AbstractIn situ hybridization is a commonly used technique for the detection of nucleic acid sequences but its efficacy can be affected by many parameters. Human papillomavirus (HPV) infection of the genital tract is a useful model for determining how the various parameters may effect the hybridization signal. In this study, the rate of detection of HPV DNA was compared for 294 genital tract lesions fixed in Bouin solution and 306 lesions fixed in buffered formalin. The detection rate in low grade vulvar and cervical intraepithelial lesions fixed in buffered formalin was 96% and 91%, which compared to 83% and 76% , respectively, of tissues fixed in Bouin solution. The detection rate was 73% and 70% for vulvar and cervical high grade intraepithelial lesions fixed in buffered formalin, respectively, versus 50% and 47% for the comparable tissues fixed in Bouin solution. Thus, the decrease in the detection rate was greater for both the vulvar and cervical high grade lesions compared to the low grade lesions for tissues fixed in Bouin solution (33% and 29%) than for tissues fixed in buffered formalin (23% and 21%). This relative greater decrease in the detection rate for Bouin fixed tissues reflects the lower copy number found in high grade lesions relative to low grade lesions. These findings demonstrate that both the fixative and the histologic findings impact on the in situ hybridization results for HPV related lesions. (The J Histotechnol14:13, 1991)

AbstractA modification of a conventional immunocytochemical assay for demonstration of estrogen receptors (ER-ICA) in breast cancer tissue sections resulted in a sensitive quantitative assay for identification of ER low positive subpopulations of malignant cells. The imprint technique for specimen preparation was improved, the time of antigen exposure to antibodies and DAB incubation time was extended to increase the signal for an image analysis based measurement of the intranuclear amount of peroxidase label (the ER marker). The Imagescan, a commercially available software, was upgraded to calculate this amount per single cells and identical cell populations.The ER positive human breast cancer cell line, MCF-7, was the model to test our irnmunocytochemical method against ER-ICA imprint and other immunohistochemical methods. Tumor tissue specimens from a group of breast cancer patients was used to check the diagnostic sensitivity and specificity. Data obtained by our new Quantitative Immunocytochemical ER Assay (QUICERA) were compared with the content of ER in breast tissue homogenates as measured by a conventional dextran coated charcoal binding assay, with ER positivity on frozen tissue sections assessed by the ER-ICA Monoclonal kit and the amount of the intranuclear ER positivity inside breast cancer cells freshly isolated from the same tumor tissue specimens on imprints (ERICA Imprint).QUICERA was demonstrated to be more sensitive than other methods and suitable for search of ER positive and ER negative subpopulations of cells in tumors at the borderline range of biochemical positivity. AbstractA microwave radiation staining method for demonstrating canaliculi and other bone structures is described. Although simultaneous fixation and decalcification of bone tissue and staining were performed in a 650 W domestic kitchen microwave without temperature control, the procedures were closely monitored at 30% power level to prevent boiling of solutions. Compared to other methods for the calcification and ground section to show bone osteon, this technique of microwave stimulated diffusion for fast processing of bone tissue. It also gave excellent morphological results with substantial time and cost savings. It would be very useful in studies directed toward evaluating the presence of metabolic bone disorders in which structural and compositional changes affect bone in health and disease. It could also be adapted to other tissues (like teeth) and other stains, depending on the structure desired to be observed. (The J Histotechnol14:27, 1991)

AbstractMicrowave irradiation has repeatedly proved to accelerate and enhance histological staining. It can be applied to the identification of a wide variety of antigens by immunohistochemistry with 3-amino-9-ethylcarbazole (AEC) as the chromogen. However, to achieve adequate staining of some of the antigens, a combination of microwave and room temperature incubation is necessary. Further, it has not been possible to get a single, microwave only protocol applicable to all antibodies evaluated. In this report, a more rapid, nearly universal staining is demonstrated for most of the antigens tested using the chromogen diaminobenzidine (DAB) as an alternative to AEC. AbstractA simple, rapid, sensitive, consistent and economical method for plaques and tangles of Alzheimer's disease is presented. Sections were sensitized in 0.5% ammoniacal silver nitrate, followed by treatment in gum mastic and immersion in a physical developer consisting of hydroquinone, gum mastic and a very low concentration of silver nitrate. They were then treated with gold chloride and sodium thiosulfate. The procedure gives comparable results to those obtained with Bielchowsky and its modifications in about 30 minutes. AbstractWe have developed a rapid and reliable method for staining plastic embedded autoradiographs for in situ labeling protocols that eliminates several problems sometimes encountered with other methods, such as displacement of silver grains and uptake of stain by the emulsion layer itself. The procedure features the brief (less than one minute) application of toluidine blue to plastic embedded autoradiographs with moderate warming. The quality of staining obtained by this method is both consistent and flexible, allowing varying degrees of intensification without the danger of damaging the emulsion layer. The principle advantages of this procedure over other techniques to stain plastic embedded autoradiographs are its simplicity, rapidity and reproducibility. (The J Histotechnol14:43, 1991)

AbstractA method for freezing and cutting of frozen tissues is described in which the status of the tissue is not altered from the time of initial freezing to the mounting of sections on slides. AbstractThe technique described permits serial sections to be cut from the pupae ofPhaormia terranovawithout having to remove the epidermis before fixation. The pupae were prefixed for 1 hr at high temperature (85°C) in Bouin fluid. Their cuticulae were then removed before the pupae were postfixed at room temperature in Bouin fluid.
