AbstractHigh-resolution immunolight microscopy was carried out on a series of neoplasms embedded in Spurr plastic by means of computerized robotic capillary action. The techniques and computer program are delineated. The results demonstrate the highly resolved immunostaining with low background attained in plastic sections. Tissues were dewaxed, reembedded in Spurr, and reacted on the autostainer with no qualitative loss of immunoreactivity. Advantages of employing the autostainer include quality control (both positive controls and unknowns can be immunoreacted in tandem), ability to carry out immunoreactions on different cases at the same time, use of far less quantities of antibody and reagents than for manual procedures, and better use of technologist time (other laboratory procedures can be done while the sections are being robotically stained). AbstractImprovements in the reliability of immunoperoxidase techniques have resulted in an increase in their use in diagnosing human disease. A large number of antibodies against human antigens are available commercially, which has expanded the use and application of these techniques in human pathology. Veterinary pathology has the same requirements for diagnosing diseases encountered in other species. With that need in mind, an assessment of the cross-species reactivity of antibodies to human antigens using an immunoperoxidase technique was conducted. The majority of the antigens selected for testing in the two nonhuman species showed a strong reactivity to the antihuman antibodies. The results and the recommendations for obtaining optimum results are reported. (The J Histotechnol13:255, 1990)

AbstractVictoria blue (VB)-hematoxylin and eosin (H&E) staining, originally described by Yokokawa et al in 1983, has been used extensively in our surgical pathology lab as a routine stain for all surgical specimens with cancer. This simple staining sequence is useful for the demonstration of venous invasion by cancer cells as VB dye specifically demonstrates elastic fibers in the background of H&E-stained tissues. AbstractA dependable method for silver staining of renal tissue for electron microscopy is described. This method enhances the light microscopic information available to the morphologist and provides additional information to that obtained with conventional uranyl acetate and lead citrate stained sections for transmission electron microscopy. The method is a variation of the technique published in 1957 by Jones with periodic acid and silver methenamine. Thin sections are placed onto nickel grids and treated with periodic acid followed by a silver methenamine solution. The reaction needs to be carefully monitored throughout the procedure as the temperature and the staining time are critical to the methodology. The stained sections are viewed with a transmission electron microscope, at relatively low magnification. The method, once established, gives the pathologist another option apart from the routine uranyl acetate-lead citrate stains used for electron microscopy and may be of vital importance to the diagnosis of renal disease. AbstractA simple, reliable, consistent and economical nonbuffered acid acid method for melanin is presented. After a short presensitization in silver, the sections are treated in gum mastic and then immersed in a physical developer consisting of hydroquinone, gum mastic, and a very low concentration of silver nitrate. The sections are placed in thiosulfate and counterstained with nuclear fast red. The entire procedure takes under 40 minutes to perform. (The J Histotechnology13:279, 1990)

AbstractA method is reported that allows investigators to gain information quickly from whole body autoradiograms in which tritium (3H) is used as the tracing radioisotope. Through the use of enhancers, weak beta emissions are changed to photons of light that can be recorded on regular X-ray film. The process is simple, inexpensive, and rapid. AbstractFreezing tissue for intraoperative diagnosis often introduces artifacts that make subsequent histologic evaluation of routinely processed tissue difficult. While attention has been given to methods of improving the quality of the frozen section itself (microwave irradiation and pretreatment with dimethyl sulfoxide), little attention has been given to the production of optimal microscopic image quality in the permanently processed“frozen section control.”One hundred sections of liver from a single human autopsy were each frozen in a cryostat in OCT compound at−20°C; the sections were then transferred to one of five postfrozen section solutions-formalin, Omnifix, Tris buffer (.05 M, pH 7.6), B5, and ethylene glycol. Routine hematoxylin and eosin sections were prepared. Each of the 100 sections was graded blindly with assessment of four features: preservation of chromatin detail, preservation of nucleoli, preservation of cytoplasmic detail, and maintenance of cell borders. The superior postfrozen medium proved to be 10% formalin, which provided a greater preservation of nuclear and cytoplasmic detail than did the other four media. (The J Histotechnol13:289, 1990)

AbstractA commercial marking system, the Davidson Marking System, was used in our hospital to identify the surgical margins of excisional diagnostic biopsies. The colored dyes were easily applied and withstood both frozen section preparation and routine tissue processing.
