AbstractFour commercially available immunocytochemistry kits were compared for their ability to detect human cytomegalovirus (HCMV) in tissue culture and histologic sections using HCMV specific murine monoclonal antibodies. The procedures evaluated included two avidin-biotin complex (ABC) kits with either alkaline phosphatase or peroxidase coupled to biotin, an indirect immunogold silver enhancement (IGSS) kit using goat-antimouse antibody-5 nm gold sol, and a streptavidin- 5 nm gold sol kit. For HCMV detection in tissue culture, the ABC-alkaline phosphatase kit was superior to the ABCperoxidase and indirect IGSS kits. The streptavidin IGSS kit was also superior to the indirect IGSS kit in its ability to detect HCMV in tissue culture. Of the three silver enhancement techniques, the IntenSE I1 provided the best balance between ease of administration and intensity of staining. The streptavidin IGSS kit performed well in demonstrating HCMV-infected cells in histologic sections. These studies demonstrate that streptavidin IGSS is a useful immunocytochemical procedure that can be used for studies involving HCMV diagnoses, and HCMV pathogenetic studies. AbstractThe effects of different fixatives on the immunohistochemical staining of lymphoid surface antigens in paraffin embedded sections were studied using avidin-biotin-peroxidase complex (ABC) technique. We found that the best results were achieved in B-5 fixed tissues. The demonstration of T and B cell antigens in formalin fixed tissues gave satisfactory results and did not require trypsinization, with the exception of leucocyte common antigen and MTl. Fixatives such as Bouin's fluid and acid formalin showed good preservation of the antigenicity of only B cells, and the use of trypsin digestion did not improve the results; on the contrary, it abolished the immunostaining. AbstractThe spatial distribution of biologically relevant elements within bone matrix was investigated using electron probe X-ray microanalysis of frozen-dried bulk specimens of cortical bone tissue. Compositional images of calcium, phosphorus, sulfur, and chlorine are presented along with the corresponding secondary electron images: These data indicate that calcium and phosphorus X-rays detected from regions of bone matrix vary little in X-ray intensity as a function of location within the matrix. Most of the sulfur and chlorine X-rays detected originate in regions corresponding to the marrow cavity or to surrounding connective tissue. However, higher magnification X-ray maps reveal the presence of both elements within bone matrix with localizations occurring in regions corresponding to lacunae and surrounding bone matrix. AbstractWhen the anionic dye acid red 137 (CI 17755) was applied to B-5 formalin fixed smears or tissues after clearing of the specimen in Lugol's solution and sodium thiosulfate, granules of eosinophils stained bright red orange. Most other cells stained pale cream color or not at all. Used as a selective stain for eosinophils, the stain holds promise as a means for quantitating eosinophils in urinary sediments, lymph node and bone marrow biopsies, as well as tissues such as liver, lung, gallbladder, and urinary bladder. AbstractThis paper describes the modification of the Lieb erichrome technique for staining myelin sheaths' embedded in glycol methacrylate plastic. The technique permits excellent morphologic detail of myelinated and demyelinated axons in the central nervous system and the characterization of pathologic abnormalities in the spinal cord of mice persistently infected with a demyelinating strain of Theiler's virus. It is a simple, rapid technique that allows 25–30 spinal cord cross-sections to be embedded, sectioned and stained at one time. (The J Histotechnol12:35, 1989).

AbstractPneumocystis carinii (P. carinii), a microorganism often associated with AIDS patients, stains a brilliant rose-purple with cresyl echt violet. The original procedure used solutions that were unstable, giving irreproducible results. This modification produces a short reliable method. 摘要:
AbstractCalcofluor white, a chemical substance used as a brightening agent in the fabric and paper industries, was the initial substance of this class to be employed in histology. This agent induces certain microorganisms to fluoresce in ultraviolet light, materially aiding in their tissue identification. Other fabric brighteners, identified as Phorwite compounds, have recently become available for potential use as fluorochromes in histology and histopathology. These have been tested in our laboratories on animal tissues and the results compared. Of ten such substances studied, five were found to possess real ability to delineate cellular morphology, especially the fine distribution of capillaries and red cells within parenchymatous organs, clearly establishing the presence of congestion and petechial hemorrhage. AbstractA method combining the acetylcholinesterase (AChE) staining technique and a thick section method is presented for studying the topographical anatomical structures of freshly frozen specimens containing bone. Fresh frozen monkey orbits embedded in an 8% carboxymethylcellulose (CMC) solution were sawn serially into 2 mm thick sections. The sections were fixed in 4% sucrose form01 solution and rinsed in distilled water. After AChE incubation, autonomic nerves were stained intensely and cranial nerves were stained weakly. The sections were stored in petri dishes or in polymethylmethacrylate (PMMA) chambers filled with pure glycerol.This method allowed the examination of the topography of autonomic nerves and their target organs, smooth muscle fibers of the orbital and the inferior and superior tarsal muscle, the lacrimal gland, and the ophthalmic artery. Small branches of autonomic nerves could be traced up to microscopical level. (The J histotechnol12:47, 1989).

AbstractThis paper describes the detailed salvage technique used for research specimens that had been infiltrated and embedded in glycol methacrylate (GMA). During the second infiltration step the GMA spontaneously polymerized,“trapping”the tissue cassettes in a“mass”of prematurely polymerized plastic.
