Abstract.Employing standardized IgG‐coated51Cr‐labelled human red blood cells (RBC), variation in rosette formation among monolayers obtained from 10 healthy donors was defined, as well as variation among 24 sequential monolayers obtained from one healthy donor and 8 sequential monolayers from a second healthy donor over a 4‐month period. Rosette formation, expressed as eluate cpm per plate, averaged 220 (range 35–472) for the 10 donors; 24 determinations on one donor averaged 201 (range 59420), 8 determinations on the second donor averaged 244 (range 69–395). Striking increases in rosette formation were observed employing monolayers from 4 of 4 normal subjects during the course of viral infections, with peak values exceeding 5, 8, 13 and 14× the normal mean. Elevations were also observed with monolayers prepared from blood of selected patients with hematologic disorders; a remarkable value 168 ×the normal mean was repeatedly obtained for a patient with myeloid metaplasia whose monolayer consisted almost exclusively of mature granulocytic rosette‐forming cells. Abstract.Human heterophile antibodies reactive with various tissues of different species may be detected by indirect immunofluorescence. Results of screening sera obtained after rhesus immunization and before and after blood transfusion and renal transplantation suggest that such antibodies can appear following alloimmunization of group A and O individuals. Eleven positive sera selected for detailed study were found to have anti‐B isohaemagglutinins together with rat erythrocyte agglutinins. Absorption studies suggested that there may be some relationship between the group B antigen and the rat antigen concerned but the precise explanation for alloimmunization remains to be determine

Abstract.The major sialoglycopeptide released from bovine erythrocytes by papain has been purified and characterized. The glycopeptide contains 82% by weight carbohydrate in molar ratios of galactose ‐5.5:N‐acetylglucosamine ‐3.6:sialic acid ‐2.6:N‐acetylgalactosamine ‐1.0. The carbohydrate and amino acid composition is quite different from the glycoprotein extracted from bovine erythrocyte stroma with hot 75% ethanol. The glycopeptide is devoid of reactivity with Paul‐Bunnell heterophile antibody of infectious mononucleosis ‐ an activity expressed to high degree on the bovine erythrocyte and associated with glycoprotein. Abstract.Antisera obtained by immunizing rabbits with desialized M and N blood group glycoproteins (M' and N' glycoproteins) agglutinate desialized erythrocytes and precipitate desialized glycoproteins, but they do not react with sialic acid containing erythrocytes or glycoproteins. By absorption of anti‐M' serum with N' glycoprotein, and of anti‐N' serum with M' glycoprotein, reagents specifically agglutinating M' or N' erythrocytes were obtained. Abstract.The number of available C (rh', Rh 2) antigen sites within the Rh system was estimated using trace‐labelled antibody. Abstract.The widespread utilization of Groupamatic equipment for routine immunohaematological tests has increased the demand for serological procedures that can be performed with this machine. We describe the test method which was developed and with which the detection of HBs antigen has now been carried out routinely for 1 year. It is an automated haemagglutination inhibition reaction which is called GIPHA (Groupamatic inhibition of passive haemagglutination). The sensitivity corresponds to 2 CNTS units, the antigen weight for 1 unit being from 1 to 4 ng. The reproducibility is good; the percentage of false positive reactions is about 0.5% and that of technical problems 0.7%. The additional advantages of the machines are numerous, among which we have the possibility of 11 other simultaneous reactions. One of them is the detection of antibodies against HBs antigen, which is a passive haemagglutination test called PHAG (passive haemagglutination on Groupamatic).

Abstract.The coil planet centrifuge (CPC) can be used to measure the osmotic fragility of erythrocytes. Fragility measured by this method alters when different salts are used. The CPC and Parpart methods were used to measure the changes during storage in red cell osmotic fragility in ACD or CPD blood with or without adenine. More marked changes were detected by the CPC method, especially in old cells. The changes of fragility of erythrocytes during storage seem to occur mainly in old cells. Adenine is effective in preventing such changes.
