AbstractThe demonstration of nucleolar organizer regions (NORs) has received increasing popularity in the study of basic cell biology and tumorigenesis. The most widely used method requires a 50% silver nitrate gelatin emulsion (AgNOR) and dark room manipulation. Morphologic detail is often poor. We investigated Bielschowsky (Biel) and Grimelius (Grim) silver techniques and nuclear fast red (NFR) and metanil yellow (MY) counterstains. We compared them with the AgNOR technique. Five breast carcinomas and 5 malignant melanomas were stained and counted in a single blinded fashion. Comparisons to AgNOR were made and correlation coefficients (r) calculated as follows: AgNOR vs AgNOR + NFR, r = 0.96; AgNOR vs Biel, r = 0.96; AgNOR + NFR vs Biel + NFR, r = 0.98; Biel vs Biel + NFR, r = 0.93. Grim and MY were unsatisfactory.These data demonstrated that Bielschowsky offers a comparable alternative to AgNOR that may be more suitable for smaller laboratories. NFR improved morphologic detail and did not significantly alter the overall NOR count. (The J Histotechnol17:99, 1994)

摘要:
AbstractAn 0.1 125% zinc chloride, 50% isopropyl alcoholic, 10% unbuffered formalin postfixative for routine surgical pathology tissues is described. Integration of this zinc isopropyl alcoholic formalin postfixative into all automated routine tissue processing produces lymphoid tissue morphology similar to that produced by B5, but without the mercurial hazard. Antigenicity is also preserved and possibly augmented (The J Histotechnol17:105, 1994)

AbstractThe aim of this study was to determine the optimal conditions for the processing and staining of specimens obtained by fine needle tissue aspiration biopsy (FNTAB) of the testis. We assessed the 5 most commonly cited fixative solutions, the 6 most popular staining schedules, the effects of 6 hour and overnight dehydration and infiltration cycles, and 2 slide incubation temperatures. Comparisons were obtained from sources allocated by 5 independent observers to 7 histological features from 20 specimens.Sections incubated at 37°C prior to staining were graded significantly higher than those incubated at 60°C. Similarly, samples processed on the overnight dehydration and infiltration cycle were found to be better preserved than those processed on the 6 hour cycle. Phosphate buffered formalin and zinc formalin were graded significantly lower than the other fixatives for all features. Scores allocated to zinc sulfate, Bouin, and Stieve solutions differed slightly. Stieve solution was selected for FNTAB because of its proven ability to maintain the immunoreactivity of testicular tissue.The various staining techniques highlighted different elements of the tissue, with the Giemsa and hematoxylin&eosin schedules providing the best overall representations. A slight variation ofthe hematoxylin&eosin schedule was selected for FNTAB as it provided particularly clear nuclear definition. (The J Histotechnol17:111, 1994)

AbstractBecause paraffin embedded tissues of cases that were reactive to the antibody for estrogen receptor in the Abbott ERICA kit stored at room temperature (RT) as 3–5μm sections on glass slides lost reactivity, freshly cut sections were retested for antigen-antibody reaction to assure reactivity was not lost when tissues were stored at RT in the block. In an attempt to identify the time frame in which reactivity was lost as well as identify a better slide storage method, 4 methods were studied: RT; 4°C; deparaffinization and storage in 10% sucrose in PBS at 4°C; and deparaffinization and storage in Abbott storage medium at 4°C. Methods were assessed at 1 wk, 2 wk, 4 wk, 8 wk, and4 mo. Slides archived for at least 10 mo at RT were also studied. Antigen reactivity was scored by intensity of stain and percent of cells positive. Storage at RT produced the most degradation of intensity over the first 4 wk and significant loss of stain in the archival material. Slides stored in sucrose or Abbott storage medium showed the most preservation at 4 wk. All methods showed continued degradation of staining beyond 4 wk, although sucrose storage had the least alteration. Sucrose storage also showed improvement of staining quality even after short periods of time. (The J Histotechnol17:115, 1994)

