Abstract.A microwave oven has been used to obtain rapid and controlled thawing of frozen plasma packs for cryoprecipitate production. The resulting factor VIII yields were higher than the average of 77.8 IU (in a volume of 17.2 ml) obtained by the slow overnight thaw procedures in routine use. Assays on 10 individual packs showed mean results of 142 ± 24.5 IU factor VIII procoagulant activity, 188.6 ± 51.68 IU factor VIII‐related antigen, 0.152 ± 0.05 g fibrinogen and 218.9 ± 66.5 U fibronectin in a volume of 23.2 ± 6.6 ml. The results of this preliminary study indicate that microwave thawing of plasma is worthy of further investigation and should be reconsidered for routine production of cryoprecipitate and high yield fibro

Abstract.The metabolism of guanosine in human erythrocytes has been studied in two different experimental systems – direct incubation and dialysis incubation – the latter allowing continuous addition and removal of substances. Intra‐ and extracellular purine compounds were analyzed using high‐performance liquid chromatography (HPLC). At 37°C, a normal pH (7.4) and a favorably high concentration of inorganic phosphate, guanine nucleotides were synthesized at a substance rate of about 0.17 mmol · h‐1(calculated per liter erythrocytes) when guanosine was kept at a concentration of 25 μmol · I‐1. At a higher guanosine concentration the rate of synthesis increased only moderately. Erythrocytes loaded with guanylates lost these nucleotides at a rate of 0.023 mmol · h‐1at a normal phosphate concentration and somewhat slowlier at a higher phosphate concentration. Abstract.A protocol for large‐scale screening of blood donors for IgA deficiency using a tanned cell passive haemagglutination‐inhibition (PHI) technique and enzyme‐linked immunosorbent assay (ELISA) is described. Evidence presented suggests that native serum from a case of IgA paraproteinaemia, suitably diluted and heated to 56°C for 60 min, can act as a satisfactory and simple alternative to purified IgA preparations in the PHI test.When PHI and ELISA techniques were used in combination to screen 41,851 random donors, 58 were found to contain less than 200 ng/ml of IgA and were classified as totally IgA‐deficient (1:721). Abstract.23 bone marrow transplant recipients whose donors where major ABO‐incompatible received marrow depleted of red cells prior to infusion utilizing gravity sedimentation in hydroxyethyl starch. The in vitro red cell‐depletion procedure effectively removed 97.8% (mean) of the red cells from the harvested marrows and preserved 86.8% of the nucleated cells and 98.2% of the CFU‐C activity in 25.4% of the original volume. All recipients had a significant quantity of isohemagglutinins of both IgM and IgG classes demonstrable in their serum at the time of the marrow infusion. Patients were premedicated and well‐hydrated prior to the infusion and tolerated the infusion well. These patients demonstrated bone marrow engraftment at the same rate as did those patients whose marrow donors were either ABO‐identical or minor ABO‐incompatible. There was no difference in the incidence of or time to development of graft versus host disease, the incidence of graft rejection, or patient survival among the groups. Recipients of red cell‐depleted major ABO‐incompatible bone marrow transplants demonstrated production of donor‐type red cells somewhat later and required slightly more red cell transfusion support that did the other groups of recipients. Abstract.A patient with chronic cold agglutinin disease and anti‐I antibody was studied. The titer of the patient's serum at 4°C was 700 with adult I RBC, 256 with cord RBC, 256 with adult i RBC. At room temperature the titers were decreased and the serum also reacted at a titer of 4,000 with enzyme treated adult I RBC. Dithiothreitol treatment of the serum and purified antibody decreased reactivity only slightly, indicating that the antibody was IgG. The heat eluate contained 1.2 mg/ml IgG kappa, but only 80 μg/ml IgM kappa. A hybridoma made by fusing the patient's peripheral blood lymphocytes with a human myeloma cell line contained only IgM kappa (30 μg/ml) anti‐I activity. The IgM fraction of the heat eluate reacted similarly to the hybridoma supernatant. The IgG fraction resembled the serum and heat eluate. In this case study, IgG kappa and IgM kappa immunoglobulins, both possessing cold agglutinin activity, were present in the patient's serum. Abstract.The class III gene markers, C4A, C4B, C2 and BF were determined in 26 HLA genotyped families. In 6 of these families the inheritance of the C4A electrophoretic phenotype, C4A 3, A 2 could not be explained assuming the genetic model that the C4A component is controlled by only one locus(C4A)and suggests that both C4A 3 and C4A 2 are encoded by the same chromosome. This apparently duplicatedC4Alocus haplotype is common in Northern Europeans occurring at a frequency of at least 6%. Abstract.Monoclonal antibodies (McAb) specific for human IgA, the IgA1 and IgA2 subclasses and the iso‐allotypic epitope nA2m(2) have been produced. Three distinct McAb recognise an IgA1‐specific epitope expressed in the Cα2 domain or the hinge region whilst a further McAb is directed possibly to an IgA1 hinge region epitope. The McAb having nA2m(2) specificity recognises an iso‐allotypic epitope expressed within the Fab region. IgA1 and IgA2 epitopes were detected in gorilla but not rhesus or baboon sera suggetting that the IgA subclasses represent a recent gene duplication event.
