SummaryP was originally conceived as a unique serum substance, which in the assumed absence of Ab, inactivated C and, in conjunction with C, participated in the killing of bacteria, the inactivation of viruses, and the hemolysis of abnormal red blood cells. More recent evidence has indicated, on the contrary, that Ab is an essential requirement for these reactions. In this revised orientation of the relationship of P and normal serum activities, the part played by P is uncertain and remains to be determined. There is no rigorous evidence at this time that P is involved in nonspecific resistance to infection or in neoplastic growth. The introduction of P as a serological entity nonetheless contributed a great deal to scientific advance by stimulating and revitalizing research efforts in many neglected areas, particularly those related to non‐specific resistance to infection.ZusammenfassungDas Properdin wurde ursprünglich als eine besondere Serumsubstanz aufgefaßt, welche in Abwesenheit von Antikörpern Komplement inaktiviert und zusammen mit diesem imstande ist, Bakterien abzutöten, Yiren zu inaktivieren und abnormale Erythrozyten zu lysieren. Neuere Untersuchungen zeigten jedoch, daß Antikörper bei diesen Reaktionen eine ausschlaggebende Rolle spielen. Die Bedeutung des Properdins ist heute unklarer als je; seine Beziehung zu den normalen Serumaktivitäten bedarf erneuter Bearbeitung. Zur Zeit fehlen bindende Hinweise dafür, daß dem Properdin bei der unspezifischen Resistenz oder beim neoplastischen Wachstum eine wesentliche Bedeutung zukommt. Trotzdem war die seinerzeitige Einführung des Properdinbegriffes in die Immunologie von beträchtlichem heuristischem Wert, als dadurch brachliegende Forschungsgebiete, besonders das Problem der unspezifischen Resistenz gegen Infektionen, in den Brennpunkt des Interesses rückten.RésuméInitialement, la properdine avec le complément était considérée comme la seule substance sérique qui, en l'absence supposée d'anticorps après inactivation du complément, participaient à la destruction de bactéries, à l'inhibition des virus et à l'hémolyse d'érythrocytes anormaux. On a eu récemment la preuve que, au con‐traire, l'anticorps est un élément indispensable pour ces réactions. Par suite de cette nouvelle conception, le rôle joué par la properdine est devenu très incertain et reste à déterminer. Il n'y a pas actuellement d'évidence nette que la properdine participe à la résistance non spécifique aux infections et à la croissance néoplastique. Il n'en demeure pas moins que la properdine, en tant qu'entité sérologique a contribué grandement au progrès scientifique en stimulant la recherche dans de nombreux domaines tenus à l'écart et plus particulièrement dans

SummarySo‐called naturally‐occurring antibodies seem, as a rule, to be composed of 19 S globulin even when they behave as haemolysins (anti‐P + P1or as partially incomplete antibodies (anti‐Le1). 19 S naturally‐occurring antibodies may be accompanied by 7 S antibody with the same specificity which has possibly been produced as an immune response; an example of anti‐Leaconsisting partly of 7S and partly of 19S globulin is described.Immune incomplete antibodies seem normally to be composed of 7S gamma globulin but it appears that they may be composed partly of 19 S globulin; in the present paper two antibodies of this kind, one anti‐D and one anti‐K, are described.ZusammenfassungDie sogenannten natürlichen Antikörper scheinen in der Regel aus 19 S‐Globulin zu bestehen. Dies gilt selbst dann, wenn diese Antikörper den Charakter von Hämolysinen (Anti‐P+P1) oder partiell inkompletten Antikörpern (Anti‐Lea) aufweisen. Neben den natürlichen 19 S‐Antikörpern kommen 7 S‐Antikörper mit derselben Spezifität vor, die wahrscheinlich auf Immunisierungen zurückzuführen sind. SummaryThe serum of a patient suffering from general arteriosclerosis and chronic pyelonephritis with mild anaemia was investigated because of difficulties during compatibility tests prior to blood transfusion. Red cells exposed to the serum were strongly agglutinated when subsequently washed in saline. The