AbstractFour testicular sex cord stromal tumors (3 Leydig cell tumors and 1 Sertoli-Leydig cell tumor) and the Sertoli and Leydig cells of 10 nonneoplastic testes were studied for their immunohistochemical reactivity to intermediate filaments. Low molecular weight cytokeratin positivity was present in the Sertoli cell component of the Sertoli-Leydig cell tumor and in Sertoli cells in atrophic seminiferous tubules of 4 of 10 nonneoplastic testes. Vimentin reactivity was present in the majority of Sertoli and Leydig cells in neoplastic and nonneoplastic testes. There was uniformly no reactivity for epithelial membrane antigen, carcinoembryonic antigen, and S-100 protein in either cell type. Our studies suggest that expression of intermediate filament may distinguish testicular Sertoli from Leydig cells and neoplasms derived from them. (The J Histotechnol17:119, 1994)

AbstractA microwave modification of the Mallory phosphotungstic acid hematoxylin stain is detailed. The technique eliminates the use of lengthy toxic mercuric chloride mordanting and dezenkerization by substituting zinc chloride. Staining is carried out at low heat levels to preclude boiling and stain breakdown and results in sharp, contaminant-free, dark blue muscle striations within about 20min in paraffin and glycol methacrylate tissue sections. AbstractA rapid one step Gomori trichrome with aniline blue (GTAB) stain is described. The original Gomori trichrome stain was modified by replacing the light green stain with an aniline blue stain. The aniline blue stained the necrotic myocardial fibers deep blue in hearts of patients undergoing autopsies following death due to early acute myocardial infarction. The aniline blue not only stained the fibrous components of the myocardial interstitium but also the necrotic myocardial fibers. The GTAB stain became very valuable in differentiating early acute hemorrhagic myocardial infarction from acute myocardial hemorrhage following coronary aorta bypass surgery. AbstractA modification of the silver methenamine/Masson trichrome (SMMT) method was used routinely to stain 1μm paraffin sections of over 2000 renal biopsies. This method provided a reliable means of differentiating the morphological changes in renal biopsy tissue. The glomerular and tubular basement membranes were outlined by a fine black layer of silver nitrate; and, in contrast, proteinaceous deposits, red blood cells, and fibrin stained a bright red; connective tissues stained different shades of green.A detailed evaluation of the use of the SMMT technique for the light microscopy of glomerular, vascular, and interstitial changes indicated this to be an excellent, inexpensive method for evaluating most aspects of renal pathology. The use of thin sections combined with high magnification enabled the detection of minute immune complex deposits on or within the basement membrane or in mesangial regions. Although this method cannot replace ultrastructural and immunocytochemical techniques, it offers considerably more information than is presently appreciated and in many cases it has provided adequate information for the pathological classification of the tissue. AbstractProcessing of histological specimens requires the use of reagents that are costly as well as hazardous to the technician and the environment. One such reagent, xylene, is routinely used as the clearing agent. This study evaluated the processing of liver, brain, and breast tissue by substituting selected clearing and infiltration mixtures (CIMs) for xylene. Four CIMs, consisting of 2 parts paraffin mixed with either 1 part xylene or a monosaturated, unsaturated, or saturated oil, were studied via alternate regimens. A fifth regimen was substitution of paraffin only for xylene. Half the alternate processing regimens employed isopropyl alcohol at 37°C as the dehydrant alcohol; the other halfused isopropyl alcohol heated to 50°C to facilitate tissue impregnation. All experiments were run in triplicate and compared to the standard processing regimen (control). Technical evaluations considered embedding, microtomy, and hematoxylin and eosin (H&E) staining procedures. Professional evaluation considered the suitability of the processed tissue for diagnosis.Our results indicated that the experimental CIMs could be used as effective xylene substitutes for satisfactory processing of liver, brain, and breast tissue. Only occasional minor difficulties in embedding, microtomy, or H&E staining procedures were observed. Ninety-six out of the 99 tissue preparations were satisfactory for making effective pathological diagnoses. (The J Histotechnol17:137, 1994)
