by virtue of containing rigid ring systems and phosphate ester groups were more potent inhibitors. Included in this group are phenolphthalein diphosphate, α‐naphthyl phosphate, riboflavin‐5′‐phosphate, and α‐tocopherol phosphate, the latter giving the strongest inhibition at the lowest concentration. That the inhibition is probably not specific is indicated by the lack of effect on Coombs reactions and by the inhibition of blood group systems unrelated to Rh. Treatment of red cells with acid and alkaline phosphatase, ribo‐nuclease, and the phosphodiesterase of Russell's viper venom failed to produce a consistently significant effect on Rh agglutination reactions.RésuméEn accord avec les rapports publiés, lee auteurs constatent que des dérivés de l'ARN, le sulfate de cytidine et l'acide cytidylique, inhibent l'agglutination Rhésus, mais seulement faiblement. Un nombre d'autres composants de structure apparen‐the par le fait qu'ils contiennent des systèmes à anneaux fixes et des groupes phos‐phoriques estérifiés sont des inhibiteurs plus puissants. Font partie de ce groupe le diphosphate de phenolphthaléine, l'α‐naphtyl‐phosphate, le riboflavine‐5′‐phosphate et l'α‐tocopherol phosphate, ce dernier donnant l'inhibition la plus forte à la concentration la plus basse. Cette inhibition n'est probablement pas spécifique étant donné l'absence de ralentissement sur la réaction de Coombs et l'inhibition d'autres systèmes de groupe sans rapport avec le système Rhésus. Le traitement des érythrocytes avec les phosphatases alcalines et acides, les ribonucléases et la phospho‐diesterase du venin de vipère Russell ne produisent pas un effet significatif sur les réactions d'agglutinations Rhésus.ZusammenfassungIn Übereinstimmung mit publizierten Berichten wurde festgestellt, daβ die RNS‐Derivate Cytidinsulfat und Cytidylsäure Rh‐Agglutinationen zu hemmen vermögen. Der Effekt ist aber wenig ausgeprägt. Eine Reihe strukturell verwandter Verbindungen, die starre Ringsysteme und Phosphatestergruppen enthielten, zeigten stärkere Hemmwirkungen. Zu dieser Gruppe gehören Phenolphthaleindiphosphat, α‐Naphtylphosphat, Riboflavin‐5‐phosphat und α‐Tocopherolphosphat, wobei die letztgenannte Verbindung die stärkste Hemmwirkung aufweist. Die Hemmung ist wahrscheinlich unspezifisch. SummaryA man of genotypeRu1Ryis reported who, on transfusion, developed anti‐cand anti‐D and became direct Coombs positive. Study of his direct Coombs negative sister of the same phenotype established the genotype of both, which was refined toR1abcd.Ry.The sister's cells reacted with 31 of 45 random anti‐D sera, reaction being more frequent with high titer incomplete antibodies than with high titer sera with a saline component. They reacted with fewer sera and reacted less strongly than didR1cdWiel.r orRoac(d)Wiel.rcells. Anti‐D was eluted from the man's cells, while his sister's cells took up anti‐D which could then be eluted from them. Summary1. With the use of a fully automated continuous flow system, routine blood typing may be accomplished with the same serological accuracy as with the manual method. ABO cell typing, serum and Rh typing, including Du, are feasible. Furthermore, there is an objective permanent record of the reactions.2. Reagent requirements are generally those of the manual method; total cost would appear to be no greater.3. The capacity of the system is in the order of 40 complete typings per hour; between 300–600 complete typings per day appear possible.4. The influence of several serological variables of importance in manual blood typing have been shown to be important in the automated system; these include time, temperature, saline concentration, enzyme treatment of cells and macromolecular suspending media.5. The application of the system to a routine laboratory operation awaits the development of a multiple channel machine which can integrate all the results the several tests being performed upon a single sample of blood.Résumé1. En utilisant un système continu automatique, on peut déterminer les groupes sanguins habituels avec le měme degré de sécurité que celui obtenu par la méthode manuelle. La détermination des groupes ABO, Rhésus, y compris Duest praticable. En outre, il y a un enregistrement objectif permanent des réactions.2. Les réactifs requis sont en général identiques à ceux utilisés pour la méthode manuelle; le coǔt total ne semble pas ětre plus grand.3. La capacité du système est de l'ordre de 40 déterminations à l'heure; 300 à 600 déterminations complètes semblent possible en une journée.4. Im ABO‐System ist die Bestimmung der Zellantigene wie der Serumagglutinine möglich; im Rh‐System gelingt es, Du‐Blutproben zu erfassen. Die Methode liefert darüber hinaus einen objektiven Beleg des Reaktionsausfalles.2. Der Reagentienbedarf liegt in derselben Gröβenordnung wie bei der manuellen Methode. Die Gesamtkosten dürften daher ebenfalls in derselben Gröβe liegen.3. Die Leistungsfähigkeit des Systems liegt bei 40 kompletten Bestimmungen in der Stunde. Eine Tagesleistung von 300–600 Bestimmungen scheint durchaus möglich.4.Eine Reihe von Reaktionsbedingungen, die bei der manuellen Methode wichtig sind, müssen auch bei diesem System beachtet werden; es handelt sich um die Zeit, die Temperatur, die Kochsalzkonzentration, die Enzymbehandlung der Zellen und die Art der makromolekularen Aufschwemmungsflüssigkeit.5.