responsible substance was identified as a γ‐globulin of high molecular weight, also possessing some but not all of the usual rheumatoid factor activities.ZusammenfassungWegen Schwierigkeiten bei der Kreuzprobe wurde das Serum eines Patienten mit generalisierter Arteriosklerose, chronischer Pyelonephritis und leichter Anämie näher untersucht. Erythrozyten, die in diesem Serum aufgeschwemmt waren, warden beim nachfolgenden Waschen in physiologischer Kochsalzlösung kräftig agglutiniert. Die für dieses Phänomen verantwortliche Substanz erwies sich als hochmolekulares γ‐Globulin, welches einige, jedoch nicht alle Eigenschaften des Rheumafaktors aufwies.RésuméLe sérum d'un patient atteint d'artériosclérose généralisée et de pyelonéphrite chronique avec anémie a été examiné par suite des difficultés que ce sérum présentait lors des épreuves de compatibilité pratiquées en vue de transfusions. SummaryStudies of the blocking of e sites by anti‐G indicate interesting interrelationships of e and G. They seem to indicate that all the different Rh specificities produced by a Rh gene are contained in a single molecule. Some of the results suggest that all the Rh specificities produced by a heterozygote may be on the same molecule, in which case the concept of unit agglutinogens produced by single genes [2] would have to be modified.ZusammenfassungUntersuchungen über die Blockierung von e durch Anti‐G‐Antikörper ergaben interessante Hinweise über die gegenseitigen Beziehungen der Rhesusantigene e und G. Die Versuchsergebnisse zeigten, daß wahrscheinlich sämtliche durch ein Rh‐Gen bedingten Rh‐Spezifitäten in einem einzigen Molekül enthalten sind. ZusammenfassungEs wird über experimentelle Untersuchungen zur Erzeugung der postnatal induzierten, «erworbenen Immuntoleranz» berichtet. Insgesamt wurden 24 Kaninchen unmittelbar nach der Geburt hochgereinigtes Albumin, γ‐Globulin und ein weniger gereinigtes antihämophiles Globulin vom Rind intraperitoneal injiziert. Im Abstand von 3 bis 5 Monaten wurde mit Antigen reinjiziert und die Antikörperbildung mit dem Gel‐Diffusionstest nachOuchterlonyund dem Hämagglutinationstest nachBoydenkontrolliert.Folgende Ergebnisse wurden erhalten:1In Übereinstimmung mit den Angaben der Literatur wurde gefunden, daß die Erzeugung der Immuntoleranz von der Antigendosis abhängig ist. Während durch Vorbehandlung mit 25 mg Rinderalbumin die Antikörperbildung nicht vollkommen unterdrückt werden konnte, fiihrte die Verabreichung von 50 mg zu einer Immuntoleranz bisher bis zu 22 Monaten Dauer.2Mit 50 mg Rinder‐γ‐Globulin konnte lediglich eine partielle Immuntoleranz erzielt werden.3Versuche mit einem Mischantigen, das sich zu gleichen Teilen aus Rinderalbumin und Rinder‐γ‐Globulin zusammensetzte, ergaben eine Immuntoleranz gegen Albumin, nicht gegen γ‐Globulin.4Der Zusatz eines Adjuvans (Al(OH)3) zum Antigen hat bei der Erzeugung einer Immuntoleranz den gleichen Antigen‐einsparenden Effekt wie bei der aktiven Immunisierung. Es konnte mit 5 mg Rinderalbumin unter Zusatz von Aluminium‐hydroxyd eine totale Immuntoleranz gegen Rinderalbumin erzielt werden.5Mit einem relativ ungereinigten antihämophilen Globulin aus Rinderplasma wurde nur eine partielle Immuntoleranz erhalten.6Zwischen den Seren der immuntoleranten und der antikörperbildenden Tiere bestand kein Unterschied im γ‐Globulingehalt.7Es gelang nicht, eine bestehende Immuntoleranz gegen Rinderalbumin durch die Infusion großer Mengen von Antikörpern gegen Rinderalbumin aufzuheben.8Durch eine Geburt wurde eine beim Muttertier seit längerer Zeit bestehende Immuntoleranz aufgehoben. Die Jungtiere waren erwartungsgemäß nicht immuntolerant.Abschließend wird das Wesen der Immuntoleranz diskutiert und kurz auf die klinische Bedeutung eingegangen.SummaryIn experiments designed to induce postnatally acquired immunological tolerance. 