Abstract.Seven subjects, who were shown to have biphasic cycles were treated with Neogynon®(50 μgethinyl estradiol+250 μg D‐norgestrel). Before and during the treatment cycle serum levels of LH, 17ß‐estradiol and progesterone were estimated by radioimmunossay daily starting on the 8th day of the cycle. In addition BBT, the karyopyknotic index of vaginal smears, cervical function, Spinnbarkeit and crystallization were recorded. Similarly, 5 volunteers were given Neoviettar®* (30 μg ethinyl estradiol+150 μg D‐norgestrel), and both central and peripheral parameters were registered. Results of our experiments indicated that ovulation was suppressed in both groups of patients. It was interesting to note that inhibition of both central and peripheral parameters took place in the patients who were treated with the lower dosed oral contraceptive even in the first medication cycle. Another 5 subjects, who had taken Neogynon®for six months were switched to Neovletta®, and were monitored during their 6th cycle of treatment with the higher dosed combined preparation as well as during the 1st and 3rd cycle of administration of the lower dosed regimen. Analysis of data provides further evidence that ovulation was inhibited even in the first cycle after the switch. The suppression of serum levels of LH, 17ß‐estradiol and progesterone during the Neogynon®cycle was statistically indistinguishable from that during the 1st and 3rd Neovletta®cycle. This observation suggests that the degree of suppression of parameters of cycle function during the administration of combined preparations is apparently influenced by the progestagen‐estrogen ratio. In addition, data obtained in the 1st Neovletta®cycle were not different from those of the 3rd cycle of treatment. Similar results were recorded for the cervical function and the karyopyknotic index. The data of this pilot study combine to provide evidence that Neovletta®provides a very high degree of contraceptive protection even in the first treatment cycle after a switch‐over from a higher dosed pill, since it contains a back‐up mechanism in the case of a breakthrough ovulation. Abstract.An oral contraceptive containing D‐norgestrel 150 μg. and ethinol oestradiol 30 μg. (Neovletta®) was tested in 682 British women during more than 6,000 cycles. The pregnancy rate was 0.2 per 100 womanyears with the exclusion of pregnancies that followed admitted errors of tablet‐taking, and 0.8 per 100 woman‐years when they were included. 92.2 per cent of cycles were within 28 ± 3 days. Withdrawal‐bleeding was missed in 3.3 per cent of cycles. Spotting occurred in 9.3 per cent of cycles and breakthrough bleeding in 5.8 per cent. Total bleeding averaged 4.71 days per cycle. Concurrent symptoms likely to be attributable to the pill were few and only 10.3 per cent of subjects withdrew because of them. Abstract.The reliability and the overall clinical acceptance of a new combined oral contraceptive, containing 30 mcg of ethinyl estradiol and 0.15 mg of D‐norgestrel, was studied in 102 patients over 1,731 cycles. The results have been accumulated over a period of four years, some of the subjects still using the presented formulation. No pregnancy occurred during the observation period. Out of the totally observed cycles, 94.3 per cent had a duration of 28 ± 2 days. In only 1.7 per cent of the cycles did withdrawal bleeding fail to appear. Spotting and breakthrough bleeding occurred in 3.6 and 3.8 per cent of the cycles respectively. Abstract.Neovletta®, a new combined low estrogen‐low progestogen oral contraceptive was investigated in 226 women over 1921 cycles at three different clinics. This report constitutes a preliminary evaluation of a large seale multicenter study being conducted in Sweden. No pregnancy was reported during the observation period. The length of menstrual cycle was generally 28 days and withdrawal bleeding during the tablet free period was missed in only 0.9 per cent of the cycles. The duration of menstruation was shortened by about one day. The amount of menstrual flow, as registered by the women, showed a reduction during treatment. Spotting and breakthrough bleeding were increased during the first three treatment cycles, whereas from the fourth to 12th cycles the incidence of these bleeding irregularities was not much different from pretreatment values. Abstract.In 45 somatically healthy women the possible occurrence of psychic effects of a new low dose combined oral contraceptive was studied using different psychodiagnostic test methods. All the participants were previous users of some of the standard combined oral contraceptives, but had discontinued either due to experienced side‐effects, due to fear of such effects or due to other reasons. Four patients dropped out before the end of the six‐month‐observation period. Women who discontinued previous use of oral contraceptives due to side‐effects or due to fear of side‐effects exhibited a more pronounced degree of neuroticism compared to those who terminated due to other reasons. Those who experienced side‐effects during earlier medication had initially a higher depression score than the two remaining groups suggesting that women's basic psychic nature seems to play an important role in the development of psychic symptoms during oral contraceptive therapy. None of the three groups developed additional signs of depression during treatment. Moreover, no impairment of the sexual function, assessed by a number of parameters, was found.
