SummaryTo study the specificity of antiglobulin reactions which are dependent upon C‘ uptake by the sensitized erythrocytes, “antisera to C’” were prepared by immunizing animals with washed specific precipitates containing bound C‘. These antisera, as well as antisera to whole human serum, agglutinated the red cells of cold hemolytic anemia, Leasensitized red cells, and sensitized sheep erythrocytes exposed to sublytic doses of C’. Antisera to GAhuC‘ (specific precipitate with bound human complement) and antisera to whole human serum, were assayed for complement antiglobulin reactions by cross‐titration with sensitized sheep cells (EA) exposed to decreasing sublytic amounts of human C’ (EAhuC‘). Because EAhuC’ red cells are agglutinable with anti‐human γ globulin sera, reagents for cross‐titration should first be freed of such antibodies by specific precipitation with human y globulin. In addition, heterophile (anti‐sheep cell) antibodies should be absorbed from reagents and sources of C‘, and controls maintained to ensure absence of “rheumatoid” effect.Antisera to GAhuC' and to whole human serum agglutinated the erythrocytes from ordinary refrigerated clotted blood specimens of adults and some newborns. For this reason, these reagents are not generally applicable for compatibility testa involving clotted donor pilot tubes, or erythroblastosis tests where the clotted cord blood has been chilled.Complement, or an indistinguishable co‐factor of C', or an antigen resulting from C' activity, appeared to be the specificity of those antiglobulin reactions that are positive only when there has been complement uptake by the sensitized erythrocytes. Therefore, the term complement antiglobulin test has been suggested for these reactions.RésuméPour étudier la spécificité des réactions à l'anti‐globuline qui se produisent à la suite de la fixation du complément sur des érythrocytes aensibilisés, les auteurs ont préparé des anti‐sérums anti‐complément (anti‐C') en immunisant des animaux avec des précipités spécifiques lavés contenant du C'. Ces anti‐sérums, au même titre que lee antisénuns anti‐séums humains agglutinent les érythrocytes d'anémie hémolytique à autoanticorps froids, des érythrocytes sensibilisés par l'anti Leaet lea érythrocytes de mouton sensibilisés sur lesquels on a fixé du C' a des doses sublytiques.Les auteurs ont tenté de pratiquer des réactions à l'anti‐globuline pour le complément en testant des érythrocytes de mouton sensibilisés EA sur lesquels ils ont firé des quantités décroissantes des complément humain sublytique (EAhuC') avec des anti‐sérums anti‐GAhuC' (précipités spécifiques contenant du complément humain) et des antiaérums anti‐sérum humain. C'est pour cette raison que ces réactifs ne sont généralement pas utilisables pour des tests de compatibilité pratiqués avec des sangs récoltés dans les tubes pilotes ordinaires ou, dans les cas d'érythroblastose, avec des sangs du cordon préalablement refroidis.Le complément C', ou un facteur non décelable de C', ou nn antigène résultant de l'action de C' apparait être responsable de la sécificité de ces réactions à l'anti‐globuline qui ne sont positives que lorsqu'on fixe le complément SUI des érythrocytes sensibilisés. Diese Antiseren, sowie solche, welche durch Immunisierung mit Gesamtseren gewonnen wurden, agglutinierten Erythrozyten von Patienten mit kälteantikörperbedingter erworbener hamolytischer Anämie, Erythrozyten, welche mit Anti‐Lea‐Seren sensibilisiert worden waren sowie sensibilisierte Hammelerythrozyten, welche subhämolytische Mengen von menschlichem Komplement gebunden hatten. Mit den Antiseren gegen GAhuC' (Immunpräzipität mit gebundenem memchlichem Komplement) und den Antiseren gegen menschliches Gesamtserum wurden Komplement‐Antiglobulinreaktionen an sensibilisierten Hammelerythrozyten (EA), welche su bhämolytische Mengen von menschlichem Komplement gebunden hatten (EAhuC'), durchgeführt. Da die EAhuC'‐Erythrozyten durch Antiseren gegen menschliches γ‐Globulin agglutiniert werden, müssen die bei solchen Titrationsversuchen