Abstract.Red cells stored in SAGM medium for 42 days at +4°C were rejuvenated by bicarbonate, pyruvate and adenosine. Autologous 24‐hour posttransfusion survival was determined in untreated as well as rejuvenated cells and showed an improvement from 77.4±4.7 to 89.2±7.2%. The erythrocyte adenylate energy charge decreased relatively more than the total adenylate concentration during storage, but the latter correlated better with posttransfusion red cell survival. Considerable deteriorations in red cell morphology (expressed as morphology index) and in deformability (measured as red cell fluidity) were observed during storage but were partly reversed by rejuvenation. The morphology index and the posttransfusion survival showed a significant correlation (r = 0.95, p<0.005) after, but not before, rejuvenation, indicating that the remaining changes are more permanent and decisive of survival. Abstract.We studied the storage characteristics of platelet concentrates prepared in polyolefin (PL‐732) and thin‐film polyvinyl chloride (CLX) plastic bags and stored on a newly designed flatbed reciprocal motion agitator with a 1.5 inch (3.8 cm) lateral movement and an oscillation frequency of 70 cycles/min. We also studied the ability of this device to resuspend the platelet button formed after preparative centrifugation. Results showed that platelet concentrates stored on the 1.5‐inch shaker had storage characteristics equivalent to those reported for the conventional 70 cycles/min, 1‐inch (2.5‐cm) lateral movement flatbed shaker. Due to a more rapid acceleration and deceleration rate, however, the 1.5‐inch shaker resuspended the platelet button formed after preparative centrifugation much more efficiently than did the less dynamic 1‐inch shaker. Abstract.Blood donated to the Natal Blood Transfusion Service was screened by an enzyme‐linked immunosorbent assay (ELISA) for anti‐lipopolysaccharide (anti‐LPS) anti‐bodies. Plasma units with high concentrations (>40 μg/ml) of anti‐LPS IgG were pooled and fractionated to obtain a gamma globulin (lot LG‐1). The binding of LG‐1 antibodies to LPS prepared from 14 bacterial species and strains was found to be the highest to LPS fromShigella flexneri, Salmonella abortus equiandSalmonella typhimuriumand intermediate withKlebsiella pneumoniae, Pseudomonas aeruginosa, Salmonella enteritidisandEscherichia coli026: B6. Differential absorption experiments showed that LG‐1 contained a mixture of specific and cross‐reacting antibodies. A large proportion of antibodies binding toSh. flexneriLPS were mainly specific, while those binding toS. typhimuriumand the otherSalmonellaspecies tested were largely cross‐reactive. There was little correlation between the spectrum of activity of the LG‐1 antibodies and the incidence of gram‐negative bacteria in blood cultures taken from hospital patients in an area covered by the Transfusion Service. Abstract.Interest in human plasma fibronectin (Fn) as a potential clinical product for replacement therapy in septic patients has prompted the search for stabilizers to protect the protein from heat denaturation during pasteurization designed to inactivate hepatitis viruses. Fn was pasteurized (60°C, 10 h) in the presence of either citrate, tricarballylate, sucrose or four mixtures of lysine, glucarate, gluconate or citrate which had been found to increase the denaturation temperature of Fn by 19°C. All but a citrate/gluconate mixture were effective in preventing aggregation as measured by dye fluorescence, light scattering, gel filtration and electrophoresis. Binding to gelatin was retained and immunological activity was only slightly diminished compared to a sample heated without stabilizers. Opsonic activity was measured as ability to mediate the uptake of125I‐gelatin‐coated polystyrene beads by attached human monocytes. Fn heated without stabilizers underwent a transient increase in activity which was traced to formation of aggregates having elevated specific activities. Pasteurized samples had slightly elevated opsonic activities with no detectable aggregates present, while the unstabilized control was inactive. These results indicate that the physical properties of Fn as well as the functional activities of the gelatin‐ and cell‐binding domains can be protected against thermal denaturation by various

Abstract.Monoclonal antibodies (OKT series) have been used to investigate possible modifications of T lymphocytes and T lymphocyte subsets in 65 multiply transfused β‐thalassemia patients. No significant difference was observed in percentage and absolute number of OKT3‐, OKT4‐, and OKT8‐positive cells when compared to controls. A subgroup of patients (10 patients, 15.3%), however, could be selected who showed a reversal of OKT4/OKT8 ratio. These patients did not differ from the others as to age, number of transfusions, frequency of splenectomy, ferritin levels, hepatitis B markers, chronic liver disease incidence, and numbers of B lymphocytes and natural killer cells. The features distinguishing this group from the remaining patients were: (1) decreased mitogen responsiveness; (2) early age when first transfused; (3) high incidence of males (90%). Immunological investigation was done in 2 occasions, 1 year apart, but no significant modification was observed in these patients. These findings suggest that in β‐thalassemia patients transfusion therapy started very early in life may be responsible for persistent immunological modifications.
