SummaryTwo genetic determinants of human γ‐globulin, Gm(s) and Gm(t), were identified.RésuméDeux »nouveaux« déterminants génétiques des gamma‐globulines humaines ont été identifiés; ce sont les déterminants Gra(s) et Gm(t).1Type Gm(s +) occurred in 38% of 176 Japanese, 31% of 75 Eskimoes of Western Greenland, 21% of 188 Suriname Negroes, 14% of 110 West African Negroes, and only in about 1% of 910 Europeans.2All Gm (s+) sera were Gm(b3+); and all Gm(b1‐b3+) sera were Gm(s +). This suggests an allele at theVilocus, calledGmsbor Gmsb(3), that is partly identical with and partly different fromGmb(=Gmb(1,3)), — the most commonViallele in Caucasians.3Gm(s) appeared to be inherited together with the determinants Gm(a) and (b3) in the Mongoloid populations and in 4 of the 6 European families studied. This suggests thatGmsbis as a rule coupled with aGmagene at theWelocus in Mongoloids and often (but not always) coupled withGmaalso in Caucasians.Gmb, by contrast, appears to be coupled withGmfor withGmafin Caucasians and Mongoloids.4Gm(t) was found together with Gm(s) in Japanese, but not in Negroes. The Gm specificities are ascribed to 3 separate genzetic regions: one region that determines Gm(a); a second that determines Gm(f); and a third that determines the Gm(b) specificities and probably Gm(c), (s), and (t). SummaryResults of Lp tests on unrelated individuals and members of families are presented.The Lp types are independent of sex and age. The calculations of the Lp frequencies in the Norwegian population are now based on tests of 1109 unrelated individuals. A frequency of 0.1948 was found for the geneLpa.Members of 175 Norwegian families with a total of 646 children have been tested for the Lp(a) factor (94 of the families have been included in an earlier paper). The distribution of the Lp types in the family material confirms that the Lp(a) factor is determined by a simple, autosomal, dominant mode of inheritance. No exception from the postulated mode of inheritance, in addition to the one reported earlier, was found.The results of the population studies so far performed, are presented.RésuméLes résultats concernant le système Lp chez des individus non apparentés et chez des membres de même famille sont présentés.Les types Lp sont indépendants du sexe et de l'âge. SummaryPrecipitating anti‐β‐lipoprotein isoantibodies were found in the sera from six patients out of 80 multiply transfused thalassemic children. Very weak isoprecipitins could be observed in four other cases.Thirteen sera from non‐transfused thalassemic patients were tested as well, but with negative results.The antisera reveal apparently five different specificities within the β‐lipoproteins of human serum.Splenectomy was found to play a significant role in the development and concentration of anti‐β‐lipoprotein antibodies.RésuméDes iso‐anticorps anti‐β‐lipoprotéines furent trouvés dans des sérums provenant de six patients compris dans un lot de 80 enfants thalassémiques ayant reçu plusieurs transfusions. On a retrouvé également une très faible isoprécipitine dans 4 autres cas.Treize sérums de patients thalassémiques non transfusés ont aussi été testés; mais il n'y eut aucun résultat positif. SummaryInformation about association of two attributes with frequencies p1and p2in a sample of size n is measured by p1(1 ‐ P1)p2(1‐p2)n. Examination of the 55 associations within multiple allelic systems for factors studied in northeastern Brazil shows that while many of them are significant in small samples, many others could be detected only in extremely large samples. The low power of tests of association is discussed and illustrated, and the conclusion is reached that only linkage tests provide powerful and unambiguous corroboration that a factor does, or does not, belong to a given system.The number of significant associations between systems is consistent in the parental sample with the nominal significance level of the test, showing that racial stratification, correlation between relatives, phenotypic interaction, multispecific reagents, and other disturbing factors are negligible in this material. However, the frequency of “significant” associations in children is five times the nominal type I error of the test, illustrating the disturbing effects of nonbinomial variation in samples of close relatives.The terminology and logical relations among factor pairs are discussed, and a system is defined as the unit of closely linked genetic information whose phenotypic factors are nonrandomly associated in panmictic populations of higher organisms.RésuméLa recherche de 55 associations entre systèmes d'allèles multiples étudiés dans le Nord‐Est du Brésil montre que si plusieurs d'entre elles sont décelables dans de petits échantillons avec une fréquence significative de nombreuses autres