AbstractGlycolmethacrylate (GMA) is now a widely accepted plastic embedding medium for histopathological uses although technical conditions of polymerization are not fully understood. We found that benzoyl peroxide initiated GMA medium accelerated with dimethylaniline (NN-DMA) polymerizes at different rates according to the volume left to polymerize and the size of the embedding molds. This phenomenom appears to be sustained by an unexpected and complex thermodynamical process that we were unable to resolve. However, we did find that NN-DMA concentration is a polynomial function of the initial GMA volume, and the proposed equation may help in adjusting the techniques when varying the amount of methacrylate embedding medium or when changing the size of the embedding molds. AbstractMicrowave technology has been used in numerous areas of pathology and research. Applications have been developed for special and routine stains and a limited number for immunostaining. In this report, microwave enhanced immunohistochemical staining in the standard avidin-biotin methodology is demonstrated for fourteen diverse antigens. Staining of paraffin or frozen sections was achieved for actin, desmin, GFAP, HCG, IgA, IgG, IgM, insulin, kappa and lambda light chains, myoglobin, prostatic acid phosphatase, S-100 and vimentin. No single procedural step exceeded 5 minutes, and some incubations were reduced to as little as 10 seconds. The entire immunohistochemical staining procedure can be completed in 10–30 minutes with results that parallel conventional methods. (The J of Histotechnol12:95, 1989)

AbstractThe objective of this research was to develop a rapid morphological test for proliferating cells using diaminobenzidine as the substrate marker. This technique can be employed to visualize proliferating cells by using a monoclonal antibody to bromodeoxyuridine and a standard immunoperoxidase technique. This paper describes methods for the rapid demonstration of S-phase nuclei in routine paraffin tissue sections. The tissue was fixed by exposure to microwave irradiation for various times and temperatures. The subsequent staining results are reliable and can easily fit into an existing immunoperoxidase program. (The J Histotechnol12:101, 1989.)

摘要:
AbstractA rapid silver method to demonstrate glomerular basement membranes is described. The method uses the dilute ammoniacal silver solution as described by Churukianet al.for the demonstration of fungus andPneumocystis carinii. The ammoniacal silver solution replaces the silver methenamine solution in the modified Jones' method for basement membranes. The heat needed to impregnate the basement membranes with silver is provided by the microwave oven, thus decreasing the total staining time. The staining results are consistent, and each glomerulus throughout the biopsy is evenly stained with no background silver debris. Renal biopsies fixed in both Helly's and 10% buffered formalin are stained equally well. (The J Histotechnol12:105, 1989.)

AbstractCells have been found to contain tissue specific intermediate filaments (7–10nm). Exact determination of cellular origin can be made by using antibodies against intermediate filaments. Epithelial cells are rich in intermediate filaments of the keratin type. Using this knowledge and a polyclonal antibody to keratin filaments, individual cells from menstrual blood flow, a mixed cellular population, can be localized and specifically identified.Cells cultured from menstrual blood flow were fixed in cold methanol and stained with anti-keratin polyclonal antibody using immunoperoxidase techniques. Positively stained cells, indicating endometrial origin, revealed a diffuse filamentous network throughout the cytoplasm. Cells rounded during trypsinization, resulting in the alteration of the pattern of keratin filaments. In addition to diffuse staining throughout the cytoplasm, a dense droplet was seen juxta nuclear. By using immunogold staining techniques, localization of the keratin filaments before and after trypsinization was studied. (The J Histotechnol12:109, 1989.)

AbstractLectin cytochemistry was used to detect viral specific proteins in infected bovine turbinate cells as a means of diagnosing the presence of various virus. Six lectins having different properties were evaluated for binding to bovine respiratory syncytial virus (BRSV), bovine viral diarrhea (BVD), bovine herpesvirus-I (BHV-l), and bovine herpesvirus-II (BHV-2). BRSV, BHV-1, and BHV-2 were detected. Three lectins,Phaseolus limensis, Phaseolus vulgaris, andGlycine maxcould not detect any virus, Three lectins that could detect one or more virus includedVicia villosa B4, Helix pomatia, andHelix aspersaLectins as a detection tool have a considerable level of specificity for binding to virus associated proteins. This work shows that this characteristic and others allow lectins to be a powerful means to diagnose viral infection. (The J Histotechnol12:115, 1989).

AbstractDifferences among the useful lifetimes of stains are well known to histotechnologists. Though problems of fading have been studied extensively by chemists, these data received little attention in histotechnology. Yet, such information is invaluable for the development of new reactions and modifications of existing technics to improve fastness properties. The history of light fastness determinations, current fastness ratings, light absorption by dyes, and fluorochromes were reviewed in part I of this report. This discussion will deal with relations between structure and fastness properties of dyes, the importance of molecular orbital systems, effects of metals, substrates, moisture, and of various reagents on the fastness, i.e. permanence of stains. AbstractTwo cases of idiopathic amyloidosis are described. AA protein was found to be the major constituent of tissue amyloid in both, based on sensitivity of Congo red birefringence to pretreatment of slides with potassium permanganate and immunoperoxidase staining with a monospecific antiserum. However, an underlying inflammatory, infectious or neoplastic disorder was not identified. The occurrence of AA amyloidosis without clearcut underlying disease is rare, having been described in only thirteen previous instances. Recent clinical series suggest that such cases may have a different course and response to therapy from“primary”amyloidosis due to systemic light chain deposition. Such cases may also underscore the importance of genetic factors in the pathogenesis of AA amyloid and the importance of characterizing the amyloidoses biochemically, as well as clinically.
