AbstractAn automated method for lectin staining using the Fisher Code-On™series, with a modification of its general program,“Immunocode,”is described. The lectin wheat germ agglutinin (WGA) was used for staining procedures. Comparisons were made between the lectin staining quality of formalin- vs modified B5-fixed lung tissue, using manual and automated staining techniques. Although automated and manual methods for lectin staining were comparable, the results obtained by automated staining were highly reproducible and less subject to inconsistencies due to human error. Automated staining also decreased consumption of costly reagents and saved laboratory personnel time (~3.0 hr, automated vs 4.8 hr, manually). The intensity and discreteness of WGA staining was superior in the modified B5- vs formalin-fixed lung tissues with both automated and manual staining methods. (The J Histotechnol14:149, 1991)

摘要:
AbstractOne of the most popular and reliable histochemical techniques to demonstrate carbohydrates and mucosubstances in tissues is the periodic acid-Schiff (PAS) technique. In the course of this technique, the aldehyde group produced in the section by periodic acid oxidation reacts with the Schiff reagent to form a magenta colored final product. All such substances that take up magenta color are known as PAS-positive substances. The new histochemical technique described in this paper demonstrates all PAS-positive substances without using the Schiff reagent. The technique uses a nondecolorized dilute solution of basic fuchsin in combination with a dilute solution of periodic acid to form a crimson final reaction product with all types of PAS-positive substances. The new technique, which is economical and simple to carry out, requires a shorter time and can be performed on formalin fixed, paraffin processed tissues. (The J Histotechnol14:155, 1991)

AbstractA modified Mayer hematoxylin stain that can be combined with either phloxinelsaffron or eosin for general staining 01 as a counterstain is presented. This method uses 10% less alum, and chloral hydrate is replaced by absolute ethanol, a more effective penetrator and bacteriostat. Phenol is recommended for formalin pigment removal in formaldehyde fixed tissues to effect a more esthetically pleasing section with most techniques. Saffron bought through the health food stores is 8 times less expensive than that bought commercially. (The J Histotechnol14:163, 1991)

AbstractA rapid method for macroscopic and microscopic investigation of human CNS is proposed. After formalin fixation, gelatin or agarose embedding, and cryoprotective treatment, frozen human spinal cords, brainstems, or hemispheres can be serially cut into 0.7 mm thick slices. Stained with gallocyanin- chromalum, these slices facilitate cytoarchitectonic, neuropathologic, and quantitative examination. Regions of interest from parallel formalin-stored unstained slices can be embedded into paraffin and stained by any immunocytologic and histologic stain compatible with formalin fixation and paraffin embedding. 摘要:
AbstractFrog sciatic nerves containing Teflon-coated platinumiridium wire implanted in vivo were embedded in glycol methacrylate (GMA) and microscopically examined for tissue-metal biocompatibility. Nerves containing metal implants were serially sectioned at 2-8μm on a retracting, automated microtome with triangular glass knives and then stained. Stained sections were examined for demyelination, axon destruction, scar tissue and general tissue response to surgical and implantation procedures. GMA provided necessary support to section tissue containing metal that could not be furnished by paraffin. It also provided versatility in staining that allowed flexibility in specimen analysis. 摘要:
AbstractIntraoperative identification of Pneumocystic carinii generally involves the use of special stains, which are time consuming and require expertise. The hematoxylin-eosin (H&E) stain readily demonstrates the organism in smears but not in tissue sections. The method described affords identification of these organisms within 5-10 min. A smear obtained from bisected lung biopsy tissue is immediately fixed with cytosol spray, dried for a few sec, and stained with H&E. When slides are examined with either high dry or oil immersion objectives, the smear shows foamy amorphous clusters of organisms, macrophages, and epithelial cells. The method stains both the cysts and the trophozoites, and the results correlate with those from special stains on tissue sections. It is rapid, simple to perform, low in cost, and easy to interpret. AbstractThe storage of glycosaminoglycans (GAGs) within cells of the central nervous system of dogs with mucopolysaccharidosis I (MPS I) was studied. Lectins that detect these stored GAGs were applied to formalin fixed tissues from 5 MPS I-affected dogs, 2 of which were previously treated by littermate bone marrow transplantation (BMT), and from 2 unaffected littermate controls.In all transplant recipients there was reduced reactivity of the central nervous system (CNS) neurons with soybean agglutinin and horse gram agglutinin indicating that BMT results in reduced levels of those GAGs stored excessively in MPS I. Wheat germ agglutinin was found to react less strongly with cells from treated or untreated dogs despite its predicted binding to sugars in GAGs that are known to accumulate in MPS I. These findings, coupled with previously reported total brain GAG content, show that neuronal storage of GAG is reduced following BMT in canine MPS I. (The J Histotechnol14:183, 1991)

AbstractSeveral recent studies suggest a diagnostic and a possible prognostic role for silver stained nucleolar organizer region (AgNOR) determination in the assessment of human neoplasia. Technical pitfalls, especially silver grain precipitate, may hamper accurate counting of AgNORs. We have identified pH as a main factor causing silver precipitation and overstaining. An improved silver staining method, which raises the pH to 3.0, optimizes visualization by eliminating silver precipitation and improving contrast between AgNORs and surrounding tissue components.