24 newborn rabbits were given intraperitoneal injections of highly purified bovine albumin, gamma globulin, and antihemophilic globulin (less highly purified). Three to five months later, antigen was reinjected and tests for antibody formation were performed using Ouchterlony's gel diffusion and Boyden's hemagglutination method. The results were as follows:1In accordance with the experience of other workers, the induction of immunological tolerance was found to be dependent upon the antigen dose. Thus, with 25 mg. of bovine albumin, antibody formation could not be entirely suppressed, but 50 mg. produced complete tolerance lasting up to 22 months so far.2With 50 mg. of bovine gamma globulin only partial tolerance could be achieved.3A mixture of equal parts of albumin and gamma globulin produced tolerance against albumin but not against gamma globulin.4The use of Al(OH)3as an adjuvant had the same antigen‐saving effect in the induction of immunological tolerance as in active immunization. Thus, 5 mg. of bovine albumin, when injected with the adjuvant, were sufficient to induce complete tolerance against the antigen.5With a relatively impure preparation of antihemophilic globulin from bovine plasma, only partial tolerance was obtained.6There was no difference in serum gamma globulin content between tolerant animals and those which had formed antibodies.7Tolerance against bovine albumin could not be abolished by large doses of anti‐bovine‐albumin antibody.8Tolerance of considerable standing in a female animal was abolished by birth. As expected, no tolerance was found in the young.The nature and clinical significance of immunological tolerance are discussed.RésuméLes auteurs rapportent sur les recherches expérimentales destinées à créer une tolérance immunitaire acquise induite postnatale. En tout, 24 lapins reçurent, au moment de la naissance, des injections intrapéritonéales d'albumine hautement purifiée, de gamma‐globuline et de globuline antihémophilique non purifiée de bœuf. Après 3 et 5 mois, on réinjecta des antigènes aux animaux et la formation des anticorps fut contrôlée au moyen du test de diffusion en milieu gélosé d'Ouchterlony et du test d'hémagglutination de Boyden. Les résultats suivants furent obtenus:1° En accord avec les données de la littérature, la formation de la tolérance immunitaire dépend de la dose d'antigène. Alors qu'un prétraitement par 25 mg d'albumine bovine ne peut pas inhiber complètement la formation d'anticorps, une injection de 50 mg d'albumine bovine produisait une tolérance immunitaire allant jusqu'à 22 mois.2° Avec 50 mg de gamma‐globuline, on obtient seulement une tolérance immunitaire partielle.3° Des mélanges d'antigènes composés par moitié d'albumine et par moitié de gamma‐globuline bovine produisent une tolérance immunitaire à l'égard de l'albumine, mais pas de tolérance à l'égard de la gamma‐globuline.4° L'adjonction à l'antigène d'un adjuvant (Al[OH]3) a, lors de la formation de la tolérance immunitaire, le même effet d'économie de l'antigène à injecter que celui observé lors de l'immunisation active. (Avec 5 mg d'albumine bovine auxquels on adjoint de l'hydroxyde d'aluminium, on obtient une tolérance immunitaire totale à l'égard de l'albumine bovine.)5° La globuline antihémophilique non purifiée, préparée à partir du plasma bovin, ne produit qu'une tolérance immunitaire partielle.6° Entre les sérums des animaux immuno‐tolérants et les sérums des animaux formant des anticorps, on n'observe pas de différence quant à la quantité de gammaglobuline sérique.7° II n'a pas été possible de supprimer une tolérance immunitaire à l'égard de l'albumine bovine en injectant une grande quantité d'anticorps anti‐albumine bovine.8° La tolérance immunitaire d'un animal, depuis longtemps induite par la mère de cet animal, cesse aussitôt après la naissance. ZusammenfassungIn dem Serum eines 2 1/4 Jahre alten Knaben, bei dem im Rahinen einer fieberhaften Erkrankung eine akute passagere autohämolytische Krise mit massiver Hämoglobinurie eingetreten war, konnte eine nichtsyphilitisehe Variante desDonath‐LandsteinerschenAntikörpers nachgewiesen werden. In quantitativen serologischen Untersuchungen wurde sie mit einer syphüitischen Variante der gleichen Antikörperkategorie verglichen. Dabei erwiesen sich in beiden Fäüen alsidentisch: die Abhängigkeit der Ambozeptorbindung vom