AbstractPreviously described immuno-electron microscopic techniques have been unnecessarily complex and often gave variable results with excess background labeling. By refining existing techniques and standardizing fixation and embedding protocols, we have established an improved procedure for the ultrastructural localization of cytoskeletal filament proteins. This procedure, which may also be used for the identification of hormones and viruses, is described and results are illustrated. All antibodies and tissues examined showed an ultrastructural immunoreactivity pattern with a high degree of specificity and minimal background staining. The technique combines the advantages of increased specificity, simplicity and reproducibility, making it suitable for routine laboratory application. (The J Histotechnol13:9, 1990)

AbstractThe cytospin method was utilized to prepare fine needle aspirates of primary, recurrent, or metastatic breast cancers for evaluation of estrogen and progesterone receptors (ER and PR) by immunocytochemistry. The cytologic material was fixed in formaldehyde phosphate buffered saline (PBS) and pelleted by centrifugation. Following resuspension in 1–2 ml of PBS, slides were prepared with the cytospin technique and the cells were fixed in cold methanol and acetone. ER and PR were detected by an immunoperoxidase method utilizing specific monoclonal antibodies.The cytospin method yielded adequate numbers of tumor cells for ER and PR analysis. Tumor cells were confined to a small area, allowing rapid evaluation and minimizing requirement for monoclonal antibody. This technique is also easily adaptable for analysis of hormone receptors in malignant effusions. (The J Histotechnol13:23, 1990)

AbstractMeasurement of estrogen and progesterone receptors (ER and PR) is very important in selecting treatment for patients with breast cancer because some tumors respond to hormonal treatment. The standard methods for receptor measurements have been the steroid binding assays, especially the biochemical cytosol. These assays have several limitations, among which are that they can not identify tumor cells and they can not be used with routinely fixed and processed tissue. Immunocytochemical techniques with monoclonal antibodies are compatible with the study of receptor content in malignant cells, which is believed to be a good prognosticator of long term survival. One commercial estrogen receptor-immunocytochemical assay kit (ER-ICA) has received Food and Drug Administration approval for diagnostic use; however, it was designed for use on fresh frozen, mildly fixed sections.This study examined the parameters that would permit successful immunostaining with the ER-ICA in routine histologic sections: enzyme digestion, repeated application of primary antibody, and imidazole in the color development step. Results from ER-ICA performed in paraffin correlated well with those from the cytosol assay, and the technique could be of great value in cases where there is insufficient material for biochemical assay or when frozen material is unavailable. 摘要:
AbstractA microwave version of Steiner's method for demonstrating spirochetes and bacteria is described. The method differs froi Steiner's method and its modifications in that the concentration of the uranyl nitrate and silver nitrate solutions was reduced from 1% to 0.5%. Decreasing the concentration of the uranvl nitrate solution reduces exposure to this radioactive substance. Uranyl nitrate sensitization, silver nitrate impregnation, and the reduction were performed by microwave irradiation at temperatures well below the boiling point of the solutions. The entire procedure took about ten minutes to perform with even, consistent silver impregnation of the organisms. (The J Histotechnol13:45, 1990)

AbstractStaining for mast cells in undecalcified sections of bone was evaluated with a variety of toluidine blue (TB) preparations. Attention was given to color change in granules, nuclear clarity, background staining, and appearance of bone. Our buffered TB stain, which was developed to highlight bone detail, resulted in violaceous staining of the granules in mast cells, often restricting their identification. An acidified TB stain and a new stain of TB dissolved in 30% ethanol resulted in purple granules. Prestaining with Congo red prior to acidified TB allowed good bone detail and full expression of metachromasia with red appearance of the granules, giving the best view of the mast cell. AbstractThe objective of this study was to evaluate dimensional changes of orthopedic bone cement used in conjunction with Spurr and acrylic embedding technique. Prepolymerized cement was machined into blocks of three sizes. These were measured with a dial caliper prior to embedment. The embedding techniques of Bloebaum et a1 (1979) with Spurr medium and of Rhinelander (1979) with butyl/methyl methacrylate were compared. To minimize the possibility of technique error, the laboratory staff of each of these investigators performed the actual embedding of specimens. Once embedding was completed, the specimens were returned to our laboratories and ground to remove excess embedding media, then remeasured with an optical comparator. All of the butyl/methyl methacrylate specimens, regardless of size, had expanded 1 mm. The Spurr specimens, however, had no measurable dimensional changes. Although both embedding techniques exhibited similar visual variations, including lucent margins, a columnar orientation of the cement matrix in these lucent margins, and scalloping of the edges, the butyl/methyl methacrylate specimens exhibited crazing and a greater degree of morphological variation than that observed in Spurr specimens. Thus, when bone cement integrity is of importance, Spurr medium appears to be the embedding material of choice for minimal histological preparation artifacts. (The J Histotechnol13:55, 1990)

AbstractThis paper describes a new technique to assist in polishing plastic embedded specimens that are too thin to grind ade- quately when held with the fingers or mechanical devices. This technique, which we call the glue-glove technique, allows the successful grinding and polishing of thin specimens (1–2 mm thick) while minimizing specimen damage and eliminating finger injuries. AbstractTransformation of myelodysplastic syndrome to hematologic malignancies is not uncommon, but involvement of three committed progenitor cell lines at one time is seldom seen. We present a case of myelodysplastic syndrome with blast transformation, neoplastic cells that were difficult to classify morphologically, and basophilia found in marrow and peripheral blood. Differential diagnosis included myeloblastic leukemia, erythroblastic leukemia, megakaryoblastic leukemia, basophiloblastic leukemia, and systemic mast cell disease (SMCD). By using a battery of cytochemical techniques niques and irnrnuno~tochemicalm ethods, we were able to demonstrate the presence of myeloblasts, erythroblasts, and megakaryoblasts, thus excluding the possibility of SMCD and basophiloblastic leukemia.
