摘要:
AbstractA possible and important sequalae of lung injury is the development of pulmonary fibrosis; however, conventional embedding procedures which use thick paraffm sections may not afford the pathologist the ability to identify an early fibrogenic response. Methacrylate-embedded tissue allows the histologist to section lungs at 1.0 micrometers, increasing the resolution for the identification of early histopathological changes. This latter method, in combination with a modification of a published trichrome staining method used to emphasize the pulmonary fibrotic response to an array of xenobiotics, is presented in this paper.

摘要:
AbstractA method is described for obtaining thin, undecalcified, ground sections of bone. The procedure consisted of grinding gradually a plastic embedded 150uM bone section to 10uM thick using the Maruto Speed Lapping Machine ML-150 Dc. Sections prepared in this manner were suitable either for quantitative or qualitative bone histomorphometry.

AbstractFor maximal color contrast with the dark green copper salt formed in the Uzman reaction, a light red counterstain is required. Nuclear fast red, which stains nuclei light red and cytoplasm a faint pink, gives excellent color and tonal contrasts. This results in high visibility of copper salts and delicately stained tissues, allowing the viewer both to identify tissues and to appreciate the location of the copper deposits. Nuclear fast red, eosin, methylene blue, metanil yellow, picric acid and Harris' hematoxylin were evaluated as counterstains. Nuclear fast red was the counterstain of choice for visual observation, color photomicrography and monochrome photomicrography. Counterstains of eosin and methylene blue were adequate; metanil yellow and picric acid were barely adequate. Hematoxylin and no counterstain were unacceptable for monochrome photomicrography. Nuclear fast red counterstained Uzman sections required a Wratten #11, #59 or #58 filter in monochrome photomicrography, the exact filter depending on the density of the counterstain. Lightly nuclear fast red counterstained tissues required the use of a #58 filter, the more densely counterstained sections, a #59 or #11 filter.

摘要:
AbstractA method is described for distinguishing type I from type II muscle fibers and visualizing their motor endplates. Muscle biopsies from the region of the neuromuscular junction are snap frozen and sectioned on a cryostat. The sections are first incubated in a substrate containing beta nicotinamide adenine dinucleotide, reduced form, and nitroblue tetrazolium, then in a substrate containing alpha naphthyl acetate and hexazotized new feuchsin. This procedure takes only thirty-five minutes to do, no cholinesterase inhibitors are used, and both fiber type and neuromuscular junction are demonstrated simultaneously. It can be used routinely to help diagnose neuromuscular disorders and for myoneural research, in general.

摘要:
AbstractStudents and faculty affiliated with the Program in Histotechnology at the University of Texas School of Allied Health Sciences felt the need for an assessment of the attitudes of histologic technologists-technicians toward the newly established HTL registry. It was determined that a nationwide survey of Schools of Histotechnology could be used to accomplish this objective.The results of our survey showed that a majority of the respondents have yet to appreciate-the value of education in histotechnology. We submit that advancement in the field of histotechnology is intimately associated with advancements in histotechnology education and research. We concluded that on the basis of our survey, further research could be done in this area to identify and stimulate those ideas which would best benefit the profession of histotechnology.

AbstractAn eighty-seven year old female presented with complaints of pain in the left eye. She had a history of chronic open angle glaucoma that was under control with medication. She had lost all vision, and the eye had become very painful and inflamed. She was admitted with a diagnosis of endopthalmitis. Enucleation was performed and inspection of the specimen revealed a firm globe with a white mass extending over the iris and across the pupil in the anterior chamber. Microscopic evaluation with the hematoxylin and eosin stain revealed a plasma cell infiltrate involving the ciliary process, extending into the posterior chamber, occlusion of the retinal artery, and extensive loss of nerve fibers within the optic nerve. An accumulation of a pinkish and somewhat fibrillar material within the tumor and optic nerve was also observed. Stains for myelin and amyloid and a methyl green pyronine were performed, and immunoperoxidase for IgG, IgA, IgM, kappa and lambda.

AbstractThe two commonly used methods of adhering tissue sections to slides are gelatin to the warm waterbath before cutting is started or smearing egg albumin onto the individual slide. Problems with both methods have led many histotechnologists to seek alternative ways of adhering tissue sections to the slides. Elmer's Glue, which we had used as an adhesive, was found to candida Sp. Non-human organism. It is effective if not used in staining fungi.