pH‐Wert mit einem Optimum in der Cegend von pH 7,7, das Maximum der Ambozeptorbindung bei 0° und der steile Abfall der hämolytischen Aktivität mit zunehmender Serumverdünnung bei relativ niedrigem absolutem Titerwert.Unterschiedezeigte dagegen die Wärmeamplitude des hämolytischen Ambozeptors, die bei der nichtsyphilitischen Variante breiter war und bis etwa 25° reichte. Der klinische Verlauf macht es wahrscheinlich, daß die Wärmeamplitude im akuten Krankheitsstadium noch höher gewesen ist. Damit wäre erklärt, daß es bei dem Patienten trotz warmer klimatischer Bedingungen zu einer autohämolytischen Krise kommen konnte. Die Befunde zeigen, daß die nichtsyphilitische Variante desDonath‐LandsteinerschenAntikörpers in einer Modifikation auftreten kann, die in bezug auf die autohämolytische Potenz pathogenetisch und klinisch maligner als die syphilitische ist, von der sie sich aber auch durch eine relativ rasche spontane Rückbildung unterscheidet.SummaryIn a boy aged 2 1/4 years an acute, transitory attack of autohemolysis with massive hemoglobinuria occured in the course of a febrile disease. In the boy's serum a non‐syphilitic example of Donath‐Landsteiner antibody was found. Its serological properties were compared with those of a syphilitic example of the antibody. Featurescommonto both types were the following: the binding of the antibody was dependent upon pH, the optimal pH being 7.7; binding of the antibody was maximal at 0°; serial dilution of the sera caused a steep drop in hemolytic activity, i. e., their hemolytic titre was relatively low. The followingdifferenceswere found: the termal amplitude of the non‐syphilitic antibody was wider than that of the syphilitic one, its upper limit being 25°. It would appear that at the height of the hemolytic attack the thermal amplitude must have been still wider; this could explain why the attack occurred in May, when outdoor temperatures were high for the time of year. The non‐syphilitic type of Donath‐Landsteiner antibody would thus appear to be more dangerous than the syphilitic one with respect to autohemolytic potency; it differs from the syphilitic type by its fairly rapid spontaneous retrogression.RésuméDans le sérum d'un garçon âgé de 2 ans présentant une affection fébrile, il a pu être mis en évidence une variante non syphilitique de l'anticorps de Donath‐Landsteiner au cours d'une crise passagère d'automéholyse avec hémoglobinurie massive. Les examens sérologiques quantitatifs ont permis d'établir un parallèle avec la variante syphilitique de ce genre d'anticorps. Dans les deux cas, on observait que le pH optimal pour la fixation de l'ambocepteur se situait aux environs de 7,7, que la température optimale pour la fixation de l'ambocepteur était de 0°C. et que, en diluant le sérum progressivement, il y avait une diminution rapide de I'activité hémolytique dès les premieres dilutions. Par centre, on observait des différences dans l'amplitude thermique de l'ambocepteur hémolytique: en effet, la variante non syphilitique atteignait une température d'environ 25°. L'évolution clinique de ce cas laisse supposer que cette amplitude thermique devait être encore plus haute dans les poussées aiguës de l'affection. Ceci explique le fait que le patient a présenté des crises d'auto‐hémolyse malgré un climat assez chaud. Ces recherches démontrent que la variante non syphilitique de l'anticorps de Donath‐Landsteiner peut présenter des modifications qui expliquent au point de vue pathogénique la puissance du processus autohémolytique et au point de vue clinique la malignité plus grande que celle de l'anticorps syphilitique; il

