AbstractConnective tissue and mucosal mast cells differ in morphology, chemistry, and location and require different histologic techniques for demonstration. Connective tissue mast cells are readily preserved by most common laboratory fixatives and stained with toluidine blue. Mucosal mast cells can not be demonstrated after the above treatment but are visible after Carnoy fixation and astra blue staining.

AbstractThe meridional portion of a fetal canid eye at gestation day 55 was embedded in methyl methacrylate. Full sagittal sections, from 1 to 4 microns, passing through pupil and optic papilla or through peripapillary zones of dysplastic retina, were obtained using a Leitz base-sledge microtome with a profile d knife.Sections were floated onto glass slides with 50% ethanol, flattened with 95% ethanol, and dried on a 45' C warming plate. Plastic was removed with Cellosolve acetate, permitting a variety of routine and histochemical staining procedures.

AbstractThe histologic requirements for preservation of lymphocyte membrane antigens were discussed by the participants of a conference on leukemia and lymphoma. The risk of false negative staining is minimized if surgical lymph node biopsy specimens are processed promptly into OCT and quick-frozen with liquid nitrogen. Cut sections must be adequately dried and fixed in“dry”acetone.

AbstractOn standing, hematoxylin converts to its oxidation product, hematein. Hematein produces a dye lake when it is combined with a mordant, ferric alum in this procedure. Myelin has a special affinity for a basic charged lake as it is a complex material containing cholesterol, phospholipids, cerebrosides, and other components. A dilution of a 1 % solution of hematoxylin in absolute alcohol combined with an equal volume of 1% aqueous ferric alum provides sufficient staining to demonstrate the white fatlike substances forming a sheath around peripheral nerve fibers.

AbstractA fifty-five year old male with an appendicular abscess had 100 ml of pus drained from the right lower quadrant. Two months later, an interval appendectomy was performed. The macroscopically normal appendix had a portion of fibroadipose mesoappendix attached. Microscopic evaluation using routine stains showed redundance and diverticular extrusion as well as foci of ectopic gastric glands. Immunohistological studies for detection of gastrin immunoreactivity revealed gastrin containing cells in foci of ectopicgastric glands. This represents the first report in which gastrin was demonstrated in an appendix, confirming the presence of heterotopic gastric mucosa.
