Abstract.This review deals with the rationale for the use of hemodilution in patients not subjected to open heart surgery. The claim for an optimum of circulatory oxygen transport at 30% hematocrit has been disproved; hemodilution thus simply means acute normovolemic anemia. Accordingly, it generates a cardiovascular strain and particularly jeopardizes cerebral and myocardial oxygen supply. Potentially serious clinical side effects have been reported. Abstract.In man following the autologous transfusion of blood previously frozen with 14% low molecular weight‐hydroxyethylated amylopectin (cryo‐HES), the clearance of this material from the intravascular space was compound, and appeared to consist of exponential components. The overall half‐life ‐ however, was 10.6 ± 3.0 (SD) h. Approximately 17% of the total infused cryo‐HES was excreted in the urine 1 h postinjection, and 40% by 72 h. The erythrocyte sedimentation rate (ESR) was not affected by the presence of this substance in the bloodstream of the recipient. Abstract.An immunoadsorbent was prepared by coupling rabbit and human antibodies against human factor VIII to Sepharose CL‐2B. The resulting insoluble antibodies completely removed factor VIII and fibrinogen from normal human citrate plasma. Other coagulation factors were satisfactorily recovered in the eluted plasma. Following addition of fibrinogen, the factor VIII‐deficient plasma was used for calibration of the one‐stage factor VIII assay and compared with the hemophilic plasma. Parallel straight lines were obtained in the log‐log plot against VIII:C, indicating that the artificial reagent can be used as a substitute for hemophilic plasma in determination of factor VIII procoagulant activity. Abstract.Employing the Haemonetics Blood Processor (IFC), a relatively pure platelet concentrate can be prepared by collecting only the first portion of the PRP leaving the centrifuge bowl (Fraction I). A subsequent fraction containing RBC and WBC contaminants (Fraction II*) can be purified by means of a second centrifugation, using a conventional blood bank centrifuge (Fraction II), if transfusion of these contaminants would be detrimental to the recipient. Utilising the new 1.4% Na3‐citrate anticoagulant, platelet metabolic parameters (ATP, ADP, AMP, lactate and pyruvate) as well as O2‐uptake, were determined in Fractions I and II prepared from 10 normal healthy subjects. In contrast to previous studies reporting marked dysfunction in platelets contained in Fraction II when standard ACD‐A was used during IFC, we observed no significant difference (Student'sttest) in the present study between Fractions I and II, in regard to platelet metabolism, when using the new anticoagulant. Abstract.In vitroproliferation and differentiation of hematopoietic stem cells was studied in a patient with aplastic anemia. Prior to therapy his peripheral blood contained a very low number of myeloid progenitors but a normal number of cells forming lymphoid colonies. Moreover, peripheral blood lymphocytes of this patient were able to suppressin vitroformation of myeloid colonies but not lymphoid colonies. This suppression effect was found to be sensitive to prednisolone and antithymocytic globulin. Following treatment with prednisolone, during which an apparent hematological recovery was observed, the level of lymphoid progenitors fell, but myeloid committed cells returned to normal and hematopoietic suppression was no longer detectable. These results indicate that cells suppressing hematopoiesis may circulate in the peripheral blood of some patients with aplastic anemia and the detection and testing of susceptibility of these cells to immunosuppressive drugs may be helpful in monitoring treatment and prognosis.

Abstract.After 8 months of storage of normal red cells in liquid nitrogen, red cell enzyme activities were essentially unchanged, except for triose phosphate isomerase which was reduced by about half. Mutant pyruvate kinase (PK), PK Tokyo I, showed no changes in kinetic or electrophoretic properties after 8 months of storage. These findings indicate that red cells stored for a long term in liquid nitrogen can still be used for the study of mutant PK.

Abstract.Cryoprecipitate prepared by a rapid thawing technique was pooled in batches of 600–720 donor units and washed with ice‐cold Tris‐citrate‐NaCl solution. After dissolving at 37°C, it was adsorbed with Al(OH)3and kaolin, and cleared by centrifugation. The supernatant, diluted with 5% dextrose was passed repeatedly through a bed of Celite, filtered through a 293 mm × 0.3 μm membrane disc and lyophilized. Typical composition was 15 U ml‐1factor VIII, and 40 mg ml‐1protein with a yield of 300 U/l of starting plasma.
