B6D2Fι female mice were intravenously administered 0.4 mg of glucan. 1, 5, 11, and 17 days later, the total nucleated cellularity (TNC) and the numbers of pluripotent hemopoietic stem cells (CFU-s), granulocyte-macrophage progenitor cells (GM-CFC), and erythroid colony-forming (CFU-e) and burst-forming (BFU-e) cells were assayed in the bone marrow and spleen. Bone marrow TNC was not altered, but splenic TNC increased approximately twofold on day 5 and remained increased on days 11 and 17 after glucan treatment. The concentrations of bone marrow and splenic CFU-s and GM-CFC both significantly increased (p < 0.01) by 5 days after glucan administration; however, they returned to control levels by day 17. Splenic CFU-e concentration increased on days 5, 11, and 17, whereas splenic BFU-e concentration increased only on day 11 after treatment. By contrast, bone marrow CFU-e and BFU-e concentrations were either unaffected or slightly decreased by glucan treatment. When peripheral blood was assayed for CFU-s and GM-CFC, no detectable increase in the concentrations of these progenitors was noted at any time after glucan treatment. The relevance of these effects of low-dose (0.4 mg) glucan treatment is discussed with respect to previously reported effects of higher-dose (e.g., 4.0 mg) glucan treatment

Following intrarectal administration of high molecular weight urokinase (molecular weight 53,000; 124,000 IU/mg protein) and its functionally active heavy chain (molecular weight 31,000; 212,000 IU/mg protein) to mice, activation of plasma fibrinolysis was observed. The plasma levels of pyro-Glu-Gly-Arg-pNA amidolysis and euglobulin fibrinolytic activities reached a maximum (43.4 ± 5.3 nmol/ml and 10.8 ± 9.4 mm2/0.03 ml, respectively) at 1–2 h after the administration of 300 IU of either enzyme per mouse, and thereafter, both activities declined slowly. Platelets incubated with prostaglandin synthetase inhibitors, i.e. aspirin (ASA) and diclofenac-sodium (Voltaren) show a decreased capability to aggregate. While ASA induces the complete loss of platelet pseudopodia, Voltaren causes the appearance of long, needle-shaped pseudopodia, a finding which may suggest a rather increased tendency to aggregate. Platelets labeled with ferritin hydrazide and examined with a transmission electron microscope and by X-ray analysis show a greater deposition of ferritin hydrazide after incubation with Voltaren in comparison to those incubated with ASA and to the controls. This finding suggests an increased negative charge of the stretched platelet membrane after incubation with Voltaren with a subsequent increase in the repulsive forces between platelets.

Severe stress, with increased secretion of adrenaline, is likely to cause platelet activation. The aim of the present study was to investigate if moderate stress, which usually is not accompanied by adrenaline secretion, could induce activation of platelets, as measured by changes in the plasma concentrations of platelet factor 4 (PF-4). Noise stimulation (100 dBA for 10 min) caused a significant increase in the diastolic (10%, p < O.Ol) and mean arterial pressures (4%, p < O.Ol) of 10 healthy male volunteers. The plasma levels of PF-4 and the venous platelet concentrations were, however, unaffected during noise exposure. The results therefore suggest that stress not accompanied by adrenal medullary activation, does not induce platelet activation.

An immunoradiometric assay (IRMA) for the determination of factor IX inhibitors in haemophilia B was developed. The assay was based on competitive binding between radiolabelled anti-IX and inhibitors in the test material of immobilized IX:C. 5 haemophiliacs with known inhibitors were investigated with the new method and with a conventional neutralization test. An inhibitor titre as low as 5 × 10-4 U/ml could be measured by IRMA, showing it to be about 500 times as sensitive as the conventional neutralization assay used, and in 2 cases the inhibitors could only be detected by IRMA. The new method is thus useful for the investigation of patients with low inhibitor titres, or when inhibitors with divergent properties are suspected

Superoxide dismutase (SOD) activity and reduced glutathione (GSH) content in erythrocytes (RBC) of uremic patients on chronic dialysis were determined. Specific SOD activity and GSH content were found lower in RBC from uremic patients before hemodialysis compared to normal RBC; after hemodialysis, the specific activity of SOD and GSH content were significantly increased.

A 70 year-old woman developed typical clinical symptoms of pure red cell anemia (PRCA) following a history of rheumatoid arthritis (RA). The patient’s bone marrow erythropoietic progenitors cells were cloned in a micro agar culture system several times over a period of 11 months, revealing a diminished frequency of bone marrow erythroblasts paralleled by a markedly reduced number of CFU-e and BFU-e in vitro. No inhibitory activity in the patient’s IgG fraction could be detected either by preincubation with IgG and/or rabbit complement, or in the continuous presence of IgG. Depletion of T lymphocytes from the patient’s bone marrow cells led to an improved in vitro erythroid proliferation. Cytostatic therapy with cyclophosphamide clinically induced a marked increase in the bone marrow erythroblast and reticulocyte number, correlated in vitro by normalization of CFU-e levels and increase in the number of BFU-e. Nevertheless, BFU-e values never attained normal levels, which could be attributed to a reduced stem cell pool resulting from previous therapy with cyclophosphamide and/or antirheumatic drugs. A 40-year-old man with Philadelphia chromosome-positive chronic granulocytic leukaemia developed blast crisis with a mixture of lymphoid and myeloid blasts. A bone marrow relapse with predominantly myeloid features was followed by menin-geal leukaemia of purely lymphoid differentiation. The terminal crisis showed a homogenous population of large unclassifiable blasts with simultaneous expression of lymphoid and myelomonocytic surface antigens. The repeated phenotypic conversions were documented by morphological studies and by cell marker analysis with monoclonal antibodies.

A patient with hairy cell leukaemia is described who developed progressive lymphadenopathy, which proved to be due to Mycobacterium kansasii infection. Clinicians should be alert to this rare but well-described complication of hairy cell leukaemia. Early lymph node biopsy is recommended in patients developing otherwise unexplained fever and lymph node enlargement.
