The effects of various human sera, platelet lysates and platelet-derived growth factor (PDGF) on the proliferation of human bone marrow-derived fibroblastoid colony-forming cells (CFU-F) were examined. We obtained nearly identical growth curves of fibroblastoid colonies with sera, platelet lysates and PDGF as stimulants and concluded that PDGF was a main growth factor for CFU-F in human serum. In contrast to colony size, CFU-F number was irrelevant to the concentration of PDGF. Removal of culture medium containing hemopoietic cells after short-term incubation of bone marrow cells reduced both colony number and size in CFU-F cultures. When each of bone marrow-conditioned medium (BMCM), peripheral blood mononuclear cells (MNC) and phytohemagglutinin-stimulated leukocyte-conditioned medium (PHA-LCM) was added to the cultures, CFU-F number and colony size recovered. The role of PDGF and the factors present in BMCM, MNC and PHA-LCM in the growth of CFU-F and their precursor cells were discussed

A soluble inhibitor of granulocyte macrophage colony growth, to which we shall refer to as T-de-rived colony-inhibiting activity (Td/CIA), was obtained from the supernatant of T cells from 5 healthy donors and 5 patients with severe aplastic anemia (SAA) in remission, following immunosuppressive therapy. The su-pernatants were purified by an AC A 44 column and the suppressor activity found in fractions of 70,000–80,000 daltons. Experiments were then performed to test for (a) endogenous productions of Td/CIA in normal marrow cells (NBM), (b) reversibility of suppression, and (c) competitive inhibition with human placenta-conditioned medium (HPCM). The results of this study can be summarized as follows: (1) the endogenous production of Td/CIA can be elicited by addition of mitogens to NBM, and is prevented if the marrow is T-depleted or treated with cyclosporin A; (2) suppression is completely reversible if Td/CIA is removed from NBM by washing at 1, 48, 72 and 96 h; (3) CFU-c which have been exposed to Td/CIA once, and freed from Td/CIA by washing, are still sensitive to a second exposure of Td/CIA; (4) there is no clear competitive inhibition between Td/CIA and HPCM. Scanning electron microscopy has shown a typical cell surface morphology in hairy cell leukemia. Scanning immunoelectron microscope techniques, utilizing monoclonal antibodies and colloidal gold particles, have recently become available. Eight patients with hairy cell leukemia have been studied with a panel of monoclonal antibodies of which Bl, BA1, OKM1, anti-TAC and LeuM5 were shown to be suitable for scanning immunoelectron microscopy and reactive with hairy cells. The combination of typical cell surface features with reactivity for the Bl and LeuM5 monoclonal antibodies permits accurate diagnosis of hairy cell leukemia, particularly when low percentages of hairy cells are present as in hypocellular or in interferon-treated patients

摘要:
A patient who developed a meningeal leukemia during prolymphocytoid transformation of B-chronic lymphocytic leukemia is described. The central nervous system (CNS) complication occurred after 5 years of relatively benign course. Almost all CNS mononuclear cells showed the typical morphology and immunology of prolymphocytic leukemia cells, whereas a mixture of prolymphocytoid elements and small-sized lymphoid cells was observed in peripheral blood. In this case the more aggressive component was present at diagnosis and showed an uncommonly slow progression allowing the occurrence of the CNS involvement

Megakaryocytopoiesis was investigated with a polyclonal (26 cases) and a monoclonal (20 cases) antibody to factor-VIII-related antigen (factor VIII RAg) with the indirect peroxidase-antiperoxidase (PAP) method on air-dried bone marrow aspirates (BM). Numerous megakaryocyte precursors were identified in 5 patients with essential thrombocythemia (ET) and the 2 patients with acute myelogeneous leukaemia (AML) after recovery from therapeutic aplasia. Small megakaryocyte precursors were rare in controls (C), patients with reactive thrombocytosis (RT) and immune thrombocytopenia (IT). In 2 patients with severe alcoholism the number of mature megakaryocytes increased after 5 and 7 days of abstinence, respectively

The platelet element content was determined by semiquantitative X-ray microanalysis in 6 patients with chronic renal failure. The patients showed a different degree of thrombocytopathy expressed by impaired adhesiveness, epinephrine-induced aggregation and platelet factor 3 availability. The microanalysis indicated significantly increased quantities of phosphorus and copper in patients’ platelets. Sodium and zinc showed a decrease in about half of the patients, whereas iron was significantly increased in at least 50% of the patients. Red blood cells (RBC) from iron-deficient rats were found to generate more malonyldialdehyde after in vitro incubation with H2O2 than RBC from control rats (p < 0.001). The iron-deficient RBC, however, had a higher content of superoxide dismutase (SOD) than control RBC (p < 0.02). 42 different samples of human erythrocytes were tested for glutathione-peroxidase activity (GSH-Px) in an attempt to study the inhibitory effect of Bromsulphalein (BSP). The mean activity of the enzyme was 11.90 ± 3.61 U/g Hb, with no significant difference between males and females. BSP was used at different concentrations from 1 to 45 mMand inhibited GSH-Px activity; the inhibition curve showed a sinusoidal pattern. The major effect was obtained at 30 m M BSP when almost 65% of the initial activity was inhibited. The inhibition of GSH-Px by BSP has also been confirmed using partially purified GSH-Px obtained from human erythrocytes, as well as purified bovine GSH-Px. Some difference was noted between males and females: females may be divided into two subgroups, one with a lower and a second with a higher level of GSH-Px. 1 mM BSP increased the activity in the first group, whereas it reduced the activity in the second group. The inhibition by BSP was positively correlated with the basal value of GSH-Px and this effect was particularly evident in females (r = 0.865; p < 0.001). We present a young man with Mediterranean type glucose-6-phosphate dehydrogenase (G6PD) deficiency and insulin-dependent diabetes mellitus whose brittle course was characterized by recurrent bouts of hypoglycemia and diabetic ketoacidosis (DKA). While neither of the episodes of DKA was complicated by hemolysis, hemolytic anemia consistently followed the recurrent attacks of hypoglycemia. Stringent control of the patient’s blood glucose levels in the upper limit and slightly above the normal range successfully prevented recurrence of hypoglycemia and recrudescence of hemolytic anemia. Severe hemolysis occurred in a 51-year-old female after a 17-day course of intravenous amoxicillin. A strongly positive direct antiglobulin test (anti-IgG titer 1:2,000) ensued which disappeared after withdrawal of the drug. Both the patient’s serum and eluate obtained from the patient’s red cells contained an IgG antibody which reacted with red blood cells coated in vitro with amoxicillin, but not with uncoated cells. In addition, high-titer antipenicillin, antiampicillin and antiamoxicillin IgG antibodies could be demonstrated in her serum by a RAST-based solid-phase radioimmunoassay.
