Abstract.Quantitation of Rh antibody with the Auto‐Analyzer is remarkably precise during short periods of replication. As the duration of operation extends over several hours considerable variation develops. This can be minimized with frequent calibrations. However, repeated intralaboratory tests on the same specimen over a period of several months show the range in values can vary from the mean by ±30%; with interlaboratory testing this may reach ±50%. Nevertheless, the AA technique does represent a distinct advance in the accuracy of Rh antibody quantitation when compared with established manual methods. With better control of the sources of variation such as drift in sensitivity and of cell base, and with extension of the accurate part of the calibration curve or limiting determinations to the most accurate portion, it should be possible to improve considerably the accuracy of the system. The correlations of prenatal serologic data determined quantitatively on the AA with severity of disease, were found, in these studies on a limited number of cases, not to be significantly better than those obtained with the semiquantitative manual methods employed. Plasmin agglutinators are natural IgG anti‐Fab antibodies. These antibodies bind to erythrocytes which are coated with the Fab fragments of anti‐Rh antibodies that have been hydrolyzed with plasmin. The plasmin agglutinators are demonstrable in most human sera albeit in low titer. These agglutinators can be inhibited with Fab fragments that have been hydrolyzed with subtilisin, bromelin, trypsin, and plasmin but cannot be inhibited by Fab fragments from pepsin, ficin, chymotrypsin, papain or elastase digests.The Fab fragments from naturally fragmented immune serum globulin gave identical inhibition reactions to those produced by plasmin hydroly

Abstract.A new method of C 3 type determination on high voltage agarose gel electrophoresis is described. The influence of some electrophoretic variables on the results has been investigated and a standard technique developed. The method is quick, simple and gives good separation of C 3 proteins. The three common C 3 phenotypes and the four variant types so far studied are easily demonstrated.Preliminary allele frequencies in a Norwegian population sample is presented. The values found,C 3S= 0.80 andC 3F= 0.19, are in good accordance with the allele frequencies in two population groups of North American Caucasians.

Abstract.The authors studied the degree of qualitative and quantitative deleucocytation of whole blood that can be obtained by washing and filtering. Based on the studies of 20 samples of blood it was established that successful deleucocytation can be expected only it combined procedures of washing and filtering are employed.The purpose of our present investigations was to determine to what extent can whole blood be deleucocyted by using current methods of blood preservation, or by combining these methods, as well as to assess the degree of damaging effects on red blood cells produced by these methods.

Abstract.ABO agglutinin titres and haemolysin tests on individuals living in urban areas of Northern England showed no significant variation. There was, however, a small but significant seasonal variation in individuals who lived in rural areas with outdoor occupations and it is suggested that this may be related to climatic factors. This might explain the variable results reported from different areas of the world.

Abstract.The incidence of H deficient A2blood samples in the Sheffield area was found to be 1 in 169 and 1 in 13 of group A2and A2B blood samples respectively. Family studies are described on an Aintand on examples of the new variant phenotypes, AintH↓., A2↑HW, A2↑BHWand A2BHW. The sera of some of the variants contained cold anti‐Hs/HIs/HIc/LebHantibodies, but none contained the Bombay type of anti‐Hsactive at 37°C. The authors suggest that the modified A status of most of the variants is due to reduced expression of theA1genes as a result of insufficient production of H precursor substance by weakHalleles.
