摘要:
AbstractA modified Avidin-Biotin-Peroxidase complex (ABC) technique is described for use on frozen tissue sections and compared with the PAP technique. Increasing the incubation temperature and decreasing the incubation time resulted in (1) increased sensitivity with less background staining, (2) reduced turnaround time and (3) decreased cost with the modified ABC technique.

摘要:
AbstractThree variants of Weigert's resorcin fuchsin stain for elastic tissue were compared with the original. The resulting slides were virtually identical. Thus, the claim for specificity for one variant1was not confirmed. Since preparation of the dye solution is time consuming and messy, it is suggested that the use of the Chroma preparation is preferable.2However the recommended staining time can be reduced to 30 minutes and, after staining, the slides should be differentiated for one minute in 1 % HCI in 70% ethanol.

摘要:
AbstractA modified stain for general routine cytology use is presented. This stain is basically that of Papanicolaou, but prepared as a single prebuffered powder in 95% ethanol to make one staining solution. The tinctorial patterns are similar to its predecessor—thus cell identification presents little difficulty. The staining solution is relatively stable and repeated use does not appreciably diminish its potency. Barr bodies stain exceedingly well, eliminating special procedures for these structures.

摘要:
AbstractThe hairless mouse, whose skin in many ways resembles that of man, is increasing its appearance in the scientific literature, in a variety of fields from dermatology to immunology and cancer research. Due to the fragility of the skin of these animals, special handling is often required to obtain optimal histological results. In processing, embedding and staining, special care must be taken to ensure reproducible slides. Many techniques and stains applicable to human skin are useful on mouse skin; however, differences in skin structure, strength, and in apparent chemical specificities have shown that the methods which are most successful on human tissues are not always the best choices for use on hairless mouse skin. Necessary modifications of standard methods for satisfactory histological processing of whole skin are detailed, in addition to a description of staining techniques found most successful for use on hairless mouse skin.
