Folic acid (3H-pteroylglutamic acid or 75Se-selenofolate) administered free or bound to folate-binding protein (FABP) was rapidly cleared from the plasma but the plasma survival when bound to FABP was longer than unbound during the initial 5 min. 75Se-folate bound to FABP is more rapidly taken up by the liver than when administered as free 73Se-folate. 125I-labeled FABP was rapidly cleared from the plasma (90% in 5 min). The rapid uptake of FABP, a sialoglycoprotein, was inhibited by the preadministration of desialyzed fetuin. Following the hepatic uptake of 75Se-folate/FABP, only free 75Se-folate was found in the bile. The total biliary secretion of 75Se-folate over 240 min was 63% when administered bound to FABP and 33% when administered as free 75Se-folate. The liver retained 33.5% of the 125I activity 5 min following the administration of 125I-FABP, while only 4.46% of the 125I activity was secreted into the bile over 180 min. This data suggests that FABP may play an important role in the enterohepatic circulation of folates by directing nonmethylated folates to the liver.

Plasma and erythrocyte vitamin B12 levels were studied in pregnant Indian women consuming very little food of animal origin and compared with healthy non-pregnant women from the same dietetic group. The comparison of the mean values for routine haematological parameters revealed distinct fall in haemoglobin, packed cell volume, plasma vitamin B12, and albumin levels in pregnant women. However, the mean erythrocyte vitamin B12 levels in pregnant women with haemoglobin of more than 10 g/dl did not differ significantly from the corresponding mean observed in non-pregnant subjects. It seems that in normal pregnancy, the fall in erythrocyte vitamin B12 is much less marked than the fall in plasma levels of this vitamin.

A trimodal distribution of the amounts of Hb S in 56 sickle cell heterozygotes was observed. The variable amounts of Hb S were directly related to the mean cell volume and the mean cell Hb and were a result of the difference in net synthesis of Hb S due to the presence of the α-thalassaemia. Red blood cells (RBC) of children with terminal renal insufficiency show a reduction of the apparent 51Cr RBC half-life (16.5 ± 5.5 days). The enzymatic activities of phosphofructokinase (PFK) and glucose 6-phosphate dehydrogenase (G6PD) in the RBC of these patients do not differ from those in healthy persons. The RBC of uraemic and healthy children were separated by density gradient centrifugation in dextran medium according to maturity and age. RET count, mean cellular Hb concentration (MCHC), PFK and G6PD activities were determined in the separated cell populations. MCHC increases with increasing cell density, while the number of RET decreases. There is a distinct decrease in the G6PD activity of low-density cells compared to high-density cells. The extent of the decrease in enzymatic activity is about the same for healthy persons as for uraemic patients. A more rapid decrease in G6PD activity per unit of time can be assumed when the enzymatic activity is plotted against the distinctly shortened life span of the RBC of the uraemic children. There is a possible relationship between G6PD activity in old RBC and the premature destruction of these cells. PFK activity apparently does not decrease with increasing cell age

Haemoglobin synthesis was studied in bone marrow erythroblasts and in reticulocytes of 4 children with β°-thalassaemia major and of 7 children with β+-thalassaemia major. In patients with β°-thalassaemia the γ/α ratio was found to be lower in bone marrow than in peripheral blood. On the contrary, in patients with β+thalassaemia the β+γ/α ratio was more balanced in bone marrow, where the β-chain synthesis was higher, than in reticulocytes. Adenosine deaminase (ADA) has been assayed in plasma, erythrocytes, and lymphocytes from 29 patients with haematological and autoimmune diseases. ADA activity was uniformly low in erythrocytes and lymphocytes from patients with non-Hodgkin lymphoma and multiple myeloma (p < 0.001). High levels of ADA activity was found in plasma, erythrocytes, and lymphocytes from patients with myeloid leukemia (p < 0.001). ADA was high in plasma but low in erythrocytes and lymphocytes from patients with autoimmune diseases treated with immunosuppressive drugs (p < 0.05). 4 adults with congenital immunodeficiency showed decreased ADA activity. In the control group of normal blood donors we found a 34-year-old female with low ADA activity in plasma, erythrocytes, and lymphocytes without any immunological abnormalities. The data available for 338 cases of splenectomy in agnogenic myeloid metaplasia published since 1940 were retrieved from the literature and analyzed. The median survival of cases operated upon at an early stage of the disease (as judged by their hemoglobin level, platelet number, and splenic size) was compared with the survival reported in conservatively treated patients diagnosed at an early stage of agnogenic myeloid metaplasia. Survival time since diagnosis of patients with early splenectomy was similar to, or shorter than, that reported in conservatively treated patients with early agnogenic myeloid metaplasia.

The trend of some haemostatic parameters was investigated in a series of 186 myocardial infarction patients randomly allocated to sulphinpyrazone and placebo 15–25 days after the myocardial infarction episode in order to ascertain if one or more of these parameters may be considered as forecasting elements. The tests were performed at treatment allocation (basal values) and after 1, 6, and 12 months. In comparison with 44 healthy volunteers, the results have provided striking confirmation of ‘hyperactive’ platelets in the early phase of myocardial infarction expressed by the shorter bleeding time, increased plasmatic β-Mhromboglobulin, increased platelet factor 4 release and shorter heparin thrombin clotting time, and by the increased platelet sensitivity to threshold concentrations of adenosine diphosphate and collagen. Significant changes in bleeding time, platelet factor 4 release and heparin thrombin clotting time persist at successive testing times. Progressive antithrombin activity and the immunological levels of antithrombin III, α2-macroglobulin, and α1-antitrypsin were measured in women at various intervals during treatment with oral contraceptives and in a group of untreated women. No significant changes were observed in any of the above parameters. In both control and treated women, there was a positive statistical correlation between progressive antithrombin activity and antithrombin III immunological levels, whereas no correlation was found between the former and α1-antitrypsin or α2-macroglobulin immunological levels.