RésuméL'activité anti‐Gma, anti‐Gmb et anti‐Gmx, présente dans certains sérums d'individus normaux, est de nature euglobulinique et en relation avec les fractions macroglobuliniques des γ‐globulines (β2M).Cette activité parait être supportée soit par la β2M seule, soit par l'association β2M‐γ‐globulines.Elle existe indépendamment du facteur rhumatoi'de.L'activité anti‐Gm n'a pas été rencontrée dans des sérums présentant une macroglobulinémie pathologique.SummaryThe anti‐Gma, anti‐Gmband anti‐Gmxactivity present in the serum of certain normal individuals is a function of the euglobulin fraction and is related to the macroglobulin fraction of the γ‐globulins (β2M). This activity appears to be exercised either by the β2M fraction alone or in association with the γ‐globulins. It occurs independently of the rheumatoid arthritis factor. SummarySera from patients with idiopathic granulocytopenia and thrombopenic purpura, which were serologically positive in the anti‐globulin‐consumption test, and normal sera as controls were incubated with leucocytes and thrombocytes, respectively. The serum proteins remaining fixed onto these cells after several washings were removed by elution at an elevated temperature. The proteins contained in the eluates were studied serologically with the anti‐globulin‐consumption test, in vivo in rabbits, electrophoretically and by sedimentation analysis.The anti‐globulin‐consumption tests of the eluates containing the patient's autoantibodies were positive and the injection of these eluates into rabbits caused a cytopenic effect corresponding with the cytopenia observed in the respective patients.All eluates containing autoantibodies showed a 7 S fraction in the sedimentation diagram and in the electrophoretic evaluation a gamma globulin fraction. These eluates contained 4.6 S and sometimes 18 S fractions in the same way as the control eluates.These results indicate that it is possible to isolate by the elution method autoantibodies against leucocytes and platelets. The protein fraction with the mobility of gamma globulin and the sedimentation constant 7 S which was found in the autoantibody‐containing eluates seems to be identical with the antibody. The observation of other protein fractions in both these eluates and in the controls allows the conclusion that during incubation of autoantibody containing sera with their appropriate cells specific binding and non‐specific adsorption of proteins is involved.ZusammenfassungSera von Patienten mit idiopathischer Granulozytopenie und thrombopenischer Purpura, die im Anti‐Globulin‐Consumptionstest positiv reagierten, und normale Kontrollsera wurden mit Leukozyten bzw. Thrombozyten inkubiert. Die nach mehreren Waschungen an den Zellen gebunden bleibenden Protéine wurden bei erhöhter Temperatur eluiert. Die in die Eluate überführten Eiweißkörper wurden mit serologischen Methoden (Anti‐Globulin‐Consumption), in vivo bei Kaninchen, elektrophoretisch und mittels Ultrazentrifuge untersucht.Der Anti‐Globulin‐Consumptionstest war bei den antikörperhaltigen Eluaten positiv, und diese verursachten bei den Kaninchen einen zytopenischen Effekt, der mit der bei den jeweiligen Patienten beobachteten Zytopenie übereinstimmte.Alle autoantikörperhaltigen Eluate zeigten im Sedimentationsdiagramm eine 7‐S‐Fraktion und in der Elektrophorese eine Gamma‐Globulin‐Fraktion. In gleicher Weise wie die Kontroll‐Eluate wiesen sie ferner regelmäßig eine 4.6‐S‐ und manchmal eine 18‐S‐Fraktion auf.Diese Ergebnisse deuten darauf hin, daß man mit der Elutionsmethode Autoantikörper gegen Leukozyten und Thrombozyten isolieren kann. Die Proteinfraktion mit der Mobilität eines Gamma‐Globulins und der Sedimentationskonstante 7 S, die regelmäßig in den autoantikörperhaltigen Eluaten gefunden wurde, scheint mit dem Antikörper identisch zu sein. Die Beobachtung, daß andere Proteinfraktionen sowohl in diesen Eluaten wie in den Kontrolleluaten vorkommen, läßt den Schluß zu, daß während der Inkubation autoantikörperhaltiger Sera mit den ihnen entsprechenden Zellen, sowohl eine spezifische Bindung wie eine unspezifische Adsorption an die Zelloberfläche auftreten.RésuméDes sérums de patients atteints de granulocytopénies idiopathiques et de purpura thrombocytopéniques qui présentaient une réaction positive au test de consommation de l'anti‐globuline, et, des sérums témoins normaux ont été incubés avec des leucocytes et des thrombocytes, respectivement.Après plusieurs lavages successifs, les protéines résiduelles fixées à ces cellules ont été éluées par chauifage à température élevée. Les protéines