Abstract.Plasma samples from haemodialysis patients, renal allograft recipients and haemophiliacs were studied for the presence of mixed leukocyte culture blocking factor activity (BFA). A history of blood transfusions received within the preceding 6 months of testing was significantly and directly correlated with the appearance and persistence of BFA in the plasma of haemodialysis patients. This association was not found in renal allograft recipients. The development of BFA was found to be associated with formation of lymphocytotoxins but not haemagglutinins. The authors have also demonstrated that BFA resides in the IgG fractions of serum samples obtained from both chronic uraemics undergoing haemodialysis and patients bearing renal allografts. BFA may play a minor role in the acceptance of human renal allografts.

Abstract.We have developed a new serologic method for the study of lymphocytotoxic reactions, based on differential fluorescent staining. Living cells, labeled with fluorescein diacetate, show green fluorescence, while dead cells are stained by ethidium bromide and show red fluorescence. Since both living and dead cells can be quantitated, the test is more sensitive. Because the total number of cells present can be quantitated, the estimations can be made by automatic equipment without the danger of false negatives resulting from the absence of cells.

Abstract.A simple method is described for the preparation of leukocyte‐poor red cell concentrates from fresh and stored CPD blood. Warm normal CPD blood is passed through a nylon wool filter. After centrifugation at 1,500gfor 3 min, the plasma, the buffy coat and the top 30 ml of the red cell concentrate are eliminated. The average removal of leukocytes is better than 97%, the relation of remaining lymphocytes and granulocytes being 1:1. Red cell loss ranges around 20%. Abstract.Periodate oxidation and Smith degradation of desialized human erythrocyte glycoproteins lead to an almost complete destruction of galactose and some loss of galactosamine and glucosamine. Neutral sugar and hexosamine content of native MN substances are only slightly influenced by periodate oxidation. Destruction of galactose is accompanied by an uncovering of A‐like receptor sites detectable by agglutinins from plants and snails, which are directed against terminalN‐acetylgalactosamine. From the specificity of these antibody‐like substances, it is deduced that the amino sugar functioning as receptor is α‐glycosidically linked. Destruction of the receptor sites by subsequent alkaline borohydride treatment suggests that theN‐acetylgalactosamine is unmasked by degradation of the alkali‐labile tetrasaccharide of human erythrocyte glycoproteins. Periodate‐oxidized and Smith‐degraded NANA‐free erythrocyte and ovarian cyst glycoproteins treated by acetolysis are potent inhibitors of anti‐Tn from man, animals, and plants, suggesting that Tn‐antigen is represented by terminal, α‐glycosidic, peptide‐boundN‐acetylgalactosamine. Abstract.Saline extracts from the eggs of the snailPomacea paludosacontain two separate and potent agglutinins specific for determinants on human ABO erythrocytes. Haemagglutinins with anti‐pneumococcus type XIV and cross‐reacting A and B specificities were detected in three different extracts from eggs of this species. Human group O erythrocyte‐agglutinating activity ofPomacea urceusalbumin gland extract was inhibited by purified hog H substance and pneumococcus type XIV polysaccharide, but purifiedSassafras albidumpolysaccharide was the most potent inhibitor of this reagent. Abstract.Sera obtained from 15,500 Rh‐negative pregnant women were screened for presence of serum IgA. 12 out of 15,500 or 0.08% of the women were deficient in serum IgA, and only one of these 12 women had anti‐IgA antibody of broad specificity. Nine women with IgA deficiency received anti‐Rhoγ‐globulin within 72 h after delivery. None of them developed anti‐IgA antibodies 3–6 months following injection of anti‐Rhoγ‐globulin. Administration of Rh immune globulin to one subject (case No. 5) in this group who has class‐specific anti‐IgA was not associated with any untoward reactions, and there was no significant change
