Abstract.A sandwich‐type solid phase radioimmunoassay, FRC‐RIA (Finnish Red Cross‐RIA) for hepatitis B surface antigen (HBsAg) is described. Polystyrene test tubes coated with sheep anti‐HBsγ‐globulin serve as the solid phase. The technique is specific and has a sensitivity of about 1 ng/ml for the subtypesadandayof HBsAg when corresponding subtype‐specific antibodies are used as reagents. The total incubation time can be shortened from over 20 h to 3 h, with almost equal sensitivity, when the incubation temperature is raised from room temperature to 45°C. Abstract.Modifications were introduced into the low ionic strength system of Lalezari. Glycine was added in place of acidified glucose and the sensitisation and aggregation phases were segregated. In the automated glycine system a stable baseline is readily obtained as strong cell aggregation, with risk of cell adhesion to coil walls, is restricted to the last few turns of the aggregation phase. Abstract.A relative of the original Rh proposita, of group R1R2, shows weak expression of his Rh antigens, and is thought to be an Rhnullheterozygote. His wife and 3 of their 4 children show normal Rh antigen expression, but one daughter showed weak Rh antigen expression, as determined by quantitative haemagglutination. The observations support the proposition that the father is heterozygous for an unlinked modifier of Rh antigen expression. Stomatocytosis, observed in the Rhnullproposita and other Rhnullindividuals, was also observed, but to a lesser degree, in the blood of an other individual thought to be an Rhnullheterozygote. This observation also supports the earlier conclusion that the Rhnullphenotype of the proposita is due to homozygosity for inactive alleles at a locus which controls the biosynthesis of precursor for Rh and LW antigens. Osmotic fragility tests showed that the Rhnullcells were more fragile than cells with normal Rh antigen expression, and cells from Rhnullheterozygotes had intermediate fragility. This is consistent with the proposition that Rh antigens are normal structural components of the red cell membrane, and the Rhnullheterozygotes show a deficiency of the Rh antigenic structures.

Abstract.2,140 pregnant Sudanese women attending the antenatal clinic in Omdurman Maternity Hospital during the period November 1975 to December 1976 had ABO and Rh typing performed. Their sera were also screened by a modified papain enzyme technique for the presence of atypical antibodies. The rate of Rhesus immunization, the frequency distribution of blood groups, the atypical antibodies encountered, and the advantages of the method used for screening the sera in a developing country are discussed.

Abstract.A study is made of the influence on hemagglutination of various physicochemical parameters, such as: cell shape, cell distance, extracellular colloid‐osmotic pressure, degree of hydration of the cell surface, and cell ζ‐potential. To a varying extent these are changed by: the addition of divers soluble polymers, the action of enzymes, the presence of various salt ions, centrifugation, agglomeration at low ionic strength, complex coacervation with cationic polymers, and the interaction with different kinds of antibodies. Among the more important parameters are cell distance and cell shape (particularly when the latter tends to spiculation), while the influence of ζ‐potential appears to be of fairly minor consequence. The action of dissolved polymers is mediated through polymer bridging, increase in extracellular colloidosmotic pressure, decrease of cell surface hydration, and change in cell shape.
