AbstractA method facilitating recording of macroscopic images from glycolmethacrylate (GMA) embedded tissues and tissue-free sections is described. This method used dyes that selectively stain only tissue, only resin, or both, but in contrasting colors. The dyes were selected on the basis of simple numerical parameters like ionic weight, charge, and log Poct.Contrasting colors were readily converted into contrasting gray values and were so recorded for morphometric investigations, permitting areas of tissue and overall areas of resin sections to be easily measured with animage analysis system. The time saving compared to manual procedures was substantial, about 50%.We found that the small hydrophilic dyes (<550 Da) quickly stained tissues and could readily be removed from the GMA matrix by water washes. AbstractWe describe the method and application of a reliable thionin Feulgen stain for paraffin sections of formalin fixed, routinely processed surgical pathology specimens. The technique substitutes thionin for basic fuchsin in the classical Schiff reagent and modifies the hydrolysis times. Serial sections cut from selected paraffin blocks were processed with both a commercial kit stain and the thionin Feulgen stain. Computer assisted image analysis was used to perform a direct comparison of tissue response to the two stains. The thionin Feulgen reagent yielded results as reliable as those from the commercial kit stain, at approximately one-fifth the cost. (The J Histotechnol16:113, 1993)

AbstractA method is described for making histological sections of tumor pellets prepared from very small particulate samples of breast cancer originally pulverized for biochemical analysis of estrogen and progesterone receptors. Pulverizing the frozen tumor fractures it into fine particles with remarkably intact microscopic anatomy. This method embeds and concentrates the particles in matrices suitable for preparation of either frozen or permanent histological sections. We have used these tumor pellets and standard immunohistochemical procedures to study multiple biologically and prognostically important markers on single 50 mg samples of breast cancer from a large frozen tumor bank with long-term clinical follow-up. AbstractA simple method of labeling 3 antigenic sites on 1 tissue section with only the conventional immunoperoxidase method is described. Although procedures to label 3 or more antigens simultaneously have been reported previously, they have used combinations of fluorescence, immunoperoxidase, and alkaline phosphatase to produce chromagens of different colors. The triple-label imrnunoperoxidase procedure described is both cost- and time-effective, yields permanent results, and offers distinct advantages over other staining procedures. AbstractAdjacent semithin and ultrathin sections were used to correlate specifically immunolabeled cells at the light microscope level with the superimposed subcellular compartments at the electron microscope level. Prolactin immunoreactivity was successfully located in theparaformaldehyde-lysine-periodate (PLP)-osmium fixed, plastic embedded rat anterior pituitary glands at both the light and electron microscope levels. Immunofluorescence revealed slightly heavier immunoreactive product in the unosmicated tissues than in the osmicated. By superimposing semithin section and consecutive ultrathin section, the brightly immunofluorescent areas appeared to be secretory granules. This method may be valuable for identifying the fine structure of specific cells immunolabeled by various hormonal antigens, protein markers, and putative neurotransmitters other than in the pituitary gland. AbstractAs the numbers and diversity of orthopedic prosthetic devices increase and the service life of implants nears expiration, surgical pathology laboratories can expect to receive specimens containing softer polymeric materials such as the expanded polytetrafluoroethylene (ePTFE) comprising the Gore-Tex Cruciate Ligament Prosthesis. This report suggests a systematic approach to the processing of a surgically resected bone specimen that may contain such biomaterial. Suggestions for specimen documentation include photography specimen radiography. Protocols for decalcified paraffin processing and for undecalcified polymer processing in glycolmethacrylate and the Exakt grinding system are reviewed with consideration for the needs of both the clinical and research aspects of specimen management. In this procedure, resorcin-fuchsin staining is followed by Gill hematoxylin and eosin Y-rose bengal. AbstractQuantitation of mast cells and eosinophils is of value in studies of allergic or parasitic tissue inflammation. We describe a new method that combines two reliable stains, astra blue and vital new red, to stain both mast cells and eosinophils in the same tissue section. The mast cells stain violet-blue and the eosinophils bright red. Because this method stains both cells in tissue that has been fixed with form01 saline or buffered formaldehyde, it can be used on archival tissue that has undergone standard fixation. The minimal staining of the background makes quantitative studies of both cells simple, even at low magnification. (The J Histotechnol16:143, 1993)