contenues dans l'éluat ont été étudiées sérologiquement au moyen du test de consommation de l'antiglobuline, in vivo par injection chez le lapin, par l'électrophorèse et par sédimentation à l'ultracentrifuge.Les tests de consommation de l'anti‐globuline pratiqués avec les éluats provenant de sérums de patients contenant des auto‐anticorps étaient positifs; l'injection de ces mêmes éluats au lapin provoquait une cytopénie correspondante à la cytopénie observée chez le patient.Tous les éluats contenant des auto‐anticorps présentaient la fraction 7 S dans le diagramme de sédimentation et, à l'électrophorèse, une fraction gamma‐globulinique. Ces éluats contenaient en outre des fractions 4.6 S et parfois 18 S au même titre que les éluats provenant de sérums témoins.Ces résultats démontrent qu'il est possible d'isoler, grâce au procédé de l'élution, des auto‐anticorps antileucocytaires et antithrombocytaires. La fraction protéinique trouvée dans les éluats provenant de sérums contenant des auto‐anticorps présente la mobilité électro‐phorétique des gamma‐globulines et la constante de sédimentation 7 S et semble donc être identique à un anticorps. La mise en évidence d'autres fractions protéiniques dans ces éluats et dans les éluats témoins nous autorise à conclure que, durant

RésuméFaisant suite aux recherches deAeplietFrei, on a examiné Taction nocive sur le sang des matériaux suivants, pouvant entrer dans la construction des organes artificiels: Makrolon, Tygon S‐22–1 et adhésifs Araldite. Ces matériaux se sont révélés très peu nocifs pour les facteurs sanguins dosés, c'est‐à‐dire la prothrombine, les thrombocytes, le complément, l'hémolyse et Thématocrite. Le Tygon a Tinconvénient de fixer de la fibrine.La rugosité de la paroi n'influence pas le taux de prothrombine et le nombre des thrombocytes; en revanche, elle augmente Thémolyse.Un precédé de nettoyage par des enzymes protéolytiques a été mis au point et ses limites ont été précisées. Ces enzymes peuvent «digérer» totalement les protéines sanguines fixées aux parois après rinçage. Ce procédé peut être utilisé lorsque les matériaux entrant en contact avec le sang ne supportent pas les bases fortes, et lorsqu'on ne peut recourir à des éléments remplaçables qui ne servent qu'une fois.SummaryFollowing researches byAepliandFrei, it has been examined how blood can be altered by such materials as Makrolon, Tygon S‐22–1 and Araldite, which can be used in the construction of artificial organs. These materials have proved themselves as being practically noninjurious to the determined blood factors, which are prothrombin, platelets, complement, hemolysis and hematocrit. The disadvantage of the Tygon is that it fixes some fibrin. It has been proven that roughness of the wall has no importance for prothrombin and platelets, but on the contrary it increases hemolysis.Besides, it has been demonstrated that proteolytic enzymes are able to digest totally the blood proteins adhering to the walls after a rinsing with water. This method of “cleaning” can be used when material contacting blood cannot tolerate strong bases, or when it cannot be disposable.ZusammenfassungIn Weiterführung der Untersuchungen vonAepliundFreiwurden einige, möglicherweise für die Konstruktion von künstlichen Organen in Frage kommende Kunststoffe auf eine allfällige schädigende Wirkung gegenüber Blut untersucht. Es handelte sich um Makrolon, Tygon S‐22–1 und Araldit. Diese Materialien erwiesen sich als bemerkenswert unschädlich gegenüber folgenden Bluteigenschaften: Prothrombin, Thrombozyten, Komplement, Hämolyse und Hämatokrit. Das Tygon zeigte als Nachteil eine Fibrinablagerung. Die Prothrombinaktivität und die Thrombozytenzahl wurde durch Wandrauhigkeiten nicht beeinflußt; diese steigerten hingegen die Hämolyse.Es wurde ein Reinigungsverfahren mit proteolytischen Enzymen ausgearbeitet. Es gelingt damit, die nach dem Durchspülen noch der Wand anhaftenden Bluteiweiße völlig wegzudauen. Dieses Verfahren dürfte sich dann als brauchbar erweisen, wenn die mit dem Blut in Kontakt gestandenen Teile eine Behandlung mit starken Laugen nic
