Abstract.Graft‐versus‐host disease (GVDH) follows blood product transfusions in patients with deficient cell‐mediated immunity. The rate of mortality in this adverse effect of blood transfusions is between 90 and 100%. The historical; clinical and pathologic; and mechanism of acute and chronic GVHD are presented. The patients at risk, the clinical and pathologic features of posttransfusion GVDH have been presented in hopes that, with increased awareness of this usually fatal complication, clinicians and transfusion therapists may more intelligently advise and select patients who may need lymphocyte‐poor or irradiated blood products and may recognize the syndrome

Abstract.One B‐ and two A‐specific radioimmunoassays detected A and B antigenic determinants in saliva of ABH nonsecretors. Abstract.The distribution of antigenic determinants on human IgA was studied with fragments and mutants of IgA1. F(abc)2 and Fabc, lacking the CH3 domain, F(ab′)2 and Fab′, lacking the CH2 and CH3 domain, Fab that further lacks most of the hinge region, and Fc fragments were included in our investigations. Antibodies specific for the CH3 domain of IgA1 were found in antisera raised against an α1‐HCD protein and in anti‐Fcα antisera. The antisera detected different antigenic determinants on CH3 as was shown by inhibition with sera from nonhuman primates. An anti‐Fc5μ antiserum detected a determinant on Fc common to IgA and IgM. The serum of an IgA‐deficient individual reacted with a determinant on CH2 of four‐chain molecules only. A subclass‐specific anti‐Fabc antiserum detected a determinant which needed interaction of CH2 and CH1 or the hinge region. Abstract.To determine the degree of platelet damage produced by different modes of agitation during storage of concentrates for 5 days in CLX blood bags, we studied pH, platelet counts, release of LDH and beta thromboglobulin, morphology and osmotic recovery. Platelets were maintained at 20–24°C on elliptical, 6‐rpm circular, 2‐rpm circular and flat bed agitators. At 72–120 h platelet concentrates stored on the flat bed shaker had significantly lower pH values than units stored on the elliptical or on either of the circular rotators (p<0.05). The percent LDH discharged was highest for the units stored on the elliptical rotator (p<0.05). Remaining tests of platelet function were not significantly different for concentrates stored on any of the four agitators. Flat bed shakers were unable to resuspend the platelet ‘button’ which formed after the final preparative centrifugation. Abstract.The study of the specificity of platelet autoantibodies has made it possible to expand our knowledge about the immune characterization of idiopathic thrombocytopenic purpura. Abstract.A solid‐phase kinetic enzyme‐linked immunosorbent assay has been developed to measure binding of antibodies to purified or synthetic blood group antigens and tested in the Lewis blood group system. Chemically synthesized Lewisaantigen is used as the target for the binding of serum antibody in this sandwich‐type assay. Kinetic, rather than endpoint, determinations are used to calculate the amount of specific antibody. Data are presented showing the assay to be quantitative, sensitive, and specific. It can separately quantitate the amount of IgG or IgM anti‐Lewisapresent in patient sera. The assay uses commercially available reagents and is semiautomated.