verwendeten Antiseren vorgängig durch Immunpräzipitation mit menschlichem γ‐Globulin von ihren Anti‐γ‐Globulin‐Antikorpern befreit werden. Außerdem müssen allfällig in den Antiseren oder den Komplementquellen enthaltene heterophile Anti‐Hammel‐erythrozyten‐Antikörper wegabsorbiert werden. Im weiteren sind die verwendeten Reagenzien auf die Abwesenheit des «rheumatoiden» Effektes zu prüfen.Die Antiseren gegen GAhuC' sowie diejenigen gegen menschliches Gesamtserum agglutinierten die Erythrozyten gekühlter geronnener Blutproben von Erwachsenen und einiger Neugeborener. Aus diesem Grunde eignen sich diese Seren nicht ohne weiteree für Vertraglichkeitsproben, bei denen geronnene Spenderblutproben verwendet werden. SummaryA blood is described which is completely nonreactive with anti‐N. This blood is type M, U‐negative. The only other type M, U‐negative blood that has been tested gives similar reactions; it is possible that all type M, U‐negatives are the same. TheMgene which was studied produces no S, s, or U and nothing which reacts with anti‐N. It is suggested that M genes normally produce a small amount of the N factor, the alternative hypothesis being that they produce a previously unsuspected factor that cross‐reacts with anti‐N. The data suggest strongly that very potent anti‐N serums would always react with most type M bloods because of the small amount of N they contain, so that one must be satisfied with anti‐N reagents of low or medium strength. Carefully performed MN blood typing for anthropologic, genetic, or medicolegal purposes is in no way discredited by these data.RésuméL'auteur décrit un sang qui ne donne aucune réaction avec l'anti‐N. Ce sang est du type M, U‐négatif. Le seul autre sang de type M, U‐négatif donne des réactions identiques et il est possible que tous les sangs de type M, U‐négatif donnent des rtactions similaires. Le géne M étudié ne produit ni S, ni s, ni U, ni quoi que que ce soit qui réagisse avec l'anti‐N. L'auteur suggère que les gènes M produisent normalement une faible quantité du facteur N, ceci en opposition avec l'hypothèse selon laquelle les gènes M peuvent produire un facteur non connu qui donne une rtaction croisée avec l'anti‐N. Les résultats obtenus laissent pourtant supposer que les sérums anti‐N très puissants peuvent toujours réagir avec la plupart des sangs du type M, parce que ceux‐ci contiennent en faible quantité de la substance N; il résulte de ceci que les réactifs anti‐N de faible et moyenne puissance sont plus que satisfaisants. Les déterminations des facteurs M et N pratiquées jusqu'ici d'une manière correcte et précise pour l'anthropologie, la génétique ou la médecine légale ne sont en aucune façon discréditées par ces observations.ZusammenfassungEine Blutprobe, die mit Anti‐N völlig negativ reagierte, wird beschrieben. Diese Blutprobe war vom Typ M, U‐negativ. Die einzige mituntersuchte Blutprobe vom Typ M, U‐negativ zeigte sehr ähnliche Reaktionen. Es erscheint deshalb als wahrscheinlich, daß sich alle M, U‐negativen Blutproben identisch verhalten. In beiden Fällen bildete das M‐Gen kein S, s oder U sowie nichts, was mit Anti‐N reagiert. Es ist anzunehmen, daß die M‐Gene normalerweise entweder eine kleine Menge N‐Antigen bilden oder daß sie ein Antigen bilden, welches mit Anti‐N‐Anti‐körpern eine Kreuzreaktion eingeht. Die Untersuchungsergebnisse zeigen, daß sehr kräftige Anti‐N‐Seren mit der überwiegenden Mehrzabl der M‐Blute reagieren, da diese eine kleine Menge N‐Antigen enthalten. SummaryOn the basis of previously published experiments dealing with the solubility of plasma proteins in the presence of polyphosphate, a fractionation method for human plasma has been developed. This procedure is simple and especially designed for the production of the fractions of clinical interest.A first fraction containing predominantly fibrinogen is precepitated with 0.4% polyphosphoric acid at a pH between 5.0 and 5.3. A second fraction containing virtually all the albumin is then precipitated from the supernatant with 0.8 % polyphosphoric