摘要:
AbstractWe describe a rapid automated technique for colorimetric in situ DNA hybridization using the Code-On Immuno-DNA Slide Stainer. We report recent advances in positively charged glass slide technology, rapid limolene/xylene dewaxing, stable ribonuclease and pepsin predigestion, heat cycling during hybridization, and alkaline phosphatase enhancement. These amplify the signal to noise ratio of colorimetric molecular hybridization to the point where complete one hour assays can be performed from dewaxing to counterstaining by automation. Both biotin labeled oligonucleotide and nick translated DNA probes can localize their viral targets in situ with this single, mechanized protocol at the rate of one slide per minute. 摘要:
AbstractA rapid, automated immunocytochemical staining method was developed for detection of lymphocyte surface markers with the Code-On Immuno/DNA Slide Stainer. Acetone fixed, frozen tissue sections from snap frozen lymph nodes and tonsils were stained for various B andT cell markers by the immunoperoxidase technique with commercially available antibodies and the Vectastain Elite kit. This system produced high quality, specific staining with substantial time and cost savings over manual processing. (The J Historechnol14:231, 1991)

摘要:
AbstractProliferating cell nuclear antigen (PCNA), an endogenous cell replication marker, was detected by an improved immunohistochemical procedure (super sensitive biotin-streptavidin), in formalin fixed, paraffin embedded tissue. Small intestine, liver, kidney, testis, and esophagus from rats were stained for the detection and evaluation of PCNA as a suitable marker for cell proliferation. Different patterns of staining that are believed to correlate with the individual phases of the cell cycle (G1, S, G2, M, G0) were recognized. PCNA offers an alternative to established cell labeling techniques for investigating cell proliferation. (The J Histotechnol14:237, 1991)

摘要:
AbstractAn irnmunogold-silver stainingmethod wasused to identifyTreponemapallidumspirochetes in formalin fixed tissue. In an indirect immunohistochemical procedure, the colloidal gold particles bound to the secondary antibody were visualized by a silver developing solution to create a brown-black reaction product. This brown-black staining is similar to the stain generated with a Warthin-Starry technique. This method was superior to such techniques as the avidin-biotin complex immunoperoxidase method and indirect immunofluorescence method with ammonium hydroxide treatment and could also beused to stain other spirochetes and antigens that are difficult to visualize with other immunohistochemical techniques. AbstractA modified and simplified luxol-fast stain was used on 345 histologic sections of 23 canine stomachs to visualize parietal cells selectively. This staining procedure allowed their discrimination from other mucosal components and their quantification by computer-assisted histomorphometry. AbstractWe describe several methods for the handling, fixation, and processing of canine eyes, as well as the light microscopic results of these efforts. Fixatives included Zenker and Bouin, 4% formaldehyde-1% glutaraldehyde, and 2% and 6% solutions of glutaraldehyde. In some techniques the globe was injected with fixative; in others, a window was cut into the eye or ocular tissues were dissected and separated to facilitate fixation. Processing included ethyl or isopropyl alcohol dehydration; all eyes were embedded in paraffin. Best overall morphology was seen in specimens fixed in Zenker fluid for 5 to 8 hr and dehydrated initially in isopropyl alcohol. There was superior retinal detail with acceptable morphology of the cornea, iris and lens. Advantages and disadvantages of various techniques and common artifacts are discussed. 摘要:
AbstractBromcresol purple (BCP) dye has been used as a colored marker and pH indicator for 10% formaldehyde solution. Its distinctive coloring has advantages related to lab or hospital safety and tissue processing. The effects of the dye on tissue processing and staining were tested by routineuse for over 2 yr in a busy pathology laboratory, by fixative penetration (K-value) determinations on a protein model, and by pH indicator trials. No adverse effects were noted in these trials. Although BCP was reported to bind specifically with albumin, results of trials suggest that this property did not confer any advantage to rate of fixation nor did it affect routine staining. BCP is recommended as an additive to 10% neutral buffered formalin as a pH indicator for acidic conditions during fixation or storage that might produce artifacts. In addition, it serves as a marker for this toxic chemical. (The J Histotechnol14:257, 1991)
