AbstractWebsters New World Dictionarydefines competency as“a condition of being competent or qualified.”Both technical and professional competencies, which imply attainment of specific skills/knowledge, are required by health professionals. How does an allied health professional attain the required competencies and how does one assess the attainment of technical competency in particular? We decided to use the technique of phase training in a pilot study to teach/evaluate students in the University of Texas, School of Allied Health Sciences, Program in Histotechnology, in the operation of the Tissue-Tek I11 Embedding System.

AbstractIt is generally believed that in von Kossa's technique silver cations react with phosphates and carbonates in calcium deposits and are then reduced to black metallic silver by strong light. Perusal of von Kossa's paper showed that he was aware of significant differences between reactions of silver phosphatein vitroand in calcium deposits. He regarded only the yellow coloration of calcium deposits during early stages of the reaction as diagnostic for calcium phosphate and ascribed the blackening to organic matter. Efforts to prevent this blackening were unsuccessful. Von Kossa's experiments were reproducible in our hands. Further studies showed that bright light, generally regarded as essential for von Kossa's reaction, only causes the irreversible blackening of organic matter that masks the yellow silver phosphate. When the reaction is performed in subdued light, yellow to yellowish brown silver phosphate is visualized selectively. Silver carbonate dissolves in thiosulfate and cannot be demonstrated with von Kossa's technique.

摘要:
AbstractFour histological techniques for the demonstration of spirochetes were evaluated. The Steiner technique(1944) was the most satisfactory but suffered a few drawbacks, such as precipitate and darkening of the tissue section which obscured the spirochetes. The method was therefore modified to improve on these points. The modified technique employs sensitization in uranyl nitrate, impregnation in 1% silver nitrate for an hour, brief exposure to 2.5% gum mastic followed by reduction in a solution composed of hydroquinone, gum mastic, absolute alcohol and a trace of silver nitrate. This method gives consistent results to reveal the presence of spirochetes and will demonstrate most bacteria as well.

摘要:
AbstractModifications of the Avidin-Biotin immunoperoxidase procedure (AB) for visual identification of tissueantigens are described. The alterations in the technique result in a 75 % reduction of processing time while at the same time improving sensitivity and enhancing tissue morphology. The use of this rapid technique allows this procedure to be utilized as a routine diagnostic method with a minimal expenditure in terms of labor and equipment.

摘要:
AbstractA hematoxylin and eosin procedure is described for staining formalin-fixed tissues embedded in glycolmethacrylate with emphasis on 1.5 micron sections. The staining method utilizes Gill's hematoxylin followed by a buffered eosin Y-phloxine B counterstain. Acidophilic and basophilic components of most stained tissue sections had an intense, sharp and clean appearance when observed by light microscopy.