acid at a pH of 4.3. The γ‐globulin remains in the supernatant. The purification of the last two fractions is described in detail.The raw albumin fraction is dissolved at neutral pH. Then the polyphosphate is eliminated by passage over a low crosslinked anion‐exchanger. By subsequent desalting on a mixed bed ion exchange column a plasma protein solution is obtained which can be pasteurized at 60° C. It contains 60 % of the plasma proteins of which 80 % are albumin and 20% globulins. It is free of any risk of hepatitis transmission and seems therefore very well suited as a plasma expander.From the supernatant of the albumin precipitate the polyphosphate is removed by ion exchange. The γ‐globulin is then precipitated at a pH of 7 with 25% ethanol. The yield is excellent. This γ‐globulin has a purity of at least 90% and contains the antibodies in full potency.The composition of the fractions is recorded and it is also shown that the amount of polyphosphate eventually remaining in the final product is so small that there is no danger of toxicity. There are no indications of denaturation of the plasma proteins by polyphosphate within the pH range employed (7.0–4.3).The average degree of polymerization of the polyphosphoric acid used has virtually no effect on its precipitating action, at least as long as the lower oligophosphates are excluded.Finally the method is compared to that proposed byRane and Newhouser, in which cyclic tetrametaphosphate is used for fractionation.The polyphosphate method, as described here, may prove of practical value in preparing from human plasma the two fractions for which there is today the greatest demand from the clinicians: a stable pasteurized plasma protein solution, and an antibody‐globulin preparation.RésuméSur la base de recherches precedemment publiées concernant la solubilité des protéines plasmatiques en présence de polyphosphate, les auteurs ont mis au point une méthode de fractionnement de plasma humain; cette méthode est simple et est plus particulihrement adaptée à la préparation de fractions utilisables en clinique.Une premiere fraction, composée principalement de fibrinogène, est précipité à pH 5‐5,3 avec de l'acide polyphosphorique à 0,4 %. Du surnageant on précipite, à pH 4,3, au moyen d'acide polyphosphorique à 0,8%, une fraction qui contient pratiquement toute l'albumine. La γ‐globuline reste dans le surnageant. Le traitement subséquent de ces deux dernières fractions est décrit. La fraction brute d'albumine est mise en solution par neutralisation et est liberée du polyphosphate et des sels au moyen échangeurs d'ions et l'on obtient une solution de protéines plasmatiques que l'on peut pasteuriser, et qui semble indiquée comme solution de transfusion ne comportant pas de risque de transmission d'hépatite. Elle contient environ 60% des protéines plasmatiques totales, dont 80% d'albumine.Après précipitation de l'albumine et élimination du polyphosphate au moyen d'échangeurs d'ions, le surnageant est porté à pH 7 et est additionné de 25% d'éthanol. On obtient ainsi avec un très bon rendement une γ‐globuline d'une pureté de plus de 90 % qui contient les anticorps entièrement actifs. La composition des fractions est indiquée et il est aussi démontré que les quantités de polyphosphate éventuellement encore contenues dans les préparations terminées sont extrèmement faibles de sorte qu'aucun effet toxique n'est à craindre. II n'y a pas non plus d'indices permettant de supposer que les protéines sont dénaturées par le polyphosphate aux pH utilisés (entre 7 et 4,3), même pas à température ordinaire.Le degré moyen de polymérisation de l'acide polyphosphorique utilisé n'a pratiquement pas d'influence sur l'action précipitante tant qu'il ne s'agit pas des acids oligophosphoriques inférieurs. Pour terminer, la méthode proposée est comparée avec celle deRane et Newhouser;ces auteurs ont procédè à une précipitation fractionnée au moyen de tétramétaphosphate cyclique pur.La méthode rapportée ici représente un procédé rationnel, permettant de transformer le plasma