摘要:
AbstractA method for assessing the relative concentration and distribution of mineral in undecalcified bone sections is described. The procedure consists of gradually grinding plastic-embedded bone section to 100+M, exposing the sections to soft x-rays under vacuum on photographic film using an aluminum stepwedge as control. Maximum and minimum radiodensity in the photographic negative that correspond between the x-ray images of bone and the step-wedge were respectively compared. The total densitometric detection range of the system was arbitrarily divided into specified sub-ranges. The respective densitometric data for each sub-range was quantitated and expressed in numerical form.

摘要:
AbstractA method for staining various structures of chondro—osseous trabeculae of the growth plate is described. A pseudo Schiff reagent made from crystal violet is used in the PAS sequence as the stain for calcified cartilage. Prestaining in saturated picric acid differentiated between calcified bone and osteoid tissue before staining with a modified Goldner's or Mallory's procedure. The method has been particularly suitable for histomorphometric studies of rat growth plates using an automatic numerized image analyzer system.

摘要:
AbstractWe compared the enumeration and cytological study of blood basophils by our method to that of the chamber counting method. Smear samples were obtained by finger prick. Basophils stained with toluidine blue were counted, and characterization was done under oil immersion. The method was accurate, simple, and suitable for field use.

摘要:
AbstractWe investigated the possibility of a microwave oven to replace the standard electric oven and water bath in histological staining procedures that require increased temperature. The technical details were perfected using the Perl's stain for tissue iron. We used a temperature probe to regulate and monitor the temperature of the staining solution. This led to the successful modification of the iron staining procedure for use in the microwave oven, resulting in a reduction in incubation time of 80%, compared to the standard oven procedure. We now use the microwave oven in most of our staining procedures that require heating.

摘要:
AbstractBone marrow biopsies, aspirates and smears are important procedures for diagnoses. Morphological abnormalities like focal lesions (lymphomas, granulomas) cannot be found in smears but will be present in the biopsy. Superior histologic preparations are essential for diagnostic interpretation of bone marrow. Appropriate histologic procedures for the preparation of bone marrow sections are presented in detail. Methods of fixation, processing, sectioning, routine H&E, iron stains and certain“special stains”are described.

AbstractIn histochemistry, the terms Schiff's reagent and leucofuchsin are sometimes used as synonyms. Though these compounds are derived from the same dyes they are chemically very different. In dyes of the fuchsin group, three aniline or ortho-toluidine rings are attached by single bonds to a central carbon atom. The fourth carbon valency is unoccupied and allows passage of mobileπ-electrons from ring to ring. These molecular orbitals cause the color of triphenylmethyl dyes. In leuco Pararosanilin (leucofuchsin), a hydrogen atom is firmly attached at the fourth binding site of the carbon atom and prevents passage ofπ-electrons; therefore, the compound is decolorized. Leuco Pararosanilin cannot be recolorized under the conditions of the Feulgen or PAS reaction. The colorless carbinol derivative contains a hydroxy group instead of the hydrogen at the central carbon atom. In Schiff's reagent, a sulfonic acid group is loosely bound at the free valency of the carbon atom. This sulfonic acid group is removed during or after reaction of the molecules with aldehyde groups and the color of the dye is restored. Sinceleuco Pararosanilin cannot be substituted for Schiff s reagent and is chemically very different, the use of the term leucofuchsin as a synonym for Schiff's reagent is incorrect and should be discontinued.
