AbstractPrimary cell culture, tissue culture, and established cell line culture methods are addressed in this paper. These methods are easily applicable to the pathology laboratory. Sterile conditions to obtain, process, disaggregate the surgical specimen, and incubate the cells are emphasized. Passage to multiwell-antisera saving plates, specific streptavidin-biotin-peroxidase and phosphatase-antiphosphatase method modifications, and bromodeoxyurydine incorporation in cell culture medium are described to evaluate the cellular proliferation. We also discuss new features about conservation of easily-preserved slides that can be filed as biopsies. Satisfactory results have been obtained using immunohistochemical methods in cell cultures with antibodies against vimentin, glial fibrillary acidic protein, muscle actin, proliferating cell nuclear antigen, bromodeoxyurydine, and AE1 keratin. AbstractWe developed a method to facilitate measurements of aluminum (Al) positive lines in bone biopsies from patients with renal osteodystrophy by modifying solochrome azurine (SA) methods for tissue A1 staining so that background staining of calcium (Ca) deposits, soft tissues, and glycol methacrylate (GMA) did not interfere with gray level thresholding of Al-positive deposits in video images processed by a semiautomated image analysis system. Detailed studies of the SA staining characteristics at various pHs of each component in histologic sections and of the solubility of the resulting SA-complexes in aqueous and alcoholic solutions indicated that optimal staining of A1 deposits could be obtained by staining GMA embedded sections for 16 hr at pH 5. Subsequent destaining with deionized water, ethylenediaminetetraacetic acid (EDTA), and deionized water removed background staining without loss of A1 staining. Sections prepared by these methods were satisfactory for obtaining, enhancing, and measuring Al-positive lines with semi-automated programs designed for use with the Image 1 Video Image Analysis. Measurements obtained in this way are as reproducible and precise but faster than manual methods for measuring A1 lines, and, therefore, are an improvement in the methodology for measuring A1 lines in bone biopsies from patients with renal osteodystrophy or other bone diseases. AbstractRecent morphometric analysis of histological structures has been performed with computerized image analyzers, thus supplying previously unknown infomation. However, conventional methods are still used because few research centers have investigators trained in this technique. The basic objective of this study was to compare studies on renal tissues obtained both by the conventional method and the computerized analysis. We concluded that both methods are useful, in spite of some differences found in certain parameters, such as the glomerulus diameter and the percentage of glomerular area per glomerulus. The similarity of the results of area measurements and of glomerular sclerosis is emphasized. (The J Histotechnol19:121, 1996)

AbstractThis report compares a direct, peroxidase conjugated antibody method with a peroxidase labeled streptavidin/biotin (LSAB) method for the detection of immunoglobulins A, G and M, complement (C3c), fibrinogen and kappa and lambda light chains in neutral buffered, formalin fixed, paraffin embedded renal biopsy tissue.Twenty-five percutaneous renal biopsies were selected and 2μm serial sections cut. These were deparaffinized, rehydrated, blocked for endogenous peroxidase, and digested with 0.025% protease type VIII for 3 min. Detection efficiencies of the primary antibodies were determined by checkerboard titrations, and optimal dilutions of each antibody were applied for 1 hr at room temperature. Antibody binding sites in both methods were detected with 3,3′diaminobenzidine tetrahydrochloride (DAB).Both methods detected similar distributions and quantities of immune reactants in each biopsy. A greater detection efficiency was achieved for each antibody with LSAB, and the intensity of the reaction products was similar for both methods. AbstractHelicobacter Pyloriis a gram negative bacterium that has been shown to be the causative organism in some gastric and duodenal ulcers. It is the most common cause of antral gastritis and has been linked to the development of distal gastric carcinoma and low grade gastric lymphoma. Identification in a gastric biopsy and prompt treatment with antibiotics, can lead to a speedy recovery for the patient. Unfortunately this organism stains poorly with standard hematoxylin and eosin techniques and special staining methods can be lengthy. The usual demonstration methods are Steiner and Romanowski type stains. Toluidine blue has been used as a rapid method for the demonstration of these organisms, but the contrast is generally very poor. We have developed a counterstain for toluidine blue that shows deep blue organisms on a yellow background. The combined procedure takes about 30 min. (The J Histotechnol19:131 1996)

AbstractA modified Fite carbon fuchsin stain for Mycobacterium species and other acid-fast bacteria is described. This procedure eliminates uneven stain differentiation and areas of excess stain deposition. Harris hematoxylin is used for the counterstain to allow better nuclear detail. This stain was found to be superior to conventional Fite staining for Mycobacterium species and other acid-fast bacteria that are difficult to visual and for total tissue demonstration. AbstractUranyl nitrate has been utilized as a sensitizer in silver stains formulated to detect spirochetes since the inception of these methods, even though many other aspects of these techniques have been changed over time. We have substituted zinc formalin for uranyl nitrate in both the Warthin- Starry and modified Steiner techniques. The resulting spirochete staining is as good or better than when uranyl nitrate is used. (The J Histotechnol19:135, 1996)

AbstractThe purpose of this study was to compare the performance of the alkaline phosphatase-streptavidin based detection systems from 3 commercially available kits for in situ hybridization. Of the 65 cases analyzed, 62 were known to contain human papillomavirus (HPV), 1 contained herpes simplex virus, and the other 2 cytomegalovirus by prior molecular analyses. The detection rates for the viral DNA using biotin-labeled probes were equivalent with the 3 detection systems, being 56/65 (86%) for the product from BioGenex, 53/65 (82%) from Digene, and 21/24 (87%) for Enzo Diagnostics. The BioGenex system yielded the overall strongest hybridization signal, defined by the intensity of the colorimetric reaction and number of positive cells. The Enzo system gave the lowest background signal. The Digene system was the easiest to use as it is a 1-step detection system as compared to a 3-step system for the other companies. We conclude that each of the kits is good for the detection of a target-probe complex for in situ hybridization and each offers advantages and disadvantages when compared to the other. AbstractA simple method of rat brain embedding into egg yolk facilitates serial sectioning of the whole brain in coronal plane. The sections are compact and can easily be treated by such free floating techniques as silver impregnation or immunohistochemistry. Unembedded material is hard to process in this way, as caudal parts of the brain (including occipital cortices, cerebellar convolutions, and brain stem) form separate pieces that must be treated individually, and their unfolding and appropriate positioning onto slide is difficult. A simple and rapid embedding protocol greatly simplified processing the large number of sections. This technique may be of practical importance in several cases where frozen or vibratome brain sections are to be processed by floating technique. (The J Histotechnol19:143, 1996)