ne sont décelables que dans de grands échantillons de populations. Les auteurs discutent et soulignent la faible puissance de tests d'association. Ils concluent que les tests de linkage fournissent des réponses sans ambiguïté sur l'appartenance d'un facteur à un système ou son indépendance.Le nombre d'associations significatives entre systèmes est en rapport dans l'échantillon parental avec le taux de significance nominale du test, montrant que la stratification raciale la corrélation entre parents, l'interaction phénotypique, la multispécificité des réactifs et d'autres facteurs de perturbation peuvent être négligés dans ce matériel. Solche Tests haben eine Information von P1(1 ‐ p1) p2(1 ‐ p2) n, wobei p1und p2die Häufigkeiten der beiden Phänotypen sind und n die Zahl der getesteten Individuen darstellt. Eine Prüfung der 55 Paarbeziehungen innerhalb der Systeme mit multipler Allelie zeigt, daß eine signifikante Abhängigkeit bei vielen Tests erst bei extrem großen Stichproben nachweisbar wäre, während andere schon bei kleinen Stichproben signifikant sind. Die geringe Stärke des Assoziationstests wird diskutiert und illustriert und führt zum Schluß, daß nur Koppelungstests eine kräftige und eindeutige Bestätigung dafür liefern, ob ein Faktor zu einem gegebenen System gehört oder nicht.Die Zahl signifikanter Beziehungen zwischen verschiedenen Systemen steht bei der Elternpopulation mit dem durch die Wahl der Signifikanzgrenze zu erwartenden Wert in Einklang. Dies zeigt, daß Rassenschichtung, Korrelation zwischen den Verwandten, phänotypische Beeinflussung, multispezifische Reagenzien und andere Störfaktoren in diesem Material zu vernachlässigen sind. Bei den Kindern erreicht jedoch die Häufigkeit der »signifikanten« Kombinationen den 5fachen Betrag des erlaubten Wertes. SummaryData from 207 two‐generation pedigrees have been analysed by computer to assess their contribution to the probability of linkage between each of 66 pairs of autosomal loci of the common marker type. SummaryThe present report details the relationship of mother‐child ABO and Rh incompatibilities in a series of 520 Rh immunised mothers investigated throughout pregnancy. These mothers were divided into two series, those possessing Rh antibodies prior to the present pregnancy and those who were not immunised before but developed Rh antibodies at some stage during the present pregnancy. In this way it was possible to measure the frequency of ABO incompatible conceptions associated with Rh immunisation.The observed findings showed no significant variations in the distribution of ABO incompatibility or in the mother's ability to produce Rh antibodies. Of importance remains the significantly lower Rh antibody titre values observed among ABO incompatible conceptions as opposed to the ABO compatible mother‐child combinations.The quantitative disparity observed in the production of Rh antibodies induced by ABO and Rh incompatible foetal red cells disclosed further evidence to suggest that variations may exist in the maternal mechanism of antibody synthesis.RésuméCe présent rapport expose les relations qui existent dans les incompatibilités fœtomaternelles des systèmes ABO et Rhésus trouvées lors d'une étude faite auprès de 520 mères enceintes présentant une immunisation Rhésus.Ces mères furent divisées en deux groupes: d'une part celles qui furent précédemment immunisées et qui possèdent encore dans leur sérum un anticorps anti‐Rhésus; d'autre part, celles qui n'ont présenté aucune immunisation avant la grossesse où s'est déclarée une immunisation. Ainsi, il fut possible d'étudier les incompatibilités fœto‐maternelles du système ABO liées à une immunisation Rhésus.Les résultats ne montrent pas de variations significatives entre la distribution de l'incompatibilité ABO et l'aptitude de la mère à former des anticorps anti‐Rhésus. Mais, il est important de constater qu'on peut trouver un titre d'anticorps anti‐Rhésus significativement plus faible lors d'incompatibilité ABO tandis que ce n'est pas le cas lors de combinaisons fœtomaternelles ABO compatibles.La différence quantitative observée dans la production des anticorps Rhésus induits par des globules rouges fœtaux incompatibles pour le système ABO et Rhésus laisse supposer qu'il y a des variations du mécanisme de la synthèse des anticorps dans le rapport mère‐enfant.ZusammenfassungIm vorliegenden Bericht werden die Beziehungen zwischen ABO‐ und Rh‐Inkompatibilität bei Mutter und Kind an Hand einer Reihe von 520 Rh‐immunisierter Mütter, die während der ganzen Schwangerschaftsdauer unter serologischer Kontrolle standen, im einzelnen dargelegt. Die Mütter wurden in zwei Gruppen unterteilt, nämlich eine solche, bei welcher die Rh‐Antikörper bereits vor der betreffenden Schwangerschaft festgestellt worden waren, sowie eine