AbstractCampylobacterpylori, a gram negative spiral bacteria, has been found to be the causative infectious agent in many gastric and peptic ulcers and it is necessary to demonstrateC. pyloriin tissue sections for a prompt diagnosis. Many laboratories use a modified Steiner method to demonstrate the organism. Although this silver stain works very well, silver precipitation can become a problem with fixatives other than formalin. Precipitation renders the microorganisms difficult to identify, especially when they are present in scant amounts. The Butler Modified Wright stain, which utilizes Weigert iron hematoxylin, Wright stain solution, and safranin, adequately stainsC. pyloriwithout masking the bacteria. It is time and cost efficient and provides satisfactory results with formalin fixed tissue, as well as with Hollande and Bouin fixed tissue. (The J Histotechnol13:109, 1990)

AbstractA simple rapid method is described for the staining of Campylobacter-like organisms (CLOs) in paraffin embedded tissue sections. This method the Diff-Quik®is compared to two methods currently in use, the Warthin-Faulkner modification of the Warthin-Starry silver impregnation method and the May-Grunwald/Giemsa stain. Although staining of CLOs in gastric biopsies is acceptable in all three methods, the Diff-Quik stain reduces the time required to ten minutes. It also offers ease of performance and economy of reagents. (The J Histotechnol13:113, 1990).

AbstractAn immunocytochemical method for identifyingPneumocystis carinii(P. carinii) in formalin fixed, paraffin embedded tissue is described. The procedure utilized a commercially available monoclonal antibody and a sensitive avidin-biotin detection system. The 2G2P. cariniiantibody produced intense labeling of cyst walls, extra-cystic fibrillar material, and trophozoites, even in tissue that had been fixed in formalin for over 2 years. The procedure identified 30 of 30 knownP. cariniiinfected lung tissues. ImmunolabeledP. cariniiorganisms were easily visible under low power magnification. No false positives were observed. No immunoreactivity was observed with a variety of normal tissues and various fungi. The availability of a commercial antibody that reliably and specifically identifiesP. cariniiin histologically processed tissues will benefit pathologists, clinicians, and researchers. AbstractA method is described for the preparation of paraffin blocks containing multiple individual tissue samples by use of a commercially available skin biopsy punch. Cylindrical cores are cut from selected regions of paraffin blocks containing various tissues of interest for use in a variety of immunocytochemical techniques. From 12 to 40 separate cores are combined and reembedded in one paraffin block from which hundreds of nearly identical serial sections can be cut. This method departs from previously reported techniques for preparing blocks containing multiple tissue samples in that the samples are much larger, from 3 to 6 mm in diameter, which effectively eliminates false-negative immunocytochemical staining results. In addition, the core samples are organized within the blocks in a rectangular grid-like arrangement that enables easy microscopic identification of individual samples on stained sections. AbstractA microwave stimulated diffusion method for preparing hamster and human oral mucosal tissue block for paraffin sections within 15 min is described. The dehydration, clearance, and impregnation time was saved by a factor of 34 for animal tissue and a factor of 103 for human tissue. Light microscopically, the results were excellent and indistinguishable from those of tissues prepared by the classical method. The new method is valuable for infectious cases requiring urgent diagnosis; they can be performed without frozen sections that require unfixed fresh tissue and sometimes produce inconsistent results. Moreover, the microwave processing technique is practical in small research laboratories as a microwave oven is the only equipment required. (The J Historechnol13:127, 1990)

AbstractGlass jars are routinely used as molds for embedding hard tissue specimens such as bone and metal prostheses in polymethylmethacrylate. Our laboratory has successfully adopted the routine use of polypropylene disposable containers as the standard polymethylmethacrylate embedding mold. Polypropylene is less expensive and safer to use than glass, and the polymerization of the embedding medium is controlled, predictable, and fast. (The J Histotechnol13:131, 1990).

AbstractSporadic appearance of a nuclear artifact had hampered accurate microscopic evaluation of lymph node and bone marrow biopsies. Although troubleshooting included specimen handling, fixation and staining, all changes in methodology failed to produce consistently good quality sections. Improved morphologic detail was attained only after blocks were recut and sections were completely dried before heat-affixation. Complete drying of sections before heat-affixation was the only factor that consistently prevented the artifact. (The J Histotechnol13:135, 1990).

摘要:
AbstractThe substitution of 1% silver nitrate for 5 % silver nitrate in reticulum and argentaffin procedures results in equal or better staining. Because less ammonium hydroxide is required in the production of the ammoniacal silver solution, tissue loss associated with the strong arnmoniacal compounds is minimized, as is the danger of producing potentially explosive chemicals. In addition, a significant cost savings can be realized from the reduced amounts of silver nitrate required in the procedures. (The J Histotechnol13:137, 1990)