en un produit de transfusion ne présentant pas de risque de transmission d'hépatite, et qui est susceptible d'être conservé comme solution sous forme stérile; la γ globuline, dont l'importance devient toujours plus considérable en clinique, peut être préparée comme sous‐produit avec un excellent rendement.ZusammenfassungAuf Grund früher publizierter Untersuchungen über die Loslichkeit von Plasmaproteinen in Gegenwart von Polyphosphat wurde eine Fraktionierungsmethode für humanes Plasma ausgearbeitet, die einfach und speziell für die Gewinnung der klinisch wichtigsten Fraktionen zugeschnitten ist.Mit 0,4% Polyphosphorsaure wird bei pH 5‐5,3 eine erste, vor‐wiegend Fibrinogen enthaltende Fraktion ausgefallt. Aus dem Über‐stehenden fällt bei pH 4,3 mit 0,8 % Polyphosphorsäure eine Fraktion, die praktisch alles Albumin enthält. Das γ‐Globulin verbleibt im Überstehenden. Die Aufarbeitung der beiden letztgenannten Fraktionen wird beschrieben.Die rohe Alhuminfraktion wird unter Neutralisation gelöst und mit Hilfe eines schwachvernetzten Anionenaustauschers von Polyphosphat befreit. Durch anschlieoende Totalentsalzung mittels Ionenaustausch wird eine bei 60° C pasteurisierbare Plasmaproteinlösung erhalten, die als hepatitissicheres Transfusionsmittel sehr geeignet scheint. Sie enthält ca. 60% des gesamten Plasmaproteins, und davon sind 80% Albumin. Das Überstehende von der Albuminfällung kann nach Eliminierung des Polyphosphates durch Ionenaustausch bei pH 7 mit 25 % Aethanol gefällt werden. Dabei wird in hervorragender Ausbeute ein γ‐Globulin von über 90prozentiger Reinheit erhalten, welches die Antikörper in voller Wirksamkeit enthält.Die Zusammensetzung der Fraktionen wird angegeben und es wird auch gezeigt, daβ die in den fertigen Präparaten eventuell noch vorhandenen Mengen Polyphosphat außerordentlich gering sind, so daß keinerlei toxische Wirkungen zu befürchten sind. Es liegen auch keine Anzeichen dafür vor, daβ die Plasmaproteine durch Polyphosphat im angewendeten pH‐Bereich (zwischen 7 und 4,3) denaturiert werden, auch nicht bei Zimmertemperatur.Der Durchschnittspolymerisationsgrad der verwendeten Polyphosphorsäure hat auf die Fällwirkung praktisch keinen Einfluß, solange nicht zu den niederen Oligophosphaten übergegangen wird. SummaryA new simple method of hemagglutination (HAOS) is described. Human red blood cells of A‐group, for example, suspended in AB‐serum of non‐secretor are smeared on slide. After fixation with methanol, the smear is exposed to anti‐A (normal human B‐serum) and washed. A‐cell saline suspension is then brought into contact with the smear. Hemagglutination takes place around the fixed red cells. The reaction is strictly specific. Some fundamental observations not possible by conventional methods are described. The mechanism and possible applications of HAOS are discussed.RésuméL'auteur décrit une nouvelle métbode simple d'hémagglutination (HAOS). Des érythrocytes de groupe A, par exemple, mis en suspension dans un sérum AB d'un non secreteur sont Btales sur un porteobjet. Après fixation au méthanol, le frottis est mis en contact avec de l'anti‐A (sérum normal humain B), puis, bien havé. Une suspension d'érythrocytes A en solution physiologique est ensuite mise en contact avec le frottis. On observe alors une agglutination des érythrocytes aux cellules fixées. La réaction est strictement spécifique et l'auteur décrit quelques observations qui ne sontpaspossiblespar desmethodes usuelles. II discute des mécanismes et des applications possibles du HAOS.ZusammenfassungEine neue einfache Methode der Hamagglutination (HAOS) wird beschrieben. Menschliche Erythrozyten, beispielsweise der Gruppe A, werden in AB‐Serum eines Nichtsekretors aufgeschwemmt und auf einem Objekttrager ausgestrichen. Nach Fixation mit Methanol wird der Ausstrich mit Anti‐A‐Serum (normales menschliches B‐Serum) behandelt und gewaschen. Eine Aufschwemmung von A‐Zellen in Kochsalzlösung wird auf den Ausstrich gebracht, worauf rund um die fixierten Erythrozyten die Hämagglutination in Erscheinung tritt.