solche, bei welcher die Rh‐Antikörper zu irgendeiner Zcit während der laufenden Schwangerschaft nachgewiesen wurden. Auf diese Weise gelang es, die ABO‐inkompatiblen Konzeptionen mit nachfolgender Rh‐Immunisierung zahlenmäßig zu erfassen.Die vorliegenden Befunde ergaben keinen Hinweis für eine Beziehung zwischen einer ABO‐Inkompatibilität und dem mütterlichen Antikörperbildungsvermögen. SummaryHuman erythrocytes coated with subagglutinating doses of isoantibodies were strongly agglutinated upon the addition of subagglutinating quantities of rabbit anti‐human red blood cell serum.Similarly, erythrocytes coated with subagglutinating doses of heteroantibodies were agglutinated upon the addition of small amounts of isoantibodies. SummaryThe agglutination of group A or group B erythrocytes by natural isoantibodies showed a two‐stage character, dependent on at least two factors, a finding which was quite analogous to previously reported data on agglutination of rabbit erythrocytes by human natural antibodies. In the first stage of the reaction, the sensitizer, i.e., the γM‐antibody, sensitizes the erythrocytes. In the second stage the sensitized erythrocytes are agglutinated by a non‐specific factor called agglutinator. The agglutinator activity is mainly located in the β1‐globulin fraction of human serum but some activity resides also in the α1‐globulin fraction. The result of absorption experiments showed that the same agglutinator is involved in the agglutination of human A or B erythrocytes and rabbit erythrocytes. The kinetics of agglutination by natural isoantibodies was discussed in terms of sensitizer and agglutinator.RésuméL'agglutination des érythrocytes de groupe A et de groupe B par des iso‐anticorps naturels démontre qu'elle se produit en deux temps dépendants en définitive de 2 facteurs; cette découverte est en fait analogue à celle qui avait été décrite précédemment concernant l'agglutination des érythrocytes de lapins par des anticorps naturels humains. Dans le premier temps de la réaction, le sensibiliseur, par exemple, un anticorps γM, sensibilise les érythrocytes. Dans le second temps, les érythrocytes sensibilisés sont agglutinés par un facteur non spécifique appelé agglutinateur. L'activité de l'agglutinateur est en général localisé dans la fraction des β1‐globulines du sérum humain mais il y a également un peu de l'activité de cet agglutinateur dans la fraction des α1‐globulines. SummaryThe reactivity of the A1blood type specific phytohemagglutinin present inCrotalaria breviflorawas generally similar in F1and parent seeds grown under different ecological conditions.A detailed comparison of the reactivity of both one gram and individual seed samples with blood types A1, A2and B reveal general similarities in parent and F1plants, the F1being slightly less reactive to A2than P1. There was an increase in the ability of the F1generation to agglutinate B cells at 4°C compared to 22°C.The reactivity of both seeds was further studied by comparing the influence of two saccharides, two amino acids, an antibiotic, and secretor saliva on the inhibition reaction. Secretor saliva inhibited the agglutination reaction ofCrotalaria brevifloraagainst A1cells in all cases.RésuméLa réactivité de la phytohémagglutinine spécifique contre les érythrocytes A1présente dans leCrotalaria brevifloraest en général semblable dans les semences parentales et les semences de la première filiation qui ont crû dans des conditions écologiques différentes.Des comparaisons détaillées de la réactivité des échantillons de semence individuelle ou d'un gramme de semence avec des érythrocytes A1, A2, et B ont démontré des ressemblances chez les plantes parentales et de la première filiation, celles de la première filiation ayant une activité un peu moins forte à l'égard des érythrocytes A2que les parentales. SummaryFractions of erythrocytes differing in age were isolated by centrifugation. The course of the exchange transport of aldoses (galactose‐glucose) was evaluated on the basis of the galactose level reached in the apparent steady state. In the young population the level of galactose, entering the erythrocytes from the medium in exchange for glucose, attained a higher level than in the old erythrocytes. The possible relation between this finding and the structural changes in the erythrocytes membrane during the aging has been pointed out.RésuméDes erythrocytes d'äges différents ont été isolées par centrifugation. Le mode des échanges des aldoses (galactose‐glucose) a été évalué sur la base du taux de galactose obtenu dans un état d'homéostase apparent. Dans les populations érythrocytaires jeunes, le taux de galactose entrant dans les érythrocytes à partir du milieu en échange de glucose atteint un niveau plus élevé que parmi les populations d'érythrocytes ägées.
